We attempted to measure in situ the tensile elastic moduli of individual component polymers with a three-dimensional (3D) assembly mode in the cell walls of Sugi (Cryptomeria japonica D. Don) without isolating the polymers. To prepare wood tangential slices [50 × 6 × 0.2 mm (L × T × R)] consisting of lignin with a 3D assembly mode in the cell walls, cellulose and hemicellulose were removed using the method of Terashima and Yoshida (2006) to obtain methylated periodate lignin slices. To prepare wood slices consisting of polysaccharide with a 3D assembly mode in the cell walls, lignin was removed using the method of Maekawa and Koshijima (1983) to obtain holocellulose slices. Static tensile test was applied to determine the elastic moduli of 3D lignin and 3D polysaccharide slices. The followings were revealed. The elastic modulus of the 3D lignin slices was 2.8 GPa, regardless of the microfibril angle (MFA) in the slices. The elastic moduli of the 3D polysaccharide slices with MFAs of 14°, 23°, 34°, and 42° were 18, 12, 9, and 4 GPa, respectively. The former shows that the lignin with a 3D assembly mode behaves as an isotropic substance in the cell walls, while the latter suggests that the 3D polysaccharide slice shows marked anisotropic structure in the cell wall. Despite the fact that cellulose content increased after lignin removal, values of substantial elastic modulus of the cell wall slightly decreased regardless of MFA. Following two possible reasons were pointed out for explaining this phenomenon. First, lignin removal caused an artifactual deterioration in the polysaccharide slices at the level of macromolecular aggregate. Second, rigid and fusiform-shaped cellulose crystallites are dispersed in the soft matrix of amorphous polysaccharide, and those are loosely connected to each other by the intermediary of matrix polysaccharide. Those suggest that the rigid cellulose crystallite can optimize its strong mechanical performance in the polysaccharide framework of the wood cell wall in combination with the ligninification.  相似文献

Some reports have shown that for single species the correlation between modulus of elasticity (MOE) and modulus of rupture (MOR) in bending is quite high. Tropical timbers consist of hundreds of species that are difficult to identify. This report deals with the mechanical stress grading of tropical timber regardless of species. Nine timber species or groups of species with a total number of 1094 pieces measuring 60 × 120 × 3000 mm, were tested in static bending. The MOE was measured flat wise, while MOR was tested edge wise. Statistical analysis of linear regression with a dummy model and analysis of covariance were used to analyze the role of MOE and the effect of species on prediction of MOR. The analysis showed that using MOE as a single predictor caused under/overestimation for one or more species and/or groups of species. The accuracy of prediction would be increased with species identification. An allowable stress and reference resistance for species and/or groups of species were provided to compare with the prediction of strength through timber grading. The timber strength class for species and/or groups of species was also established to support the application of mechanical timber grading.  相似文献

In the present study, we demonstrated that heat shock protein 70 (Hsp70) was present in both canine reticulocytes and mature erythrocytes, and that the canine Hsp70 in reticulocytes was decreased along with the maturation of the cells into erythrocytes. These results suggest that the Hsp70 in canine reticulocytes might act as a chaperone to remove unnecessary proteins during reticulocyte maturation. We also demonstrated that Hsp70 was present in exosomes from reticulocytes during their maturation in in vitro culture. Furthermore, the concentration of Hsp70 in reticulocyte membranes was increased in proportion to an increase of the protein in exosomes until 48 hours after the incubation of reticulocytes in vitro. At 96 hours of the incubation, however, only a trace amount of Hsp70 was detected in the membrane, while a large amount of the protein was present in the exosomes. These results suggest that Hsp70 in canine reticulocytes might play an important role for exosome formation in reticulocytes, resulting in the maturation of the cells.  相似文献

Housedust mites, Dermatophagoides pteronyssinus (D. pteronyssinus) and Dermatophagoides farinae (D. farinae), are the important causative agents of allergic diseases in human and animals. By using 165 dogs suffering from atopic dermatitis (AD), serum levels of immunogloblin E (IgE) antibody against 25 kinds of allergen including housedust mites were determined. Housedust mites were the most frequent allergen against which 90 of the 165 allergic dogs (54.5%) by IMMUNODOT assay. With the further analysis of immunoblotting assay in the 90 dogs sensitized with housedust mites, antigenic proteins of housedust mites recognized by IgE antibodies were with the apparent molecular masses of 15, 76, 90, 98, and 170-kD. Among them, the 15-kD protein that might be identical to Group 2 antigens (Der f2, Der p2) was prominently observed (52/90). This study indicates that about a half of dogs with AD were sensitized to housedust mites, suggesting that Group 2 antigens of housedust mites may be a major allergen in canine AD.  相似文献

