ABSTRACT The witches'-broom disease, caused by the basidiomycete Crinipellis perniciosa, is the most limiting factor for cacao cultivation in Brazil. Trichoderma stromaticum is a mycoparasite of the witches'-broom pathogen of cacao that is currently being applied in the field to manage the disease in Bahia State, Brazil. In this work, molecular and traditional methods were used to study the genetic and biological diversity of this mycoparasite. Ninety-one isolates, mostly collected from farms not sprayed with the fungus, were analyzed by amplified fragment length polymorphisms (AFLP), which showed that two genetic groups (I and II) of T. stromaticum occur in Bahia State. This classification of T. stromaticum into two distinct AFLP groups was also in agreement with several other characteristics, including growth on agar media at different temperatures and sporulation on infected stem segments (broom pieces) and rice grains. Group II favors higher temperatures compared with group I. The genetic and biological differences of the isolates, however, were not evident in field experiments, where sporulation was evaluated on the surface of brooms under natural conditions. Our results show that there is considerable genetic and biological diversity within T. stromaticum in Bahia and other cacao-growing regions of South America that are affected by the witches'-broom disease. This diversity could be explored in the development of efficient biological control agents against the disease. Factors that may affect the application and performance of this biocontrol agent in the field, such as sporulation on rice substrate and on the brooms and growth at various temperatures, are discussed.  相似文献

The effects of supplementation of beef calves on weight gain, intake and digestibility of pasture, milk production and composition of their dams, and behavior of the pair cow–calf were assessed. Fifty-five beef cows with an initial average body weight of 449?±?8 kg and their respective offspring, with an initial average body weight of 138?+?3 kg and aged between 90 and 150 days, were used. Animals were submitted to an experimental period of 112 days. The experimental treatments consisted of: control?=?mineral mixture only, plan 1?=?high protein and high carbohydrate multiple supplement, plan 2?= high protein and low carbohydrate multiple supplement, plan 3?=?low protein and high carbohydrate multiple supplement, and plan 4?=?low protein and low carbohydrate multiple supplement. About 25 and 12.5 % of the protein requirements were supplied by the high and low protein supplements, respectively, and 15 and 7.5 % of total digestible nutrient requirements by the high and low carbohydrate supplements, respectively. Grazing behavior, performance, milk production, milk composition, intake, and digestibility of the cows were not affected (P?>?0.05) by the supplementation of the calves. The supplemented calves had greater (P?<?0.05) performance (as measured by final body weight, average daily gain, and final body condition score), intake and idle time, and lower (P?<?0.05) grazing time, but supplementation did not affect (P?>?0.05) suckling time and suckling frequency. It can be concluded that supplementation affects the behavior and feed intake of calves. However, it does not affect the suckling time and suckling frequency of calves. Additionally, performance, milk production, nutritional characteristics, and behavior of their dams are not affected.  相似文献

The effect of addition of grape seed tannins on the phenolic composition, chromatic characteristics, and antioxidant activity of red wine was studied. Two highly pure commercial grape seed tannins (GSE100 and GSE300) were selected, and their phenolic compositions were determined. Two types of red wines were made with Castela?o/Tinta Miu?da (3/2, w/w) grapevine varieties by fermentation on skin using two different maceration times, which correspond to the wines rich and poor in polyphenols, respectively. Each of these wines was used for experimentation with the addition of GSE100 and GSE300 before and immediately after alcoholic fermentation. Phenolic composition, chromatic characteristics, and antioxidant activity of the finished red wines were analyzed by HPLC-DAD, CIElab 76 convention, and DPPH radical test, respectively. The results showed that the addition of grape seed tannins had obvious effects of increasing color intensity and antioxidant activity only in the wines poor in polyphenols. Although GSE300 contained much higher amounts of di- and trimer procyanidins and a lower amount of polymeric proanthocyanidins, it provided effects of increasing the color intensity and antioxidant activity of the wines poor in polyphenols similar to those of GSE100. Furthermore, GSE100 released more gallic acid to wines than GSE300, although no gallic acid was detected in GSE100. Tannins added after alcoholic fermentation had a better effect on phenolic composition of red wine than tannins added before alcoholic fermentation.  相似文献

Increasing greenhouse gas (GHG) emissions from anthropogenic activities have contributed to global warming and consequently to climate change. Among all sources of emissions, the agricultural sector accounts for just under a quarter, mainly because of the intensification of food production systems necessary to supply the growing demand of the population. As ruminal fermentation is the largest source of methane emission in the livestock industry, emission by cattle has become the focus of studies. The aim of this study was to evaluate enteric methane emission and emission intensities of Nellore cattle at different ages submitted to levels of intensification of the grazing system. Twenty-four animals per cycle (age of 21.8 and 13.1 mo in cycles 1 and 2, respectively) were randomly distributed across different grazing systems: irrigated pasture with a high stocking rate (IHS), dryland pasture with a high stocking rate (DHS), recovering dryland pasture with a moderate stocking rate (DMS), and degraded pasture with a low stocking rate (DP). Methane emission was measured using the sulfur hexafluoride technique in each season of the cycle. Intensive systems provided higher yields of good-quality forage as well as superior animal performance when compared with DP. Methane yields were different between seasons and cycles. Methane emissions per average daily weight gain and dry matter digestible intake were different between treatments. Differences in the results were observed when they were analyzed per hectare, with the highest gain yield (P = 0.0134), stocking rate, weight gain, carcass production, and total methane emission (P < 0.0001) being found for the intensive systems. There were no differences in emissions per weight gain or carcass production between production systems, while a difference was observed between cycles (P = 0.0189 and P = 0.0255, respectively), resulting in lower emission intensities for younger animals. We conclude that more intensive systems resulted in a higher kilograms production of carcass per hectare; however, animals at 19 mo of age raised in the IHS and DMS systems had a lower emission intensity in kilogram of CO2-eq. per kilogram of carcass. Moderate intensification (DMS) using animals at about 19 mo of age might be an effective strategy to mitigate GHG emissions from Brazilian tropical pastures. Further studies are needed to understand the relationship between increasing productivity and decreasing environmental impacts, especially methane emission from ruminants.  相似文献

This retrospective study aimed to describe and classify cats with intraocular lymphoma, determine the proportion of cases with presumed solitary ocular lymphoma (PSOL) compared with ocular manifestations of multicentric disease and assess the clinical outcomes of these patients. One hundred seventy‐two cases identified through biopsy submissions were reviewed histologically; 163 of these cases were subtyped according to the WHO classification system. Cases were categorized as having PSOL or ocular lymphoma with suspected systemic involvement (SSI) based on submission forms and follow‐up data. The majority of cases exhibited concurrent uveitis (75%) and secondary glaucoma (58%). Diffuse large B‐cell lymphoma was the most common subtype (n = 86; 53%), followed by peripheral T‐cell lymphoma (n = 44; 27%). Other subtypes included anaplastic large T‐ (n = 8; 5%) and B‐cell (n = 4; 2.5%) lymphomas, and 15 cases (9%) were negative for all immunohistochemical markers. In sixty‐nine cases (40%), adequate clinical data and sufficient survival data were obtained to distinguish PSOL from SSI. PSOL comprised the majority of cases (64%), while 36% had SSI. When covarying for age at diagnosis, the median survival time was significantly higher (P = 0.003) for cases of PSOL (154 days) versus those with SSI (69 days); hazards ratio of 0.47 for PSOL (95% CI: 0.241‐0.937). The subtype of lymphoma did not affect survival time. Cats with PSOL represent a greater proportion of the disease population, and this subset of cats with intraocular lymphoma has a better clinical outcome.  相似文献
