Recovering native uniqueness has major importance for breeds with historic introgression. The aim of the study was to estimate population genetic parameters for two local red cattle breeds from Northern Germany and to study possibilities to reverse introgression. Genealogical information consisted of 90,783 individuals for German Angler and 187,255 individuals for Red Dual-Purpose cattle breed, with additional information on sex, born, breed, status, and conventional breeding values. It is concluded that the native genetic contribution could be included as an additional trait in the total merit index in order to recover a part of the native genetic background. Native contributions should be estimated in the long term from marker data in order to account for Mendelian sampling. The maintenance of a sufficient genetic diversity of native alleles can be achieved by an advanced OCS with appropriate constraints.  相似文献

Fire blight caused by the gram-negative bacterium Erwinia amylovora is a major disease of pome fruit like apple (Malus domestica) or pear (Pyrus communis). Detection of the pathogen is hampered by low titres usually being present during initial flower infection and the brief time frame for analysis to decide upon subsequent countermeasures. Here we describe a loop-mediated isothermal amplification of DNA (LAMP) assay for genomic DNA of Erwinia amylovora, which relies on a highly specific primer design that excludes amplification from typical DNA sources of the orchard biological system and from sample handling. The assay enables the fast detection of down to approximately 20?cfu of pure Erwinia amylovora or 100?fg genomic DNA (corresponding to approximately 25 E. amyolovora equivalents) per reaction within 45?min. Fast and reliable detection of E. amylovora in orchard samples by naked eye is achieved through a visual colour change indication with hydroxynaphthol blue. The assay avoids use of complex technical devices and is thus suitable for field testing.  相似文献

Permafrost thaw is expected to alter biogeochemistry and hydrology, potentially increasing the mobility of soil constituents. Northern latitude boreal forests where permafrost thaw is occurring also experience extreme changes in day length during the growing season. As the effects of photoperiod on plant uptake of soil constituents or interactions with the rhizosphere are unknown, our objective was to determine these interactions with three plant species from different functional groups. A tree, forb, and grass common to military training ranges in this region were grown in soil spiked with or without lead, antimony, or 2,4-dinitrotoluene and grown under 16, 20, or 24 h of light. Plant biomass, soil constituent uptake, and rhizosphere bacterial communities were compared between treatments. Photoperiod had no effect on plant uptake of any soil constituent or on rhizosphere community, indicating that plants and their associated microbial communities adapted to this environment are resilient to extremes in photoperiod. Lead uptake was not significant in any plant species and had no effect on the rhizosphere. Antimony increased the percentage composition of Saprospirales in the rhizospheres of two of the three plants, indicating an interaction between this bacterial order and antimony. Antimony uptake by white spruce (Picea glauca) was considerable, with a mean concentration of 1731 mg kg?1 in roots, while mean shoot concentration was only 155 mg kg?1, indicating its potential to phytostabilize this heavy metal. Although antimony had the strongest impact on the rhizosphere bacterial community, it was also readily accumulated by the grass and tree.  相似文献

Summary The mission was carried out within the Mediterranean Programme of the IBPGR in April/May 1983 with participants from the Socialist People's Libyan Arab Jamahiriya (Agricultural Research Centre, Tarabulus), the Istituto del Germoplasma, Bari, and the Zentralinstitut für Genetik und Kulturpflanzenforschung, Gatersleben. In Tripolitania, the Fezzan- and Ghadmis-areas 316 samples, mostly cereals and vegetables, have been collected. In Tripolitania and the Ghadmis-area the land-races have been nearly completely replaced by new varieties. In the Fezzan genetic erosion is in progress. There, several cereal- and vegetable-land-races could still be collected. The land-races may be useful for breeding programmes.
Bericht über eine Reise in die Sozialistische Libysche Arabische Volksjamahiriya 1983 zur Sammlung autochthoner Sippen von Kulturpflanzen
Zusammenfassung Die Reise erfolgte im Rahmen des Mediterranean Programme des IBPGR im April/Mai 1983 mit Teilnehmern aus der Sozialistischen Libyschen Arabischen Volksjamahiriya (Agricultural Research Centre, Tarbulus), des Istituto del Germoplasma, Bari, und des Zentralinstituts für Genetik und Kulturpflanzenforschung, Gatersleben. In Tripolitanien, dem Fezzan und dem Gebiet von Ghadmis wurden 316 Proben, meist von Getrieden und Gemüsen, gesammelt. In Tripolitanien und dem Gebiet von Ghadmis sind die Landsorten fast vollständig von neueren Sorten verdrängt worden. Im Fezzan schreitet die Generosion fort. Hier konnten noch zahlreiche Getreide- und Gemüselandsorten gesammelt werden. Die Landsorten können für Züchtungsprogramme nützlich sein.

