In the auditory epithelium of the cochlea, the sensory hair cells and supporting cells are arranged in a checkerboard-like fashion, but the mechanism underlying this cellular patterning is unclear. We found that mouse hair cells and supporting cells express the immunoglobulin-like adhesion molecules nectin-1 and -3, respectively, and that their interaction mediates the heterotypic adhesion between these two cell types. Genetic removal of nectin-1 or -3 disrupted the checkerboard-like pattern, inducing aberrant attachment between hair cells. When cells expressing either nectin-1 or -3 were cocultured, they arranged themselves into a mosaic pattern. Thus, nectin-1 and -3 promote the formation of the checkerboard-like pattern of the auditory epithelia.  相似文献

Structural and physicochemical characteristics of endosperm starch from milled rice grains of seven Japanese cultivars used in sake production were examined. Amylose content was 15.2–20.2%, number-average degree of polymerization (DPn) of amylose was 900–1,400, and the ratio of short-to-long chain amylopectin was 2.7–3.5, respectively. The degree of retrogradation of purified starch stored for seven days at 4°C after gelatinization was 20–31%. The degree of retrogradation correlated negatively with the ratio of short-to-long chain amylopectin. The effect of holding time after steaming on enzyme digestibility and starch retrogradation of steamed rice grains was investigated. The longer the holding time after steaming, the greater the extent of retrogradation, and the less the degree of enzymatic digestibility. The decreased rate of enzyme digestibility correlated with amylopectin chain length distribution. Samples with short-chain amylopectin exhibited a slow decrease in enzyme digestibility. It was determined that the structure and retrogradation properties of endosperm starch in Japanese rice cultivars affect the decreasing rate of enzyme digestibility of the steamed, milled rice grains.  相似文献

The aim of the present study was to examine the production efficiency of cloned pigs by serial somatic cell nuclear transfer (SCNT) and to ascertain any changes in the telomere lengths of multiple generations of pigs. Using fetal fibroblasts as the starting nuclear donor cells, porcine salivary gland progenitor cells were collected from the resultant first-generation cloned pigs to successively produce second- and third-generation clones, with no significant differences in production efficiency, which ranged from 1.4% (2/140) to 3.3% (13/391) among the 3 generations. The average telomere lengths (terminal restriction fragment values) for the first, second and third generation clones were 16.3, 18.1 and 20.5 kb, respectively, and were comparable to those in age-matched controls. These findings suggest that third-generation cloned pigs can be produced by serial somatic cell cloning without compromising production efficiency and that the telomere lengths of cloned pigs from the first to third generations are normal.  相似文献

Moisture distribution in individual polished rice grains was observed during soaking by magnetic resonance (MR) imaging, and a nuclear magnetic resonance (NMR) signal intensity profile (SI-profile) was generated from the MR image. The water penetration pattern during soaking roughly showed dissimilar trends between different varieties of japonica and japonica-indica hybrid rice. NMR signal intensity at the completion of water absorption varied within each grain; high at the periphery and the central region and low in the area between the periphery and the central region. High moisture content within the central region is due to loosely packed starch granules. The SI-profile was congruent for grains of the same variety harvested in different regions and years and characterized a grain moisture distribution for each variety. Moisture distribution was compared using SI-profiles for varieties with different amylose contents and new varieties bred for specific end-uses in Japan. The NMR signal intensity, which is related to the moisture content, at the surface of soaked grain was negatively correlated to the grain amylose content. The NMR signal intensities at the surface of soaked grain negatively correlated with the overall hardness of the cooked rice grain as measured by the single-grain low-high compression test.  相似文献

Isolation of Rhodococcus equi from the submaxillary lymph nodes of pigs, with or without caseous lymphadenitis, and typing of the isolates by two serological methods were carried out. The rate of isolation of the organisms from the lymph nodes of pigs was 5 times higher in the lymph nodes with caseous lymphadenitis than in those without the lesion. Of 219 isolates, 146 (66.7%) were typable by the method of Prescott, while all the 219 isolates (100%) were typable by the method of Nakazawa et al. The most frequently isolated were serotype 2 of Prescott (identical to serogroup 16 of Nakazawa et al.), and serogroup 3 of Nakazawa et al., which did not correspond with any serotypes of Prescott. Serotypes/serogroups of R. equi from pigs were thus first clarified in Japan.  相似文献

It is an urgent priority to establish in vivo bioassays for detection of hazards related to fine particles, which can be inhaled into deep lung tissue by humans. In order to establish an appropriate bioassay for detection of lung damage after particle inhalation, several experiments were performed in rats using quartz as a typical lung toxic particle. The results of pilot experiments suggest that Days 1 and 28 after intratracheal instillation of 2 mg of fine test particles in vehicle are most appropriate for detection of acute and subacute inflammatory changes, respectively. Furthermore, the BrdU incorporation on Day 1 and the iNOS level on Day 28 proved to be suitable end-point markers for this purpose. An examination of the toxicity of a series of particles was performed with the developed bioassay. Although some materials, including nanoparticles, demonstrated toxicity that was too strong for sensitive assessment, a ranking order could be clarified. The bioassay thus appears suitable for rapid hazard identification with a possible ranking of the toxicity of various particles at single concentrations.  相似文献

Serratia marcescens strain B2 is an antagonistic bacterium that produces the red-pigmented antibiotic prodigiosin and suppresses rice sheath blight caused by Rhizoctonia solani AG-1 IA. Rice sheath blight disease was suppressed when plants were inoculated with this bacterium an hour before pathogen inoculation but not when plants were treated 4 weeks before pathogen inoculation. In both cases the bacteria were detected in the rice rhizosphere 4 weeks after inoculation. Bacteria isolated from the rice plant and rhizosphere inhibited biosynthesis of prodigiosin in S. marcescens strain B2. We suggest that bacteria isolated from rice plants and rhizospheres mediate the suppression of antibiotic production of biological control agents and that such suppression is common under field conditions.  相似文献

Chitinolytic activity of the biocontrol bacterium Serratia marcescens strain B2 was inhibited by bacteria isolated from rice, even though its growth was not affected. Antifungal activity of the strain against Pyricularia oryzae was also reduced under the influence of these bacteria. In contrast, the rice-epiphytic bacterium Erwinia ananas NR1, transformed with chitinase gene chiA derived from strain B2, had high chitinolytic activity regardless of the presence of the bacteria isolated from rice. Therefore, the introduction of an antagonistic factor gene with a promoter from the recipient into the epiphytic bacteria may prove useful in the development of effective biocontrol agents.  相似文献

In vitro cultured shoots of three Chinese poplar species (Populus tomentosa, P. alba cv. Pyramidalis andP. euphratica) and an improved poplar (FS-51), as a control, were exposed to the stress of low osmotic potential, salinity and high pH, and the shoot growth was measured.P. tomentosa andP. euphratica could grow even under the high osmotic stress (0.5m mannitol; −1.7 MPa).P. euphratica shoots which were previously cultured on the medium containing 0.7m mannitol (−2.4 MPa) recovered its growth after being transplanted to the medium without mannitol. The shoot growth ofP. euphratica was not so reduced even on the medium containing 100 mM NaCl. However, growth of the other poplars markedly reduced with 10 mM NaCl. Medium pH (pH 5.8–10) also did not affect the shoot growth ofP. euphratica. These poplars have different tolerance to the stresses in cultured shootin vitro. P. euphratica was extremely tolerant to the stresses which are possible to damage plants in dry land. A part of this study was presented at the 104th Annual Meeting of the Japanese Forestry Society.
