Activated carbon was derived from waste wood pallets in Hong Kong via phosphoric acid activation and applied to adsorption of basic dye (methylene blue), acid dyes (acid blue 25 and acid red 151), and reactive dye (reactive red 23). The results showed that respective adjustment in phosphoric acid concentration, impregnation ratio, activation temperature, and activation time could maximize the surface area and pore volume of activated carbon. An increase of impregnation ratio or activation temperature significantly influenced the pore size distribution by expanding the porous structure and creating more macropores than micropores. The characterization of the carbon surface chemistry using Fourier-transform infrared (FTIR) spectroscopy, however, revealed a decrease in the amount of several functional groups with increasing activation temperature. The physical properties (surface area and pore volume) of the wood waste-derived activated carbon (using 36% phosphoric acid with an impregnation ratio of 1.5 at an activation temperature of 550°C for 1.5 h) were comparable to those of commercial activated carbon (Calgon F400). The contrasting pH effects on the adsorption of different classes of dyes signified the importance of both electrostatic interaction and chemical adsorption, which correlated to pH-dependent dissociation of surface functional groups. It is noteworthy that the physical properties of activated carbon were insufficient to account for the observed dye adsorption behavior, whereas the surface chemistry of activated carbon and the nature and chemical structure of dyes were more important. The fast kinetics and high capacity of dye adsorption of wood waste-derived activated carbon suggest that production of activated carbon from different types of wood waste should merit further investigation.  相似文献

The effect of seed source (half-sib family) and nursery bed density on the nursery stock quality and subsequent growth of black walnut (Juglans nigra L.) was investigated. Nine black walnut female genitors were selected to represent a range of phenotypes. Seeds were collected, cleaned, cold-treated, and pre-sprouted to ensure that germination was uniform and complete. The pre-sprouted seeds were planted in a randomized complete block design into standard nursery beds in Indiana, USA at three sowing densities, (11.2 plants m−2; 24.2 plants m−2; 29.4 plants m−2). After lifting, the trees were measured for height, ground-line diameter and root volume and then planted into a plantation in a randomized complete block design and re-measured after 1 and 5 years of growth. One year after planting, the effects of family (half-sib seed source) and density were significant or very highly significant for seedling height, and ground-line diameter, although family effects were greater than those for density, especially at moderate and high nursery bed density. After 5 years of growth, the same effects contributed significantly to ground-line diameter and dbh, but only family significantly influenced height. Family was more important than nursery bed density in determining the size of the trees after 5 years. Although there were no significant family × density interactions after 5 years, family variance for all the traits was considerably higher among seedlings grown at moderate and high density in the nursery. Phenotypic correlations among traits within and among years were generally very high (0.65 < r < 0.90) and insensitive to planting density in the nursery.  相似文献

Climate change is predicted to cause upward shifts in forest tree distributions, which will require seedling recruitment beyond current forest boundaries. However, predicting the likelihood of successful plant establishment beyond current species' ranges under changing climate is complicated by the interaction of genetic and environmental controls on seedling establishment. To determine how genetics and climate may interact to affect seedling establishment, we transplanted recently germinated seedlings from high- and low-elevation provenances (HI and LO, respectively) of Pinus flexilis in common gardens arrayed along an elevation and canopy gradient from subalpine forest into the alpine zone and examined differences in physiology and morphology between provenances and among sites. Plant dry mass, projected leaf area and shoot:root ratios were 12-40% greater in LO compared with HI seedlings at each elevation. There were no significant changes in these variables among sites except for decreased dry mass of LO seedlings in the alpine site. Photosynthesis, carbon balance (photosynthesis/respiration) and conductance increased >2× with elevation for both provenances, and were 35-77% greater in LO seedlings compared with HI seedlings. There were no differences in dark-adapted chlorophyll fluorescence (Fv/Fm) among sites or between provenances. Our results suggest that for P. flexilis seedlings, provenances selected for above-ground growth may outperform those selected for stress resistance in the absence of harsh climatic conditions, even well above the species' range limits in the alpine zone. This indicates that forest genetics may be important to understanding and managing species' range adjustments due to climate change.  相似文献

Single-molecule DNA sequencing of a viral genome   总被引:4,自引:0,他引:4  
The full promise of human genomics will be realized only when the genomes of thousands of individuals can be sequenced for comparative analysis. A reference sequence enables the use of short read length. We report an amplification-free method for determining the nucleotide sequence of more than 280,000 individual DNA molecules simultaneously. A DNA polymerase adds labeled nucleotides to surface-immobilized primer-template duplexes in stepwise fashion, and the asynchronous growth of individual DNA molecules was monitored by fluorescence imaging. Read lengths of >25 bases and equivalent phred software program quality scores approaching 30 were achieved. We used this method to sequence the M13 virus to an average depth of >150x and with 100% coverage; thus, we resequenced the M13 genome with high-sensitivity mutation detection. This demonstrates a strategy for high-throughput low-cost resequencing.  相似文献

Cell migration is a highly integrated multistep process that orchestrates embryonic morphogenesis; contributes to tissue repair and regeneration; and drives disease progression in cancer, mental retardation, atherosclerosis, and arthritis. The migrating cell is highly polarized with complex regulatory pathways that spatially and temporally integrate its component processes. This review describes the mechanisms underlying the major steps of migration and the signaling pathways that regulate them, and outlines recent advances investigating the nature of polarity in migrating cells and the pathways that establish it.  相似文献

The diversity of monomers available for synthesis of high polymers makes it possible to prepare a wide variety of long-chain macromolecular compounds. It is instructive to consider a hierarchical organization of structure in polymers at four successive levels--the molecular, nano-, micro-, and macrolevels--and to examine how interactions at and between these various levels of structure have important and often quite specific influences. Examples are drawn from semicrystalline polymers with flexible chains, liquid-crystalline polymers composed of rigid macromolecules, and amorphous polymers. Structural hierarchies in biocomposite systems are also discussed, particularly in soft connective tissues such as tendon and intervertebral disk.  相似文献
