Tetracapsuloides bryosalmonae (Myxozoa: Malacosporea) is the causative agent of proliferative kidney disease (PKD), which affects both wild and farmed salmonid fish. The objective of this study was to outline differences in susceptibility to PKD in different salmonid species, hybrids and breeding lineages. Susceptibility to T. bryosalmonae infection was established based on cumulative mortality, pathological findings and detection of T. bryosalmonae in the kidney using immunohistochemistry and molecular methods. Determination of pure and hybrid individuals of different species in the genus Salvelinus, and dissimilarity of rainbow trout lineages, was performed using traditional polymerase chain reaction (PCR) and microsatellite analyses. Rainbow trout displayed higher disease severity compared with brook trout and Alsatian charr. Moreover, the results indicated differences in infection susceptibility, not only among different salmonid species but also among different lineages of charr and rainbow trout. Our study indicated that some salmonid species and even different lineages of the same species are more suitable for farming under PKD pressure. 相似文献

In northern Europe, there are high risks of severe pine weevil (Hylobius abietis) damage to newly planted conifer seedlings. Site preparation is one of the most important measures for reducing these risks and as several studies have shown the damage is highly dependent on the amount of pure mineral soil around the seedlings. We investigated effects of three site preparation techniques: (1) disc trenching with a conventional Bracke T26, (2) MidiFlex unit and (3) soil inversion with a Karl Oskar unit on characteristics of the planting spots, growth and pine weevil damage and survival rates of untreated and insecticide treated planted Norway spruce (Picea abies) seedlings. All three site preparation techniques reduced pine weevil damage in comparison with no site preparation, and the proportion of spots with pure mineral soil they created was inversely related to the rate of mortality caused by pine weevil. The results indicate that the quality of the planting spots depends on the technique used. In areas where pine weevil is the major threat to seedling survival, the amount of mineral soil in the planting spots is the most important factor in order to protect the seedling from damage. Without site preparation most planting spots consisted of undisturbed humus. Generally, the Karl-Oskar created the most spots with pure mineral soil, but on very stony soils the Bracke T26 created more mineral soil spots than other methods. Site preparation is a valuable tool in order to improve survival in regeneration areas and it is of great importance to make the right choice of technique depending on the particular circumstances on the actual site. 相似文献

Immunodetection of allergens in dark chocolate is complicated by interference from the chocolate components. The objectives of this study were to establish reference materials for detecting multiple allergens in dark chocolate and to determine the accuracy and precision of allergen detection by enzyme-linked immunosorbent assay (ELISA) before and after chocolate processing. Defatted peanut flour, whole egg powder, and spray-dried milk were added to melted chocolate at seven incurred levels and tempered for 4 h. Allergen concentrations were measured using commercial ELISA kits. Tempering decreased the detection of casein and β-lactoglobulin (BLG), but had no significant effect on the detection of peanut and egg. Total coefficients of variation were higher in tempered than untempered chocolate for casein and BLG, but total and analytical CVs were comparable for peanut and egg. These findings indicate that processing has a greater effect on recovery and variability of casein and BLG than peanut and egg detection in a dark chocolate matrix. 相似文献

Mycoplasma ovis is a hemoplasma parasite of sheep, goats, and reindeer; however, natural hemoplasma infection in white-tailed deer has not previously been reported. Subsequent to finding many coccoid, bacillary, and ring-shaped organisms, consistent with hemotropic mycoplasmas, on RBCs from a 72-day-old female white-tailed fawn, we sought to (1) identify the putative hemoplasma observed in blood from the fawn, (2) evaluate others in the herd for hemoplasma infection, and (3) identify clinicopathologic characteristics of hemoplasma-infected white-tailed deer. EDTA-anticoagulated whole blood was collected from the fawn and 8 apparently healthy does in the same herd. CBCs were performed on 7 nonclotted samples from the fawn and 6 does. DNA was extracted from all samples, followed by PCR amplification of bacterial (16S rDNA) and protozoal (18S rDNA) genes. The nearly complete 16S rDNA product from the fawn's sample was directly sequenced and compared with known sequences in the GenBank database. Samples from the fawn and 7 of 8 does were PCR-positive using hemoplasma-specific and M ovis-specific protocols. The fawn was PCR-negative for Anaplasma spp., Babesia spp., and Theileria spp. The 16S rDNA sequence from the fawn (GenBank accession number, FJ824847) was most closely related to M ovis (AF338268), having 98.5% sequence identity. The fawn had a mild nonregenerative anemia, a neutrophilic left-shift with toxic change, aspiration bronchopneumonia, and gastrointestinal disease. Hematologic values, including blood film evaluation, in infected does were unremarkable. The M ovis-like organism may have acted as either an opportunistic or primary pathogen in the fawn. The high occurrence of subclinical infections in the does suggests that white-tailed deer may act as wildlife reservoirs for M ovis. 相似文献

