The availability of very-large-scale aerial (VLSA) imagery (typically less than 1 cm ground-sampling-distance spatial resolution) and techniques for processing those data into ecosystem indicators has opened the door for routinely using VLSA imagery in rangeland monitoring and assessment. However, for VLSA imagery to provide defensible information for managers, it is crucial to understand the statistical implications of designing and implementing VLSA image studies, including consideration of image scale, sample design limitations, and the need for validation of estimates. A significant advantage of VLSA imaging is that the researcher can specify the scale (i.e., spatial resolution and extent) of the images. VLSA image programs should plan for scales that match monitoring questions, size of landscape elements to be measured, and spatial heterogeneity of the environment. Failure to plan for scale may result in images that are not optimal for answering management questions. Probability-based sampling guards against bias and ensures that inferences can be made to the desired study area. Often collected along flight transects, VLSA imagery lends itself well to certain probability-based sample designs, such as systematic sampling, not often used in field studies. With VLSA image programs, the sample unit can be an entire image or a portion of an image. It is critical to define the sampling unit and understand the relationship between measurements and estimates made from the imagery. Finally, it is important to statistically validate estimates produced from VLSA images at selected locations using quantitative data of the same scale and more precise and accurate than the VLSA image techniques. The extent to which VLSA imagery will be useful as a tool for understanding the status and trend of rangelands depends as much on the ability to build the imagery into robust programs as it does on the ability to quickly and relatively easily collect VLSA images over large landscapes.  相似文献

Adapting what we currently know about ecosystems to a future where rangelands are changing is a new frontier in rangeland management. Current tools for knowledge discovery and application are limited because they cannot adequately judge ecological relevance of knowledge to specific situations. We propose development of integrated knowledge systems (KSs)—collections of resources (e.g., data, analytical tools, literature) drawn from disparate domains and organized around topics by process-based conceptual models. An integrated KS would define relevance by ecological attributes (e.g., soils, climate, vegetation) and location as a flexible mechanism for organizing, finding, and applying knowledge to rangeland management. A KS provides knowledge sources within a decision-making framework that defines what knowledge is needed and how it will be used to make decisions. Knowledge from a KS can identify appropriate spatial and temporal scales to address specific resource questions or objectives. Several factors currently limit KS development and implementation. These include limited interoperability of disparate information and knowledge systems; lack of consistent geographic referencing of knowledge; incomplete and inconsistent documentation of the origin, history and meaning of data and information; underexploited application of remote sensing products; limited ability to extrapolate and share local knowledge and unstructured information; and lack of training and education of professionals that can link ecological and technical fields of study. The proposed KS concept and recommendations present an opportunity to take advantage of emerging technologies and the collective knowledge of rangeland professionals to address changing ecosystems and evolving threats. If we keep on with a “business as usual” approach to finding and using information, we will struggle to meet our responsibilities as rangeland professionals.  相似文献

Residues of Leucaena (Leucaena leucocephala (L), Senna siamea (S) and maize stover (M) were tested to evaluate their effect on soil organic matter accumulation and composition under sub-humid tropical conditions. On an Imperata cylindrica (I) dominated grass fallow, a total amount of 30?Mg?ha?1 DM were applied within 18?months. Two months after the last application, changes in the light and heavy soil organic carbon fraction (LF and HF) and in the total soil organic carbon content (LF?+?HF) in the topsoil were observed. All organic materials increased the proportion of the LF fraction in the soil significantly. The increase in HF was 39 to 51% of the increase in total organic carbon, depending on the source of the organic material. The potential of the tested organic materials to increase total soil organic carbon content (including all soil organic carbon fractions) was in the order L?>?S?>?M?>?I, whereas the order of increase of the HF fraction was L?=?S?>?I?>?M. Cation exchange capacity of the newly formed heavy soil organic carbon was highest with L and lowest with M. Ranking of the transformation efficiency of applied plant residues into the heavy soil organic carbon fraction was I?>?L?=?S?>?M. Transformation efficiency of the residues could neither be explained by lignin nor lignin/N ratio, but rather by extractable polyphenols (Folin?CDenis extraction). The results show that accumulation of the HF fraction in tropical soils is feasible through the application of large quantities of plant residues, but depends strongly on the composition of the applied materials.  相似文献

