Enterohemorrhagic Escherichia coli (EHEC) is an important cause of diarrhoea with blood and haemolytic uremic syndrome (HUS) in children and elderly people. Infections with EHEC are a world‐wide public health problem, related to consumption of contaminated ground beef. The aim of this study was to establish whether different meat foods sold in Santiago, Chile pose an infection risk by EHEC and to evaluate three different diagnostic techniques in foods, to determine which is most applicable for use in Chile. A parallel analysis was performed on 64 samples of meat foods (23 refrigerated ground meat, 23 refrigerated long pork sausages and 18 frozen hamburgers) sold in Santiago, Chile using DNA probes, enzyme‐linked immunosorbent assay (ELISA) and polymerase chain reaction (PCR). Twenty‐four samples (24 of 64=37.5 %) were positive by DNA probes, ELISA or PCR. The positive and negative predictive values, sensitivity and specificity of ELISA were 26.7, 81.6, 30.8 and 78.4 %, respectively. The positive and negative predictive values, sensitivity and specificity of PCR were 91.7, 96.2, 84.6 and 98 %, respectively. The EHEC serogroups most frequently isolated were O158, O157, O119, O125 and O114. These results show that, although molecular techniques such as enzyme immunoassays are useful for EHEC detection in meat foods, PCR has advantages in terms of sensitivity, specificity, cost and ease of implementation in Chile.  相似文献

Effects from 34 generations of selection, either up (U) or down (D), for 3- to 9-wk weight gain on genetic direct and postnatal maternal effects, dam size and maternal efficiency were evaluated in 25 cross-fostered sets of rats. Direct genetic effects on 12-d pup weight were 14 and 9% above controls (C) for U and D lines, respectively (P less than .01). Postnatal maternal effects on 12-d pup weight were also 11% higher for the U line (P less than .01), but not different from C for the D line. Females of the U line were heavier (P less than .01) by 13% at mating, 12% at parturition and 15% at 12 d of lactation, relative to controls. They also produced 11% greater total litter weight at 12 d and consumed 9% more feed, but were only nonsignificantly lower in feed per unit of pup 12-d litter weight. Females of D line were -8, -7 and -3% relative to controls in the three weights, but did not differ from controls in 12-d litter weight, feed consumed or in feed efficiency during lactation.  相似文献

The present study supports that short‐term (7‐days) protocols based on progestagen‐impregnated sponges and progesterone‐loaded CIDRs are equally effective to induce ovulatory response in sheep. There were no significant differences in the onset of estrus behavior (32.0 ± 6.0 and 33.8 ± 4.0 hr after device withdrawal for sponges and CIDRs, respectively; p > 0.05) and preovulatory LH discharge (5.1 ± 2.1 and 5.8 ± 3.3 hr after onset of estrus behavior for sponges and CIDRs, respectively; p > 0.05). These features are similar to previously described for classical long‐term (12–14 days) treatments. Hence, short‐term CIDR‐based protocols may be implemented using the same time‐intervals for insemination than sponge‐based and long‐term protocols.  相似文献

Two field experiments (Exp. 1 and Exp. 2) were carried out to assess the physiological performance and grain yield of wheat cultivars BR‐18 (moderately resistant) and Guamirim (susceptible) inoculated with Pyricularia oryzae in plots treated or untreated with Ópera (fungicide 13.3% epoxiconazole + 5% pyraclostrobin). Results from regression analyses indicated that spike and leaf blast severity at 10–14 days after inoculation (dai) were associated with greater yield losses (highest negative slope) than severity at 18–22 dai. Relative to untreated Guamirim, there were 0.3% and 16% increases in Exp. 1 and 2, respectively, for untreated BR‐18 (resistance alone). For fungicide treatment alone, the mean yield of Guamirim increased by 20% and 61% in Exp. 1 and 2, respectively, relative to the untreated fungicide control, whereas for the fungicide treated BR‐18, the mean yield increased by 26% and 83% in Exp. 1 and 2, respectively. Fungicide application and cultivar resistance resulted in higher measures of leaf health and photosynthetic performance in both spikes and leaves than in the untreated susceptible reference treatment. The results from this study may be useful in future efforts to develop crop loss models and management guidelines for wheat blast.  相似文献