According to the current taxonomy, the Staphylococcus intermedius group (SIG) comprises of at least three distinct species. While S. intermedius and S. pseudintermedius are associated with specific hosts (pigeons and dogs, respectively), the natural host of S. delphini remains unclear. We analysed 158 SIG isolates from less studied animal species belonging to the order Carnivora, including mink (n=118), fox (n=33), badger (n=6) and ferret (n=1). Species identification was performed by nuc PCR in combination with sodA sequence analysis and pta PCR restriction fragment length polymorphism (RFLP). The results showed a consistent association between host and bacterial species. All isolates from minks, ferret and badgers belonged to S. delphini group A, whereas all fox isolates except one were identified as S. pseudintermedius. The remaining fox isolate belonged to S. delphini group A. The results indicate that Mustelidae such as minks, ferrets and badgers are natural hosts of S. delphini group A. This is in contrast with Canidae, which are primarily colonized and infected with S. pseudintermedius. These findings suggest that coagulase-positive staphylococcal species may have evolved and diverged through host adaptation. 相似文献

应用GUS基因标记技术,可简便、快速、准确、原位、直观地确定标记花生根瘤菌株形成的根瘤,从而方便地研究标记菌株与土著根瘤菌的竞争结瘤能力。无氮水培试验表明,标记菌株gusA4-5、gusA2-9分别与土著菌混和接种占瘤率为71.4%、77.0%。盆栽试验表明,接种供试菌株Spr4-5、Spr2-9占瘤率分别为57.9%、63.0%,比对照极显著增产52.5%、22.7%;接种Spr4-5比Spr2-9极显著增产24.2%。初步说明两个供试菌株的竞争结瘤力比土著根瘤菌强,菌株Spr2-9强于Spr4-5;Spr4-5比Spr2-9有效性高,是结瘤适量,竞争结瘤能力强的高效菌株。 相似文献

Urban agriculture in Cuba has rapidly become a significant source of fresh produce for the urban and suburban populations. A large number of urban gardens in Havana and other major cities have emerged as a grassroots movement in response to the crisis brought about by the loss of trade, with the collapse of the socialist bloc in 1989. These gardens are helping to stabilize the supply of fresh produce to Cuba's urban centers. During 1996, Havana's urban farms provided the city's urban population with 8,500 tons of agricultural produce, 4 million dozens of flowers, 7.5 million eggs, and 3,650 tons of meat. This system of urban agriculture, composed of about 8,000 gardens nationwide has been developed and managed along agroecological principles, which eliminate the use of synthetic chemical pesticides and fertilizers, emphasizing diversification, recycling, and the use of local resources. The growth patterns of annually resolved tree rings are good indicators of local environmental changes, making dendrochronology a valuable tool in air pollution research. In the present study, tree-ring analysis was used to assess the effects of 16 years (1991–2007) of chronic nitrogen (N) deposition, and 10 years (1991–2001) of reduced nitrogen input, on the radial growth of Norway spruce (Picea abies (L.) Karst.) and Scots pine (Pinus sylvestris L.) growing in the experimental area of Lake Gårdsjön, southwest Sweden. In addition to the ambient input of c. 15 kg N ha?1 year?1, dissolved NH4NO3 was experimentally added to a 0.52-ha watershed at a rate of c. 40 kg ha?1 year?1. Atmospheric N depositions were reduced by means of a below-canopy plastic roof, which covered a 0.63-ha catchment adjacent to the fertilized site. The paired design of the experiment allowed tree growth in the N-treated sites to be compared with the growth at a reference plot receiving ambient N deposition. Nitrogen fertilization had a negative impact on pine growth, while no changes were observed in spruce. Similarly, the reduction in N and other acidifying compounds resulted in a tendency towards improved radial growth of pine, but it did not significantly affect the spruce growth. These results suggest that spruce is less susceptible to changes in the acidification and N status of the forest ecosystem than pine, at least in the Gårdsjön area. 相似文献