Avian aspergillosis, most often caused by Aspergillus fumigatus, is a common and devastating disease affecting a range of bird species. Early diagnosis is difficult and often unreliable. The current study evaluated the utility of measuring (1-->3)-beta-D-glucan (BG) concentrations in avian plasma samples to aid in the diagnosis of aspergillosis. We evaluated a commercially available BG assay (Fungitell, Beacon Diagnostics) using 178 plasma samples from naturally infected, experimentally infected, and aspergillosis-free birds. Although there was variation in BG concentration, as reflected by high standard deviations, seabirds with confirmed aspergillosis had the highest mean BG concentrations (M = 3098.7 pg/dl, SD = 5022.6, n = 22) followed by companion avian species and raptors with confirmed aspergillosis (M = 1033.8 pg/dl, SD = 1531.6, n = 19) and experimentally infected Japanese quail (Coturnix japonica; M = 1066.5 pg/dl, SD = 1348.2, n = 17). Variation in severity of disease, differences among species of birds with and without disease, and also different levels in environmental exposure likely contribute to the differences among avian groups. The overall sensitivity and specificity of the BG test for diagnosis of aspergillosis in birds was 60.0 and 92.7%, respectively, with an overall optimized avian cut-offvalue of > or = 461 pg/dl for positive disease. Our findings suggest that, although BG concentrations are highly variable between and within different avian groups, it could serve as a useful adjunctive diagnostic test for aspergillosis that is applicable to multiple avian species in some settings, particularly as a negative predictor of infection.  相似文献

The Toll-like receptor (TLR)4 is critical for the recognition of Gram-negative bacterial lipopolysaccharide (LPS) but in porcine peripheral blood mononuclear cells (PBMCs) it may cooperate with other TLRs and lead to the production of inflammatory cytokines. Therefore, we analyzed TLR1-10 mRNA expression in porcine PBMCs stimulated with LPS over time (1-48 h) by using quantitative real-time PCR and cytokine proteins level by ELISA in culture supernatant. TLR1-10 mRNA was detectable in porcine PBMCs. When compared with the control (non-stimulated), TLR1 mRNA were increased (p<0.05) at 3 h after challenge with 1 μg/ml LPS, whereas TLR1 and TLR2 mRNA were increased (p<0.01) at 6 h after challenge with 10 μg/ml LPS. TLR4 increased (p<0.001) at 3h after challenge with LPS and remained constant. TLR5 and TLR6 mRNA increased (p<0.05) at 9 h and 1 h after of LPS stimulation, respectively. The mRNA of CD14 and MD2 were increased (p<0.001) at 1h after LPS stimulation. Additionally, at most of the time analyzed, the mRNA expression increased with the dose of LPS. The LPS concentration had influence (p<0.05) on all the TLRs expression except TLR10; whereas time had effect (p<0.05) on all TLRs expression except TLR2, 3, 6 and 10. When compared to the control, the cytokines IL1b, IL8 and TNFα proteins were increased (p<0.001) immediately at 1 h after LPS stimulation and remained constant till 48 h. IL12b was increased (p<0.001) 12 h after challenge with 10 μg/ml of LPS. Although IL8 level was the highest, the higher (p<0.05) expression of all these inflammatory cytokines indicate that upon interacting with TLRs, LPS exerted inflammatory response in PBMCs through the production of Th1 type cytokines. The production of cytokines was influenced (p<0.001) by both the dose of LPS and the stimulation time. Hence, the porcine PBMCs are likely able to express all members of TLRs.  相似文献

