Corn leaf aphids (Rhopalosiphum maidis Fitch) are found throughout the year on maize in Hawaii and occasionally cause yield loss. Sweet corn inbred Hi38-71 was observed to have high field resistance to aphids and was chosen for this genetic study. An artificial infestation technique was developed using hair-pin clip cages (2.2 cm diameter) which we devised and built. The cages were applied to field-grown plants into which three wingless viviparous adults were placed. Aphid populations were classified on a 1–10 rating scale after about 2 weeks based on digital images of the cages. Mean aphid coverage ratings were 2.97 for the resistant parent and 7.28 for the susceptible parent (representing >200 insects per cage). The F1 hybrids showed similar susceptibility (6.72), showing resistance to be recessive in nature. Six generations of the cross between Hi38-71 and susceptible inbred Hi27 were artificially infested to provide a generation mean analysis of 360 treated plants over two growing seasons. A joint scaling test showed that the fit to a 3-parameter additive-dominance model satisfactorily explained the observed variability with no assumed linkage or epistasis. We conclude that resistance to corn leaf aphid in Hi38-71 is conferred by a single recessive gene labeled aph. These results concur with a previous study under uncontrolled natural infestation in a single environment. The hair-pin clip cage method was most effective in distinguishing resistant and susceptible genotypes under diverse natural growing conditions in Hawaii.  相似文献

The isolation of 7,4'-dihydroxy-8-methylflavan (1) from the dichloromethane extract of Pancratium littorale stem was guided by an assay for free radical scavenging activity towards the 2,2-diphenyl-1-picryl-hydrazyl radical (DPPH). The structure of 1 was established by spectrometric methods including UV, EI mass spectrometry, 1H and 13C-NMR. Adventitious rooting in shoot cuttings of neem (Azadirachta indica) and karanj (Pongamia pinnata) was investigated in different seasons. The influence of auxins and B-vitamins (neem only) was evaluated. Maximum rhizogenesis coincided with the emergence of new sprouts i.e. February (neem) and March (karanj). Considering both rooting percentage and root biomass, IBA was the most effective auxin tested. Compared to auxin free controls, IBA (1000 ppm in neem and 800 ppm in karanj) significantly increased adventitious rooting percentage (80 to 100 % respectively), the numbers of roots and root biomass for both species. In neem, B-vitamin treatment yielded 40–50% rooting for cuttings. This was significantly greater than rooting for controls but equivalent to IAA treatments. In neem, some of the cuttings treated with 2000–3000 ppm IBA had well developed root systems without any sprouts. In karanj, all auxin treatments promoted sprouting. The level of total soluble sugars in the rooting zone of neem and karanj was higher in non-rooted than rooted cuttings.  相似文献

The genetic bases of leaf rust resistance in wheat (Triticum aestivum L.) line CSP44, selected from the Australian cultivar Condor, and Indian cultivar VL404, were studied. The reaction patterns of CSP44 and VL404 against Indian races 12, 77, 77-1, 77-2, 77-3, 77-4, 77-5 and 108 were different from reaction patterns shown by near-isogenic lines with known adult plant resistance (APR) genes, viz. Lr12, Lr13, Lr22b and Lr34. Although the reaction patterns of CSP44 and VL404 were similar to the near-isogenic line Tc+Lr22a, tests of allelism indicated absence of Lr22a in both CSP44 and VL404. On the basis of genetic studies, their resistances in field tests against race 77-5, the most virulent race from the Indian sub-continent, were each ascribed to two genes. One of the two genes in each wheat was identified to be the non-hypersensitive APR gene Lr34. The second APR genes in CSP44 and VL404 gave hypersensitive reaction types and were recessive and dominant, respectively. The gene in CSP44 was designated Lr48and the gene in VL404, Lr49. This revised version was published online in August 2006 with corrections to the Cover Date.  相似文献