Two experiments were carried out to assess the changes associated with photoassimilate production and partitioning in the source–sink relationship of flag leaves and spikes of wheat plants infected with Pyricularia oryzae, the causal agent of blast. Flag leaves and spikes were inoculated at 10 and 20 days after anthesis (daa) with a conidial suspension of P. oryzae. Analysis of chlorophyll a fluorescence using maximal photosystem II quantum efficiency (Fv?Fm), fraction of energy absorbed that is used in photochemistry (YII), quantum yield of non‐regulated energy dissipation (Y(NO)) and quantum yield of regulated energy dissipation (Y(NPQ)), showed an impairment of the photosynthetic performance in both infected flag leaves and spikes, coupled with reduced concentrations of chlorophyll b and carotenoids. Compared to non‐inoculated controls, there was lower capacity for CO2 fixation by RuBisCO in the infected flag leaves. Similarly, in the infected flag leaves and grains (obtained from infected spikes), there were lower concentrations of soluble sugars, while the hexoses‐to‐sucrose ratio increased in infected flag leaves. Compared to non‐inoculated controls, infected flag leaves showed lower sucrose phosphate synthase (SPS) activity and lower expression of the sucrose synthesis (SuSy) gene, while higher expression and activity of acid invertases also occurred. At the advanced stages of fungal infection, the concentration of starch in grains decreased but remained high for the infected flag leaves. There were reductions in ADP‐glucose pyrophosphorylase activity and the expression of ADP‐glucose pyrophosphorylase genes and a down‐regulation of β‐ and α‐amylase expression at the advanced stages of fungal infection on flag leaves and spikes. In conclusion, the effect of blast on both grain quality and yield can be associated with alterations in both production and partitioning of carbohydrates during the grain filling process.  相似文献

Phenolic compounds in Spanish virgin olive oils were characterized by HPLC. Simple phenols such as hydroxytyrosol, tyrosol, vanillic acid, p-coumaric acid, ferulic acid, and vanillin were found in most of the oils. The flavonoids apigenin and luteolin were also found in most of the oils. The dialdehydic form of elenolic acid linked to tyrosol and hydroxytyrosol was also detected, as were oleuropein and ligstroside aglycons. The structure of a new compound was elucidated by MS and NMR as being that of 4-(acetoxyethyl)-1,2-dihydroxybenzene. Changes of phenolic compounds in virgin olive oils with maturation of fruits were also studied. Hydroxytyrosol, tyrosol, and luteolin increased their concentration in oils with maturation of fruits. On the contrary, glucoside aglycons diminished their concentration with maturation. No clear tendency was observed for the rest of the phenolic compounds identified.  相似文献

The fatty acid composition and total lipids of various forage crops at different cutting dates and seasons, which are used in forage grazing beef production in Argentina, were determined. Three experiments were carried out at the Agricultural Experimental Station of INTA, Anguil, La Pampa, Argentina Samples of alfalfa from three different cultivars with different dormancy groups (G4‐5, G6‐7 and G8‐9) and samples of ryegrasses Bill, Florida and Queue, wheats Charrua and Guapo, and triticale Don Santiago cultivars were taken at different cutting dates and analysed for total lipids and fatty acid composition. Significant differences on the fatty acid composition, the ratios 18:3n‐3/18:2n‐6 and polyunsaturated fatty acids (PUFA) due to cultivar, cutting date and season were found. Taking into account the importance of 18:2n‐6 and 18:3n‐3 fatty acids as substrates for the conjugated isomers of 18:2n‐6 (CLA) and n‐3 PUFA concentrations in beef, the fatty acid composition of forages needs to be considered in forage grazing beef production systems. Data variation in both total lipids and individual fatty acid proportions in forages should be taken into account to establish the potential to cultivate crops for a higher content of n‐3 PUFA and hence the opportunity to deliver more n‐3 PUFA and CLA into beef.  相似文献