A method for sex identification of the Japanese black bear was examined using a polymerase chain reaction (PCR) and sequencing of a part of the amelogenin gene. This gene is located on the X and Y chromosomes, and there are 54 nucleotide deletions on the Y chromosome-specific gene. Forty-seven (26 male and 21 female) DNA samples and 23 (13 male and 10 female) DNA samples, respectively extracted from white blood cells and hairs of Japanese black bears were analyzed. The primers SE47 and SE48 from this X-Y homologous region were used in sex identification by PCR amplification. These primers amplified X- and Y-specific bands, which could be used to discriminate between sexes by a length polymorphism in all samples. We suggest that PCR amplification using the primers SE47 and SE48 is useful for sex determination of the Japanese black bear and could be applied to DNA analysis of small samples such as hairs.  相似文献

A new herbicide, epyrifenacil (S-3100), inhibits protoporphyrinogen oxidase (PPO) in plants. Repeated administration of epyrifenacil in laboratory animals led to some toxicological changes related to PPO inhibition, e.g., hepatotoxicity caused by porphyrin accumulation and anemia caused by the inhibition of heme biosynthesis. In vitro studies revealed that an ester-cleaved metabolite, S-3100-CA, is predominant in mammals, exhibits PPO-inhibitory activity, and thus is the cause of epyrifenacil-induced toxicity. To assess the human risk, the effects of species differences on the dynamics (PPO inhibition) and kinetics (liver uptake) of epyrifenacil were evaluated separately. The results of in vitro assays revealed an approximately tenfold weaker inhibition of PPO by S-3100-CA in humans than in rodents and six- to thirteen-fold less hepatic uptake of S-3100-CA in humans than in mice. Finally, it was suggested that humans are less sensitive to the toxicity of epyrifenacil than are rodents, although further mechanistic research is highly anticipated.  相似文献

Fifty-four slaughtered horses were classified into groups having adipose tissue in the crest of the neck with or without hemorrhage (AH and NH groups, respectively). Blood biochemical tests (Alb, TP, T-bil, GOT, GPT, LDH, T-cho, and BUN) and an epidemiological survey (age, gender, weight, origin, breed, BCS, CNS, and hoof disease) were performed. T-bil tended to be high, while the other parameters were normal. Weight, BCS, and CNS were higher in the AH group (P<0.05). GOT was lower in the AH group (P<0.05). It was suspected that the horses in the AH group had lipomatosis. It was assumed that the adipose tissue of the horses in the AH group contained damaged capillaries, and inflammation was confirmed based on evidence of macrophages and lymphocytes.  相似文献

More than 1300 natural microdiamonds, mostly averaging 0.0015 carat, were studied for transparency to ultraviolet radiation, lambda2537. Of most of them, of this size, 16 percent were completely transparent (presumably type II); 27 percent, completely absorbent (presumably type I). The remainder transmitted partially. The number of type-II diamonds is unexpectedly very high; a considerable proportion of microdiamonds may begin life as type II, later incorporating absorbent type-I material.  相似文献

2-(l-Menthoxy)ethanol has been frequently employed as a flavoring agent; however, data regarding 2-(l-menthoxy)ethanol toxicity remain limited. We performed a 13-week subchronic toxicity study of 2-(l-menthoxy)ethanol in male and female F344 rats, with doses of 0, 15, 60, or 250 mg/kg body weight (BW)/day orally administered by gavage using corn oil as the vehicle. No significant toxicological changes in general condition, body weight, or food intake were observed in any groups. The hematological assessment showed decreased hemoglobin, hematocrit, mean corpuscular volume, and mean corpuscular hemoglobin and increased platelet count in the male 250 mg/kg group. Serum biochemistry revealed elevated total cholesterol in the 250 mg/kg group of male and female rats, reduced triglyceride in the female 250 mg/kg group, and increased total protein in the male 250 mg/kg group, indicating effects on lipid metabolism and protein synthesis. For organ weights, absolute and relative weights of the liver and adrenal glands were increased in the 250 mg/kg group of both sexes and the male 250 mg/kg group, respectively. Histopathological analysis showed chronic nephropathy in the male 15 mg/kg or higher groups, with increased absolute and relative kidney weights, as well as elevated serum creatinine, in the male 60 and 250 mg/kg groups. However, eosinophilic granules containing α2u-globulin were identified in proximal tubules, suggesting α2u-globulin nephropathy specific to male rats and without toxicological significance. These results indicated that the no-observed-adverse-effect level of 2-(l-menthoxy)ethanol was 60 mg/kg BW/day for both sexes.  相似文献
