Viruses are highly abundant in temperate soils, ranging from 107 to 109?g?1, and outnumbering soil bacteria from 5- to over 1,000-fold. In order to determine the potential impacts of viruses on soil microbial communities, it is important to establish reliable methods for comparing changes in viral abundances within and across soil samples. The goals of this study were to optimize extraction-enumeration methods to accurately determine viral abundances in a range of soil types, to evaluate the feasibility of simultaneously enumerating bacterial cells and virus particles using a single extraction procedure, and to assess the utility of flow cytometry (FCM) for enumerating virus particles in soil extracts. Comparisons of extraction approaches indicated that sonication or blender extraction of soils with potassium citrate buffer yielded the highest viral abundances for most soil types. Combined viral and bacterial extractions underestimate abundances compared to separately-optimized extractions for each. Flow cytometric counts were anywhere between 350- and 1,400-fold higher than epifluorescence microscopy (EFM)-based counts for the same soil. Trends in viral abundance across soil types were different from those via EFM, and different relationships between viral abundance and soil properties were observed depending on the enumeration method. Thus, FCM is not currently recommended for enumeration of viruses in soil extracts. Little is known about the genetic diversity of pale flax (Linum bienne Mill.), the wild progenitor of cultivated flax (L. usitatissimum L.), and ex situ germplasm of pale flax was scarce. Effort was made to collect 34 pale flax accessions and five landrace accessions of cultivated flax in Turkey. The inter simple sequence repeat (ISSR) technique was applied to characterize this set of flax germplasm, along with one Turkish cultivar, one Russian cultivar, five winter and four dehiscent type accessions of cultivated flax. Twenty-four ISSR primer pairs detected a total of 311 DNA fragments, of which 298 bands were polymorphic across 493 flax samples (roughly 10 samples per accession). These polymorphic bands had frequencies ranging from 0.002 to 0.998 and averaging 0.38. Accession-specific ISSR variation (Fst values) ranged from 0.469 to 0.514 and averaged 0.493. There was 49.3% ISSR variation resided among these 50 accessions, 35.9% harbored among landrace, winter, dehiscent types of cultivated flax and pale flax, and 38.2% present among 34 pale flax accessions. Pale flax displayed more ISSR variation than landraces and dehiscent type, but less than winter type, of cultivated flax. Clustering 493 individual plants revealed that these assayed plants were largely grouped according to their plant types and that pale flax was genetically more close to the dehiscent type, followed by the winter type and landrace, of cultivated flax. Pale flax collected within the geographic range of 180 km displayed a significant spatial genetic autocorrelation. Genetic distances among the pale flax accessions were significantly associated with their geographic distances and elevation differences. Global warming is predicted to have a profound impact on the Greenland Ice Sheet and its contribution to global sea-level rise. Recent mass loss in the northwest of Greenland has been substantial. Using aerial photographs, we produced digital elevation models and extended the time record of recent observed marginal dynamic thinning back to the mid-1980s. We reveal two independent dynamic ice loss events on the northwestern Greenland Ice Sheet margin: from 1985 to 1993 and 2005 to 2010, which were separated by limited mass changes. Our results suggest that the ice mass changes in this sector were primarily caused by short-lived dynamic ice loss events rather than changes in the surface mass balance. This finding challenges predictions about the future response of the Greenland Ice Sheet to increasing global temperatures. 相似文献

Wood is a major pool of organic carbon that is highly resistant to decay, owing largely to the presence of lignin. The only organisms capable of substantial lignin decay are white rot fungi in the Agaricomycetes, which also contains non-lignin-degrading brown rot and ectomycorrhizal species. Comparative analyses of 31 fungal genomes (12 generated for this study) suggest that lignin-degrading peroxidases expanded in the lineage leading to the ancestor of the Agaricomycetes, which is reconstructed as a white rot species, and then contracted in parallel lineages leading to brown rot and mycorrhizal species. Molecular clock analyses suggest that the origin of lignin degradation might have coincided with the sharp decrease in the rate of organic carbon burial around the end of the Carboniferous period. 相似文献

Reptile pain and analgesia is only beginning to be understood in veterinary research and clinical medicine. The diversity of the class Reptilia also makes it difficult to extrapolate analgesic efficacy across species. Many veterinary clinicians argue that the administration of analgesic medication is risky to the patient and may mask behavioral signs of pain, which are considered evolutionarily adaptive for survival. However, veterinarians have an ethical obligation to treat painful conditions in all animals, including reptiles, because effective pain management reduces stress-induced disruption to homeostatic mechanisms and also decreases morbidity and mortality associated with trauma or surgery. Nevertheless, several obstacles limit successful analgesic use, including subjectivity of pain assessment, inadequate knowledge regarding analgesic efficacy across species, pharmacokinetics of analgesic drugs, and the unknown relationship between risks and benefits for this class of drugs. An accumulation of autochthonous cases of canine babesiosis caused by Babesia canis has been registered in a small animal clinic in the Saarland since the beginning of 2006, some cases with fatal outcome. This study aims to contribute to the explanation of strong focal occurrence of infections with B. canis in this region.Therefore, patient owners who had presented their dogs in the years 2006 and 2007 because of babesiosis and who had claimed not having left the Saarland with their dogs at least six months before the outbreak of Babesiosis, were asked for their dog walking habits. Accordingly, a selection often tick collection sites of various landscape structures was made.Tick sampling by flagging the vegetation took place every month from March to December 2008. The collected ticks were identified morphologically. In eight of ten collecting sites a total of 397 adult Dermacentor reticulatus ticks were collected from March to December with the highest frequencies during the months of May, October and November. All collected specimens were examined by genus-specific conventional PCR for the presence of Babesia-DNA. In positive samples, the PCR-products were differentiated by sequencing. ten D. reticulatus (2.5%) ticks examined were found positive for DNA of B. canis canis originating from three out of eight collection sites. A 9-year-old Labrador mixed breed dog, known to suffer from heart disease, was presented. It had recently collapsed on the street. Based on clinical findings and ultrasonography the tentative diagnosis of an atrial rupture with bleeding into the pericardial sac, resulting into a cardiac tamponade, was made. The dog's state of health improved under treatment, including infusion, sedation and analgesia. Therefore pericardiocentesis was unnecessary. The dog was released from hospital the next evening and the medical therapy of its heart disease was continued. The animal died 85 days later. 相似文献

In this study, the hard ticks, whole blood and serum samples collected from small ruminants (sheep and goat) in middle Black Sea region of Turkey where Crimean-Congo hemorrhagic fever (CCHF) human cases were observed in the past years were surveyed for the presence of RNA and specific IgG antibodies from CCFH virus (CCHFV). CCHFV RNA was found in 30 of 255 tick pools (11.76%) and nine of 105 (8.57%) leucocyte samples. No CCHFV genomic RNA was detected from animals in Yildizeli and Vezirkopru. However, CCHFV RNA was found from animals in Gerze and Resadiye. Seventy-eight of 105 goat and sheep blood serum samples tested were antibody-positive for CCHFV by enzyme-linked immunosorbent assay (ELISA) (goat: 42/63; sheep: 36/42). Viral RNA was detected from tick samples in all of four provinces. Positivity rates for the provinces varied and were as follows: Gerze 13.04%, Resadiye 35.41%, Vezirkopru 1.61% and Yildizeli 6.06%. CCHFV genomic RNA was detected in four of seven tick species tested. These results suggest that these hard ticks may act as a reservoir for CCHFV in northern Turkey. 相似文献
