Effective flow-through vacuum degasser for fish hatcheries

Since the fry of certain species of fish cannot tolerate even slight amounts of supersaturation, their water supplies must be degassed. Gas content can be reduced to subsaturated, sublethal levels by passing it through the vacuum degasser described here. The system includes a low-cost, flow-through unit easily capable of degassing (to less than saturated conditions) water flowing at 190 liters min⁻¹. In tests completed to date, dissolved oxygen content was never reduced below 7 ppm.     

Impact of bacteria on short-term storage of salmonid eggs

Unfertilized eggs of steelhead, Oncorhynchus mykiss (Walbaum 1792), were stored at a low temperature (～0°C) with antibiotics (penicillin and streptomycin) for up to 22 days and fertility assessed. Unfertilized eggs of chinook salmon, Oncorhynchus tshawytscha (Walbaum 1792), were stored at a low temperature (～0°C) either with or without antibiotics for up to 24 days and fertility assessed. There were no significant decreases in fertility under these conditions. Steelhead eggs were maintained at 4°C for up to 15 days with or without added bacteria and with or without gentamicin. Gentamicin inhibited bacterial growth during storage at 4°C and its use resulted in a less pronounced decline in fertility with storage. These results suggest that low temperatures and antibiotics play an important role in extending the period over which salmonid eggs can be successfully stored.     

Imaging of small-scale features on 433 Eros from NEAR: evidence for a complex regolith

On 25 October 2000, the Near Earth Asteroid Rendevous (NEAR)-Shoemaker spacecraft executed a low-altitude flyover of asteroid 433 Eros, making it possible to image the surface at a resolution of about 1 meter per pixel. The images reveal an evolved surface distinguished by an abundance of ejecta blocks, a dearth of small craters, and smooth material infilling some topographic lows. The subdued appearance of craters of different diameters and the variety of blocks and different degrees of their burial suggest that ejecta from several impact events blanketed the region imaged at closest approach and led to the building up of a substantial and complex regolith consisting of fine materials and abundant meter-sized blocks.     

Exome sequencing of head and neck squamous cell carcinoma reveals inactivating mutations in NOTCH1

Head and neck squamous cell carcinoma (HNSCC) is the sixth most common cancer worldwide. To explore the genetic origins of this cancer, we used whole-exome sequencing and gene copy number analyses to study 32 primary tumors. Tumors from patients with a history of tobacco use had more mutations than did tumors from patients who did not use tobacco, and tumors that were negative for human papillomavirus (HPV) had more mutations than did HPV-positive tumors. Six of the genes that were mutated in multiple tumors were assessed in up to 88 additional HNSCCs. In addition to previously described mutations in TP53, CDKN2A, PIK3CA, and HRAS, we identified mutations in FBXW7 and NOTCH1. Nearly 40% of the 28 mutations identified in NOTCH1 were predicted to truncate the gene product, suggesting that NOTCH1 may function as a tumor suppressor gene rather than an oncogene in this tumor type.     

Field performance of transgenic Russet Burbank and Lemhi Russet potatoes

Transformed Russet Burbank and Lemhi Russet clones which contained three different transgene constructs were evaluated for performance under field conditions in Idaho. The transgenic lines were characterized over two growing seasons, using plants grown from both greenhouse produced minitubers (first year) and field grown seed (second year). Individual clones were evaluated for a variety of agronomic and quality properties. Many of the transformed clones showed reduced yield and increases in percent malformed and undersized tubers. Other characteristics, such as specific gravity and fry color, showed less variability. The variation observed in non-transgenic clones regenerated from tissue culture was less than that of the transformed lines. Out of an original population of 57 transgenic lines in tissue culture, maintenance of key agronomic and quality properties of the parental material was observed in only four clones. These results suggest that experiments designed to generate transgenic lines for the marketplace should be initiated with a large number of transgenic clones.     

Efficacy of 1,3-dichloropropene plus chloropicrin application methods in combination with pebulate and napropamide in tomato

Small-plot research and large-field validations were conducted to determine the effect of 1,3-dichloropropene (1,3-D)+chloropicrin (Pic) application methods in combination with the herbicides pebulate and napropamide on pest control in fresh market tomato (Lycopersicon esculentum Mill.). In both the small-plot trials and the large-validation fields, various treatments compared the efficacy of soil fumigants and herbicides based on methyl bromide, in-bed and broadcast applications of 1,3-D+Pic, pebulate, and napropamide. The results consistently indicated that either in-bed or broadcast applications of 1,3-D+Pic in combination with pebulate and napropamide are equally effective against the weed Cyperus spp., the nematodes Tylenchorhynchus spp., Belonolaimus longicaudatus, and Meloidogyne spp., and the soilborne disease Fusarium oxysporum f.sp. lycopersici in fresh market tomato. Therefore, broadcast 1,3-D+Pic application can reduce personnel exposure and poisoning risks, without losing pest control efficacy.