Farmers within the Inland Pacific Northwest are gradually transitioning to direct seed (DS) practices that reduce soil disturbance and increase surface residue compared to conventional tillage (CT). Despite this transition the impacts of DS practices on soil properties and fauna in commercial fields has been little studied in the region. During the spring and summer of 2002 and 2003 we compared soil organic carbon (SOC), total nitrogen (TN), pH, and earthworm and cocoon densities in CT and DS fields planted to either spring wheat or pea in the Palouse region of northern Idaho. In 2002 mean SOC within the 0–10-cm depth was greater in DS fields (2.05%) than at the same depth in CT fields (1.79%), however SOC within the 30–40-cm depth was lower under DS compared to CT. Mean soil pH within the 0–10-cm depth was 5.35 under DS and 5.61 under CT indicating that pH stratification can occur when tillage is reduced. Tillage effects on SOC, TN, and pH were not found in 2003. Tillage also did not significantly influence earthworm densities, which averaged 39 individuals m−2 in 2002 and 57 individuals m−2 in 2003. Correlations were detected in 2003 DS fields between soil properties (SOC and TN) and earthworm and cocoon densities at depths above 30 cm while in 2002 correlations in DS fields occurred with cocoon density, but not with earthworm density. Direct seed management can increase near-surface SOC and TN concentrations compared to CT practices, however, SOC concentrations deeper in the soil appear to remain the same or possibly decrease. Higher SOC and TN near the soil surface, as found in DS fields, appear to promote greater earthworm densities, which may improve long-term soil productivity.  相似文献

OBJECTIVE: To review the role of mesenchymal stem cells (MSC) in bone formation and regeneration, and outline the development of strategies that use MSC in bone healing and regeneration. STUDY DESIGN: Literature review. METHODS: Medline review, synopses of authors' published research. RESULTS: The MSC is the basic cellular unit of embryologic bone formation. Secondary bone healing mimics bone formation with proliferation of MSC then their differentiation into components of fracture callus. Bone regeneration, where large amounts of bone must form, mimics bone healing and can be achieved with MSC combined with strategies of osteogenesis, osteoinduction, osteoconduction, and osteopromotion. MSC based strategies first employed isolated and culture expanded stem cells in an osteoconductive carrier to successfully regenerate a critical segmental defect in the femur of dogs, which was as effective as autogenous cancellous bone. Because MSC appeared to be immunologically privileged, a study using mismatched allogeneic stem cells demonstrated that these cells would regenerate bone without inciting an immunologic response, documenting the possibility of banked allogeneic MSC for bone regeneration. A technique was developed for selectively retaining MSC from large bone marrow aspirates at surgery for bone regeneration. These techniques utilized osteoconductive and osteoinductive carriers and resulted in bone regeneration that was similar to autogenous cancellous bone. CONCLUSION: MSC can be manipulated and combined with carriers that will result in bone regeneration of critically sized bone defects. CLINICAL RELEVANCE: These techniques can be employed clinically to regenerate bone and serve as an alternative to autogenous cancellous bone.  相似文献

Complete quantification of group A and group B soyasaponins in soybeans   总被引:1,自引:0,他引:1  
A combination of high-pressure extraction and preparative chromatography was used to purify the group A and group B soyasaponins from soy germ for use as analytical standards and for use in biological assays. A standardized sample preparation and extraction method was developed for the analysis of phytochemicals found in soy and processed soy products, which is reproducible in other laboratories. The extracts can be analyzed with standard liquid chromatography-mass spectrometry and high-performance liquid chromatography methods to identify and quantitate the group A and group B forms of the soy saponins, as well as the soy isoflavones. Complete saponin analysis of the extracts prepared from soy germ (hypocots), hulls, and cotyledons shows that a significant portion of the saponins is concentrated in the germ. The germ contains nearly all of the group A soyasaponins, while the group B soyasaponins are nearly equally distributed between the germ and the cotyledons. The hulls contain little of either isoflavones or saponins. Whole (full fat) soybeans grown on a tract in central Illinois in 2003 contain approximately 4-6% saponins on a weight basis, of which about one-fifth or less of the total saponin content are group A soyasaponins; the balance is group B soyasaponins.  相似文献