OBJECTIVE: To evaluate the effect of 2 hydroxyethyl starch (HES) preparations (ie, HES solution with a molecular weight of 600 kd and a degree of substitution of 0.7 [HES 600/0.7] and a calcium-containing polyionic HES solution with a molecular weight of 670 kd and a degree of substitution of 0.75 [HES 670/0.75]) on canine platelet function. SAMPLE POPULATION: Blood samples from 10 healthy adult dogs. PROCEDURES: Dilution of citrated whole blood was performed with saline (0.9% NaCl) solution, HES 600/0.7, and HES 670/0.75 at ratios of 1:9 (ie, 1 part saline solution or colloid to 9 parts whole blood) and 1:3. Measurements of time to platelet plug formation in a capillary tube (ie, closure time) were made by use of a bench-top platelet function analyzer with collagen and ADP platelet agonists. RESULTS: Mean baseline closure time was 68.0 +/- 15.3 seconds. A 1:3 dilution of whole blood with saline solution, HES 600/0.7, and HES 670/0.75 resulted in mean closure times of 85.8 +/- 15.7 seconds, 100.6 +/- 18.6 seconds, and 101.6 +/- 16.2 seconds, respectively. Closure time following 1:3 dilution of whole blood with saline solution was significantly different from baseline and from 1:9 dilution with saline solution. Closure time following 1:3 dilution of whole blood with HES 670/0.75 was significantly different from baseline, 1:3 and 1:9 dilutions with saline solution, and 1:9 dilutions with HES 600/0.7 or HES 670/0.75. CONCLUSIONS AND CLINICAL RELEVANCE: Saline solution, HES 600/0.7, and HES 670/0.75 affect canine platelet function by prolonging closure times; HES solutions prolonged closure time to a greater extent than saline solution.  相似文献

Leaf K/Na ratio predicts salinity induced yield loss in irrigated rice   总被引:7,自引:0,他引:7  
Salinity is a major constraint to irrigated rice production, particularly in semi-arid and arid climates. Irrigated rice is a well suited crop to controlling and even decreasing soil salinity, but rice is a salt-susceptible crop and yield losses due to salinity can be substantial. The objective of this study was to develop a highly predictive screening tool for the vegetative growth stage of rice to estimate salinity-induced yield losses. Twenty-one rice genotypes were grown over seven seasons in a field trials in Ndiaye, Senegal, between 1991 and 1995 and were subjected to irrigation with moderately saline water (3.5 mS cm-1, electrical conductivity) or irrigation with fresh water. Potassium/sodium ratios of the youngest three leaves (K/NaLeaves) were determined by flame photometry at the late vegetative stage. Grain yield was determined at maturity. All cultivars showed strong log-linear correlations between K/NaLeaves and grain yield, but intercept and slope of those correlations differed between seasons for a given genotype and between genotypes. The K/NaLeaves under salinity was related to grain yield under salinity relative to freshwater controls. There was a highly significant correlation (p < 0.001) between K/NaLeaves and salinity-induced grain yield reduction: the most susceptible cultivars had lowest K/NaLeaves and the strongest yield reductions. Although there were major differences in the effects of salinity on crops in both the hot dry season (HDS) and the wet season, the correlation was equally significant across cropping seasons. The earliest possible time to establish the relationship between K/NaLeaves under salinity and grain yield reduction due to salinity was investigated in an additional trial in the HDS 1998. About 60 days after sowing, salinity-induced yield loss could be predicted through K/NaLeaves with a high degree of confidence (p < 0.01). A screening system for salinity resistance of rice, particularly in arid and semi-arid climates, is proposed based on the correlation between K/NaLeaves under salinity and salinity-induced yield losses. Understanding and quantification of phosphorus (P) fluxes are key requirements for predictions of future forest ecosystems changes as well as for transferring lessons learned from natural ecosystems to croplands and plantations. This review summarizes and evaluates the recent knowledge on mechanisms, magnitude, and relevance by which dissolved and colloidal inorganic and organic P forms can be translocated within or exported from forest ecosystems. Attention is paid to hydrological pathways of P losses at the soil profile and landscape scales, and the subsequent influence of P on aquatic ecosystems. New (unpublished) data from the German Priority Program 1685 “Ecosystem Nutrition: Forest Strategies for limited Phosphorus Resources” were added to provide up‐to‐date flux‐based information. Nitrogen (N) additions increase the release of water‐transportable P forms. Most P found in percolates and pore waters belongs to the so‐called dissolved organic P (DOP) fractions, rich in orthophosphate‐monoesters and also containing some orthophosphate‐diesters. Total solution P concentrations range from ca. 1 to 400 µg P L?1, with large variations among forest stands. Recent sophisticated analyses revealed that large portions of the DOP in forest stream water can comprise natural nanoparticles and fine colloids which under extreme conditions may account for 40–100% of the P losses. Their translocation within preferential flow passes may be rapid, mediated by storm events. The potential total P loss through leaching into subsoils and with streams was found to be less than 50 mg P m?2 a?1, suggesting effects on ecosystems at centennial to millennium scale. All current data are based on selected snapshots only. Quantitative measurements of P fluxes in temperate forest systems are nearly absent in the literature, probably due to main research focus on the C and N cycles. Therefore, we lack complete ecosystem‐based assessments of dissolved and colloidal P fluxes within and from temperate forest systems.  相似文献
