Quinoa offers a promising alternative for staple food, considering its outstanding nutritional value and tolerance to abiotic stresses. To develop breeding programmes in quinoa, a reliable crossing method for increasing the genetic variation is required. In the following study, we aimed to develop segregating populations in quinoa. We tested the efficiency of three different crossing methods (hand emasculation, warm water emasculation and no emasculation). Moreover we developed a two-stage selection strategy based on morphological traits and molecular markers for the selection of hybrid plants. We reported hand emasculation to be the most efficient crossing method, followed by warm water emasculation and no emasculation. Our results demonstrated that crosses in quinoa can be successfully performed, despite its complicated flower structure and high self-pollination rate. Additionally, we developed 30 segregating populations from crosses between accessions of different origins with varying phylogenetic relationship, which offers a promising perspective for quinoa breeding programmes in the future.  相似文献

Mountain closure, considered an effective and economic measure for natural restoration of degraded forest ecosystems, has been widely carried out in the karst region of southwest China. The aim of this study was to evaluate microbial aspects of soil quality after mountain closure by analyzing soil microbial biomass, basal respiration, metabolic quotient, and relationships with basic chemical properties in Guizhou Province, a karst region of the upper Yangtze River. Soil quality was considered poor from the low levels of microbial biomass carbon (MBC), nitrogen (MBN), and microbial quotient (MBC/total C and MBN/total N), but high metabolic quotient (qCO2). Soil pH, showing marked variation from 4.1 to 7.9 in this karst region, was proved to significantly affect soil microbial biomass and activity. Soil microbial biomass, microbial quotient, and soil basal respiration declined significantly with decreasing soil pH, while qCO2 showed an apparently increasing, but not statistically significant, trend. The changes in microbial biomass and activity following the change in soil pH could possibly be because of a change in soil microbial composition, and more detailed research is necessary. Compared with soil pH, soil organic matter content was another, more important, factor that directly restricted microbial growth because of the serious loss as a result of disturbance. As a practical application based on microbial aspects, introduction of some N-fixing tree species may be an active and effective measure to improve soil fertility and thus to accelerate restoration of the forest ecosystem in the karst region.  相似文献

The mammalian target of rapamycin (mTOR) protein kinase is a master growth promoter that nucleates two complexes, mTORC1 and mTORC2. Despite the diverse processes controlled by mTOR, few substrates are known. We defined the mTOR-regulated phosphoproteome by quantitative mass spectrometry and characterized the primary sequence motif specificity of mTOR using positional scanning peptide libraries. We found that the phosphorylation response to insulin is largely mTOR dependent and that mTOR exhibits a unique preference for proline, hydrophobic, and aromatic residues at the +1 position. The adaptor protein Grb10 was identified as an mTORC1 substrate that mediates the inhibition of phosphoinositide 3-kinase typical of cells lacking tuberous sclerosis complex 2 (TSC2), a tumor suppressor and negative regulator of mTORC1. Our work clarifies how mTORC1 inhibits growth factor signaling and opens new areas of investigation in mTOR biology.  相似文献

This paper asks whether the technological development of a nation reduces the inter‐regional hierarchy in knowledge flow. We examine two scenarios that are the alternative to each other. The first is what we call the globalization of regional innovation system/weakening of inter‐regional hierarchy scenario: As many regions develop their niches in the global economy, the national “anchor” region loses its relative importance as the importer and distributor of new knowledge, rendering the domestic inter‐regional hierarchy less significant as a result. The second scenario is the globalization of national innovation system/persistence of inter‐regional hierarchy. The nation’s traditional anchor region becomes even more active in importing technology and distributing it to other regions of the country. To test which scenario is closer to reality, we employ social network metrics to analyse inter‐regional technology diffusion networks using Chinese patent licensing data for the 1998–2013 period. Our findings support the second scenario, showing that the influence of the traditional anchor region persists in the hierarchical network structure as new cities enter the network. We found five anchor regions: the three usual suspects—Beijing, Shanghai and Shenzhen—plus two that were slightly less expected—Dongguan and Suzhou.  相似文献

In order to change for the good flavor in green tea of middle or low grade,roast naked barley (miner ingredient) was added to it.To make tea having functional property,mixed roast naked barley-green tea with one medicinal herb (MH,miner ingredient).This study was performed to investigate the effect of drinking extracts of naked barley-MH-green tea (A group) and adlay-Angelica gigas Nakai-green tea (B group) on the body weights of rats,and volatile flavor components of best effective extract on weight reduction.Forty-eight male Sprague-Dawley rats divided into six groups were fed five different green tea extracts for 8 weeks.The result obtained in the experiment indicated that the change of body weights of rats was affected by A group,B group and others.A group showed highest reduction rate of body weights (36.40%).Reduction rate of body weights on the B group (10.10%) was much lower than A group (36.40%).Aroma compounds were extracted by the SDE method.The concentrated aroma extracts were analyzed and identified by GC and GC-MS.The main aroma components of the green tea blended with naked barley and MH were 12 pyrazines having roasty note,methyl butanals having sweet note,hexanal having briskness greenish note,and nerolidol and3-ionone having floral note.  相似文献
