The aim of this work was to develop mAbs against porcine CD205 and to conduct a comparative analysis of the CD205 protein expression on lymphoid tissues, monocyte-derived dendritic cells (DCs) and DCs isolated from the porcine skin. To conduct this study, we generated a monoclonal antibody, designated 1.F6F6, against the C-type lectin-like domain-5 of the porcine CD205 and showed that it recognizes a protein band of ~200 kDa by Western Blot analysis in mesenteric lymph nodes cells. Flow cytometric analysis showed that the mAb 1.F6F6 recognized 28.5%, 28.1% and 34.1% of cells from tonsil, inguinal and mesenteric lymph nodes, respectively, and 6% of cells from thymus. Analysis of monocyte-derived DCs showed that approximately 20% were positive and activation of the cells with LPS increased the positive population to 36%. Analysis of DCs isolated from the porcine skin showed that approximately 70% of the cell population expressed the CD205 receptor. The development of a monoclonal antibody capable of recognizing the CD205 receptor in swine opens up possibilities of applying new strategies for enhancing vaccine efficacy by using the anti-CD205 antibody for DC antigen-targeting to enhance priming of immune responses.  相似文献

Climate simulations anticipate an increase in mean summer temperature with synchronous decrease in summer precipitation during the course of the current century in Central Europe. As a consequence, transpiration of forest trees and stands may be altered along with soil water availability. In this study, the effect of reduced plant water availability to conifers was investigated into an open Pinus sylvestris forest (Erico-Pinetum typicum; P. sylvestris 60 %, Picea abies 20 %; and Larix decidua 20 %) within the inner alpine dry valley of the Inn River in Tyrol, Austria. For reducing plant water availability, we installed a transparent roof construction above the forest floor to prevent precipitation to reach the soil. The roofed area covered 240 m² and included 10 trees. A respective number of 11 trees served as controls in the absence of any manipulation. Roofing significantly reduced plant water availability as indicated in lower predawn needle water potentials. Sap flow density (Q s) was 63, 47, and 24 % lower in roofed P. sylvestris, P. abies, and L. decidua trees, respectively, as compared to control trees. Our findings suggest that P. sylvestris and P. abies behaves “isohydric” as they close their stomata relatively early under conditions of reduced plant water availability and thus stabilize their water relations, whereas L. decidua behaves “anisohydric” and maintains high transpiration rates.  相似文献

Tillage, organic resources and fertiliser effects on soil carbon (C) dynamics were investigated in 2000 and 2001 in Burkina Faso (West Africa). A split plot design with four replications was laid-out on a loamy-sand Ferric Lixisol with till and no-till as main treatments and fertiliser types as sub-treatments. Soil was fractionated physically into coarse (0.250–2 mm), medium (0.053–0.250 mm) and fine fractions (< 0.053 mm). Particulate organic carbon (POC) accounted for 47–53% of total soil organic carbon (SOC) concentration and particulate organic nitrogen (PON) for 30–37% of total soil nitrogen concentration. The POC decreased from 53% of total SOC in 2000 to 47% of total SOC in 2001. Tillage increased the contribution of POC to SOC. No-till led to the lowest loss in SOC in the fine fraction compared to tilled plots. Well-decomposed compost and single urea application in tilled as well as in no-till plots induced loss in POC. Crop N uptake was enhanced in tilled plots and may be up to 226 kg N ha−1 against a maximum of 146 kg N ha−1 in no-till plots. Combining crop residues and urea enhanced incorporation of new organic matter in the coarse fraction and the reduction of soil carbon mineralisation from the fine fraction. The PON and crop N uptake are strongly correlated in both till and no-till plots. Mineral-associated N is more correlated to N uptake by crop in tilled than in no-till plots. Combining recalcitrant organic resources and nitrogen fertiliser is the best option for sustaining crop production and reducing soil carbon decline in the more stabilised soil fraction in the semi-arid West Africa.  相似文献

The use of biochar in agriculture to achieve the dual benefits of improving soil quality whilst sequestering carbon (C) has received much attention. However, in low-intensity broadacre agricultural systems where yield is constrained by rainfall and costs associated with phosphorus (P) fertiliser, the application of biochar at rates commonly reported (>10 t ha?1) are likely to be prohibitively expensive where yield benefits cannot be guaranteed. In marginal areas where calcareous soils dominate, biochar application has no liming effect, reducing its value compared to application in acidic soils. In the present study, we use a field experiment to investigate the interaction between P fertilisation and biochar banding at low application rates (<1 t ha?1) on wheat yield and soil P fractionation (assessed by a modified Hedley method) in a highly alkaline Haplic Calcisol in a dryland broadacre cropping system. Our results demonstrate no statistically significant effect of low rate biochar banding on wheat yield in this highly P-constrained soil, but a significant effect of both biochar and fertiliser on P fractionation in both years of the study. Higher P fertiliser rates significantly increased wheat yield in all biochar treatments. The interactions between biochar, P fertiliser and P fractionation indicate shifts in potential P availability both as a result of P fertilisation and also biochar application. Further work is required in low productivity calcareous systems such as that studied here to elucidate the potential for biochar amendment to improve productivity and sequester C.  相似文献

