Near-infrared reflectance spectroscopy (NIRS) was evaluated as a possible alternative to gas chromatography (GC) for the quantitative analysis of fatty acids in forages. Herbage samples from 11 greenhouse-grown forage species (grasses, legumes, and forbs) were collected at three stages of growth. Samples were freeze-dried, ground, and analyzed by GC and NIRS techniques. Half of the 195 samples were used to develop an NIRS calibration file for each of eight fatty acids, with the remaining half used as a validation data set. Spectral data, collected over a wavelength range of 1100-2498 nm, were regressed against GC data to develop calibration equations for lauric (C12:0), myristic (C14:0), palmitic (C16:0), stearic (C18:0), palmitoleic (C16:1), oleic (C18:1), linoleic (C18:2), and alpha-linolenic (C18:3) acids. Calibration equations had high coefficients of determination for calibration (0.93-0.99) and cross-validation (0.89-0.98), and standard errors of calibration and cross-validation were < 20% of the respective means. Simple linear regressions of NIRS results against GC data for the validation data set had r2 values ranging from 0.86 to 0.97. Regression slopes for C12:0, C14:0, C16:0, C18:0, C16:1, C18:2, and C18:3 were not significantly different (P = 0.05) from 1.0. The regression slope for C18:1 was 1.1. The ratio of standard error of prediction to standard deviation was > 3.0 for all fatty acids except C12:0 (2.6) and C14:0 (2.9). Validation statistics indicate that NIRS has high prediction ability for fatty acids in forages. Calibration equations developed using data for all plant materials accurately predicted concentrations of C16:0, C18:2, and C18:3 in individual plant species. Accuracy of prediction was less, but acceptable, for fatty acids (C12:0, C14:0, C18:0, C16:1, and C18:1) that were less prevalent.  相似文献

Recent climatic trends and climate model projections indicate that climate change will modify rangeland ecosystem functions and the services and livelihoods that they provision. Recent history has demonstrated that climatic variability has a strong influence on both ecological and social components of rangeland systems and that these systems possess substantial capacity to adapt to climatic variability. Specific objectives of this synthesis are to: 1) evaluate options to mitigate greenhouse gas emissions and future climate change; 2) survey actions that individuals, enterprises, and social organizations can use to adapt to climate change; and 3) assess options for system transformation when adaptation is no longer sufficient to contend with climate change. Mitigation for carbon sequestration does not appear economically viable, given the small and highly variable carbon dioxide fluxes of rangeland ecosystems and the high transaction costs that would be incurred. In contrast, adaptation strategies are numerous and provide a means to manage risks associated with climate change. Adaptation strategies are diverse, including altered risk perception by individuals, greater flexibility of production enterprises, and modifications to social organizations that emphasize climatic variability, rather than consistency. Many adaptations represent “no regrets” actions because their implementation can be justified without emphasis on pending climate change. Adaptations specific to livestock production systems can include flexible herd management, alternative livestock breeds or species, innovative pest management, modified enterprise structures, and geographic relocation. Social-ecological systems in which adaptation is insufficient to counter the adverse consequences of climate change might undergo transformative change to produce alternative ecosystem services, production enterprises, and livelihoods. The rangeland profession is in a pivotal position to provide leadership on this global challenge because it represents the intersection of management and scientific knowledge, includes diverse stakeholders who derive their livelihoods from rangelands, and interacts with organizations responsible for rangeland stewardship.  相似文献