A primary driver of the wheat yield gap in Australia and globally is the supply of nitrogen (N) and options to increase N use efficiency (NUE) are fundamental to closure of the yield gap. Co‐application of N with phosphorus (P) is suggested as an avenue to increase fertiliser NUE, and inputs of N and P fertiliser are key variable costs in low rainfall cereal crops. Within field variability in the response to nutrients due to soil and season offers a further opportunity to refine inputs for increased efficiency. The response of wheat to N fertiliser input (0, 10, 20, 40 and 80 kg N ha‐1) under four levels of P fertiliser (0, 5, 10 and 20 kg P ha?1) was measured on three key low rainfall cropping soils (dune, mid‐slope and swale) across a dune‐swale system in a low rainfall semi‐arid environment in South Australia, for three successive cropping seasons. Wheat on sandy soils produced significant and linear yield and protein responses across all three seasons, while wheat on a clay loam only produced a yield response in a high rainfall season. Responses to P fertiliser were measured on the sandy soils but more variable in nature and a consistent effect of increased P nutrition leading to increased NUE was not measured.  相似文献

Worldwide, the nitrogen use efficiency (NUE) for crop plants is of great concern. The burgeoning world population needs crop genotypes that respond to higher nitrogen and show a direct relationship to yield with use of nitrogen inputs, i.e. high nitrogen-responsive genotypes. However, for fulfilling the high global demand of organic produce, it requires the low nitrogen responsive genotypes with greater NUE and grain yields. The lack of knowledge about precise regulatory mechanisms to explain NUE in crop plants hampers the goal of agricultural productivity. Understanding the molecular basis of NUE will enable to provide handle for crop improvement through biotechnological means. With the advent of modern genomics and proteomics approaches such as subtractive hybridization, differential display, and microarray techniques are revolutionizing to identify the candidate genes which play a pivotal role in the regulation of NUE. Beside it, quantitative real-time polymerase chain reaction technology is also being used to establish marker-trait association for NUE. The identification of potential candidate genes/proteins in the regulation of NUE will serve as biomarker(s) for screening genotypes for their nitrogen responsiveness for optimization of nitrogen input in agriculture. This paper describes the molecular basis of NUE with respect to nitrogen metabolism and its intimate relationship with carbon metabolism, use of molecular-physiological-genetics approaches for understanding the role of various genes/proteins, and their validation to use as biomarker(s) for determining genotypic potential for NUE. Since NUE in plants is a complex trait which not only involves the primary process of nitrogen uptake and assimilatory pathways but also a series of events, including metabolite partitioning, secondary remobilization, C-N interactions, as well as molecular signalling pathways and regulatory control outside the metabolic cascades. Therefore, identification of novel nitrogen responsive genes and their cis- and trans-acting gene elements is essential. Thus, fishing out a single gene, biomarker or a master regulator controlling complex trait of NUE could serve as an appropriate strategy for nitrogen management in agriculture.  相似文献

Wild Lens species/subspecies are a potential source for increasing genetic diversity in cultivated lentil. Four intraspecific crosses were attempted between cultivated and wild lentils. Viable hybrids were produced between L. culinaris ssp. culinaris × L. culinaris ssp. orientalis and L. culinaris ssp. culinaris × L. culinaris ssp. odomensis. Normal meiosis and pollen fertility were observed in the first set of crosses, whereas chromosomal abnormalities and reduced pollen fertility were observed in the second set of crosses. These crosses were also studied for some quantitative traits. The range, mean and coefficient of variation were calculated in parents, F1, F2 and BC1 generations to determine the extent of variability generated in the cultivated lentil through introgression of genes from wild lentil. The cultivated lentil × L. culinaris ssp. orientalis crosses showed substantially higher variability for all the traits than crosses involving cultivated lentil ×L. culinaris ssp. odomensis. The results of the present study indicated that these wild subspecies can be exploited for breeding purposes and their variation can easily be utilized to widen the genetic base of the cultivated lentil.  相似文献

Phenotypic divergence among the 24 accessions belonging to a collection of six species of Capsicum from different geoclimatic regions available in field gene bank of CIMAP, Lucknow, India, was quantified by multivariate analysis for 12 quantitative and qualitative traits. Based on their values, all 24 accessions were grouped into six clusters such that the genetic stocks within cluster had smaller D 2 values among themselves than those belonging to different clusters. The accessions of Capsicum annuum were distributed in different clusters in morphotyping representing different species, suggesting that taxonomic characteristics are not always related to agronomic traits. No parallelism/association was found between geographical and phenotypic diversity. Accessions 15 and 23 (clusters V and VI, respectively) had distinct identity. The three characteristics that played the greatest role in differentiation were fruit diameter, number of fruits per plant, and leaf diameter, which can be utilized as conventional/morphological markers for the improvement of chilli yield and obtaining good segregants in chilli breeding programs.  相似文献