The objective of this study was to quantify the combined effects of long-term plant biomass retention/removal and environmental conditions on soil microbial biomass phosphorus (P), bioavailable P, and acid phosphomonoesterase activity. Topsoil samples (0–2.5 and 2.5–5 cm) were collected from replicate field-based plots that had been maintained under contrasting plant biomass retention and removal regime for 21 years. Samples were collected on 14 occasions over a 17-month period and assessed for microbial P, bioavailable P, and phosphomonoesterase activity. All P measurements were consistently and significantly higher under plant biomass retention compared with biomass removal. Temporal variations in microbial P and phosphomonoesterase activity were evident in top soil (0–2.5 cm) and were driven by environmental conditions, mainly soil moisture, rainfall, and potential evapotranspiration, while bioavailable P had no temporal variation. Detailed analysis of microbial P data for the top 2.5-cm soil depth revealed that annual P flux through this pool was two times greater under biomass retention (10.3 kg P ha?1 year?1) compared with plant biomass removal (5.0 kg P ha?1 year?1). Similar and consistent trends were observed in soil from 2.5- to 5-cm sampling depth; however, differences were not significant. The findings of this study confirm the importance of the microbial biomass in determining the bioavailability of P in temperate grassland systems.  相似文献

A field experiment was laid out in Burkina Faso (West Africa) on an Eutric Cambisol to investigate the interaction of organic resource quality and phosphate rock on crop yield and to assess the contribution of earthworms (Millsonia inermis Michaelsen) to P availability after phosphate rock application. Organic resources of different quality were applied at a dose equivalent to 40 kg N ha–1 with or without phosphate rock from Kodjari (Burkina Faso) at a dose equivalent to 25 kg P ha–1, and were compared with control and single phosphate rock treatments in a factorial complete block design with four replicates. Sorghum (Sorghum bicolor L. Moench) variety SARIASSO 14 was grown. Sheep dung had the highest impact on earthworm casting intensity followed by maize straw. Combining organic resources with phosphate rock reduced earthworm casting activities compared to a single application of organic resources or phosphate rock. Addition of phosphate rock to maize straw reduced P availability in earthworm casts whereas combining sheep dung or compost with phosphate rock increased P availability. The contribution of earthworms to Kodjari phosphate rock solubilisation mainly occurred through their casts, as the available P content of casts was 4 times higher than that of the surrounding soil.  相似文献

In a survey of 666 sheep at a slaughterhouse, gallstones (concretions with a diameter greater than or equal to 1 mm) were found in the gallbladder of 50 sheep (7.5%), sludge (concretions with a diameter less than 1 mm) was found in 9 sheep (1.4%), and sludge plus gallstones were found in 7 sheep (1.1%). Gallstones and sludge were associated, and were more frequent in lambs and females, compared with adults and males. Qualitative analysis of the stones revealed all to be pigment (bilirubin) stones. There was a statistically significant increase of biliary bilirubin (total and indirect quota) only in sheep with gallstones plus sludge, compared with control sheep without sludge or gallstones. Concentrations of bilirubin, cholesterol, phospholipids, total and single bile aids, and total and ionized calcium were similar in the bile of sheep with gallstones, sludge, or both and control sheep. Bacteriologic analysis of the bile in 10 sheep with gallstones and 10 controls revealed bacteria in 50% of the first group and in 75% of the second group (Escherichia coli in all sheep and Salmonella spp also in 1 sheep with gallstones). These findings confirm our earlier findings of a high prevalence of black pigment gallstones in sheep. On that basis, we suggest that gallstones are associated with high total bilirubin concentration in the bile, and deconjugating bacteria are common in the biliary tract of these animals.  相似文献

The problem of nosocomial infections is of increasing importance in veterinary medicine. As an example, this review summarizes current knowledge regarding methicillin-resistant Staphylococcus aureus (MRSA) as a typical example, as these pathogens are the most important agents of nosocomial infections in human medicine worldwide and are being increasingly reported in veterinary medicine. MRSA are classified by their ability to be resistant against oxacillin/methicillin, this feature being confered by mecA, a gene which was acquired by horizontal gene transfer of the staphylococcal gene cassette (SCCmec). It is this genetic information that enables MRSA to be resistant against all penicillins, cehalosporins and carbapenems. In addition, MRSA are often resistant against a variety of other antiinfectives, i.e. aminoglycosides, macrolides, lincosamide, streptomycins, tetracyclin, chloramphenicol, but also against fluorquinolones and rifampicin. Presumably, these highly adapted strains are particularly able to acquire resistance genes located on plasmids or transposons.They are also able to develop point mutations, further leading to resistant phenotypes. If these pathogens are leading to infectious diseases, veterinarians may be confronted with a worst-case scenario, being left without any antiinfective therapeutic. As Staphylococcus aureus is highly tenacid, professional hygiene management is of utmost importance. The increasing number of published sporadic MRSA infections, MRSA-infectious diseases as well as MRSA outbreaks in veterinary medicine justifies their recognition as a "New Emerging Pathogen". So far, horses and dogs are mostly affected by MRSA. Although transmission between humans and animals has been reported occasionally, the sources, routes of transmission or the epidemiological relevance of MRSA infections in animals are far from being understood. Therefore, epidemiological investigations utilizing molecular typing tools are mandatory. Typing tools like multilocus-sequence-typing (MLST), pulsefield-gelelectrophoresis (PFGE), sequence analysis of the gene encoding protein A (spa-typing) as well as SCCmec-typing are all at hand.  相似文献