The “gold standard” for verification of patient positioning before linear accelerator‐based stereotactic radiation therapy is kilovoltage cone‐beam computed tomography (kV‐CBCT), which is not uniformly available or utilized; planar imaging is sometimes used instead. The primary aim of this study was to determine if the position of the bony skull, when used as a surrogate for isocenter verification, is different when orthogonal megavoltage (MV) portal or kilovoltage (kV/kV) radiographs are used for image guidance, rather than kV‐CBCT. A secondary aim was to determine the influence of intra‐observer variability, body size and skull conformation on positioning, as determined using these three imaging modalities. Dogs and cats receiving radiotherapy of the head were recruited for this prospective analytical study. Planar (MV portal and kV/kV images) and volumetric (kV‐CBCT) images were acquired before treatment, and manually coregistered with reference images. Differences in skull position when matched based on MV portal, kV/kV images and kV‐CBCT were compared. A total of 65 subjects and 148 unique datasets were evaluated. The Wilcoxon rank‐sum test was used to evaluate effects of transitioning between imaging modalities. When comparing magnitude of shifts in MV to kV‐CBCT, MV to kV/kV and kV/kV to kV‐CBCT, there were statistically significant differences. Results were not measurably impacted by body size, skull conformation or interobserver differences. Based on shift magnitude and direction, an isotropic setup margin of at least 1 mm should be incorporated within the planning target volume when MV or kV planar imaging is used for position verification.  相似文献

Swine are known reservoirs for Clostridioides difficile, formerly known as Clostridium difficile, and transmission from swine to human farm workers is strongly suggested by previous studies. This cross‐sectional study evaluated the potential role of farm environmental surfaces, including those in worker breakrooms and swine housing areas, in the possible transmission of C. difficile from swine to farm workers. Environmental surfaces and piglet faeces at 13 Ohio swine farms were sampled in 2015. Typical culturing techniques were performed to isolate C. difficile from samples, and amplification of toxin genes (tcdA, tcdB and cdtB) and PCR‐ribotyping were used to genetically characterize recovered isolates. In addition, sequencing of toxin regulatory gene, tcdC, was done to identify the length of identified deletions in some isolates. A survey collected farm‐level management risk factor information. Clostridioides difficile was recovered from all farms, with 42% (188/445) of samples testing positive for C. difficile. Samples collected from all on‐farm locations recovered C. difficile, including farrowing rooms (60%, 107/178), breakrooms (50%, 69/138) and nursery rooms (9%, 12/129). Three ribotypes recovered from both swine and human environments (078, 412 and 005) have been previously implicated in human disease. Samples taken from farrowing rooms and breakrooms were found to have greater odds of C. difficile recovery than those taken from nursery rooms (OR = 40.5, OR = 35.6, p < .001 respectively). Farms that weaned ≥23,500 pigs per year had lower odds of C. difficile recovery as compared to farms that weaned fewer pigs (OR = 0.4, p = .01) and weekly or more frequent cleaning of breakroom counters was associated with higher odds of C. difficile recovery (OR = 11.7, p < .001). This study provides important insights into the presence and characterization of C. difficile found in human environments on swine farms and highlights how these areas may be involved in transmission of C. difficile to swine farm workers and throughout the facility.  相似文献

The RAPD-PCR technique has been used to generate DNA amplification profiles from a set of advanced generation inbred lines of white clover ( Trifolium repens L.). The genetic distances between lines have been estimated and have been compared with the extent of positive heterosis for dry matter yield in hybrid crosses. The analysis clearly separated the four major groups (H, J, R and S), with little within group variation. Heterosis, previously shown to be positively correlated with the extent of phenotypic variation between parental lines, was greatest between lines from groups showing the least genetic distance. The lack of correlation between heterosis and genetic divergence is discussed in the context of results from other systems. Additionally, the data allow a comparison of the extent of phenotypic variation for a trait under selective pressure with the degree of variation at the DNA level detected with the RAPD technique. The use of pop‐up archival satellite tags (PSATs) to geolocate marine fishes in polar regions is challenging due to the brevity of periods during which there is a defined sunrise and sunset. Models using other environmental parameters are thus required to supplement geolocation data in the estimation of marine migratory routes. The objective of this work was to create a simple method that would estimate the migratory pathways of Atlantic salmon (Salmo salar) in polar regions using temperature and depth recordings. Validated geolocations from PSATs were used to test and constrict the model. The model’s predicted migratory routes were within 100 km of the light‐based geolocations calculated by the tags. By constraining the trajectories through the geolocations, bias was reduced. Sensitivity analyses demonstrated that slight alterations of the location and timing of the start and end points did not affect the mean migratory route estimates. This method is a management tool that can determine the primary habitat areas for any surface‐ or bottom‐dwelling marine species – especially in polar regions, where other methods may be impossible.  相似文献
