Pathogens such as Escherichia coli O157:H7 and Campylobacter spp. have been implicated in outbreaks of food poisoning in the UK and elsewhere. Domestic animals and wildlife are important reservoirs for both of these agents, and cross-contamination from faeces is believed to be responsible for many human outbreaks. Appropriate parameterisation of quantitative microbial-risk models requires representative data at all levels of the food chain. Our focus in this paper is on the early stages of the food chain-specifically, sampling issues which arise at the farm level. We estimated animal–pathogen prevalence from faecal-pat samples using a Bayesian method which reflected the uncertainties inherent in the animal-level prevalence estimates. (Note that prevalence here refers to the percentage of animals shedding the bacteria of interest). The method offers more flexibility than traditional, classical approaches: it allows the incorporation of prior belief, and permits the computation of a variety of distributional and numerical summaries, analogues of which often are not available through a classical framework. The Bayesian technique is illustrated with a number of examples reflecting the effects of a diversity of assumptions about the underlying processes. The technique appears to be both robust and flexible, and is useful when defecation rates in infected and uninfected groups are unequal, where population size is uncertain, and also where the microbiological-test sensitivity is imperfect. When a chemical reaction forms two molecular products, even if the state-resolved differential cross section (DCS) for each product is obtained individually, the coincident attributes of the coproducts are still lacking. We exploit a method that provides coincidence information by measuring the state-resolved, pair-correlated DCS. Exemplified by the reaction F + CD4 --> DF + CD3, a time-sliced ion velocity imaging technique was used to measure the velocity distribution of a state-selected CD3 product and to reveal the information of the coincident DF in a state-correlated manner. The correlation of different product state pairs shows a striking difference, which opens up a new way to unravel the complexity of a polyatomic reaction. 相似文献
Seventeen phenyl-fluorinated analogues of thidiazuron [N-phenyl-N'-(1,2,3-thiadiazol-5-yl)urea, TDZ] have been prepared and characterized. The effects of each fluorinated urea on growth and quality of kiwifruits (Actinidia deliciosa) were evaluated by comparison with untreated (control) and TDZ-treated fruits. The results obtained showed a clear dependence of the growth-promoting activity of these fluorinated ureas on the pattern and degree of fluorine substitution in the phenyl ring. The most effective for promoting fruit growth was N-(2,3,5,6-tetrafluorophenyl)-N'-(1',2',3'-thiadiazol-5'-yl)urea at 25 ppm (at harvest, treated fruits were 58% heavier than untreated ones) followed by N-(3,5-difluorophenyl)-N'-(1',2',3'-thiadiazol-5'-yl)urea at 10 ppm (50%). Comparatively, TDZ-treated fruits were 31% (10 ppm) and 38% (25 ppm) heavier than untreated ones. The results also indicate that the effects of the more active phenyl-fluorinated ureas on some standard quality parameters of fruits, for example, percent of fruit dry matter content, soluble solids contents, total titratable acids, shape, and internal structure, are similar to those of TDZ. Serum alkaline phosphatase (ALP), gamma-glutamyl transferase (GGT), 5'nucleotidase (5'NT), sorbitol dehydrogenase (SDH), and aspartate transaminase activities were measured in 10 clinically healthy foals, 10 yearlings, and 10 two-year-old Quarter Horses. Enzyme activities in foals at 0.5 to 3 days, 2 to 3 weeks, and 5 to 7 weeks of age were compared with enzyme activities from yearling and 2-year-old horses. Multivariate analyses of variance revealed significantly higher enzyme values in foals (P less than 0.002). This increase was mainly a result of higher ALP and GGT activities, with lesser effects due to higher SDH and 5'NT activities. Standard deviations for ALP and GGT were also larger in foals than in adult horses. The wide variation of ALP and GGT activities may limit their usefulness in the diagnosis of hepatic disease in foals. Standard deviations for serum AST, SDH, and 5'NT activities were smaller. These enzymes may be indicators of hepatobiliary disease in foals. The high serum enzyme activities in healthy foals may reflect a physiologic difference between foals and adult horses. Relative hepatic mass (as a percentage of body weight) and enzyme activity per gram of hepatic tissue are high in young animals, indicating that the high serum enzyme activities in foals are due partly to a high rate of enzyme production and release. 相似文献
Table olives constitute an important part of the Mediterranean diet and the diet of many non-olive-producing countries. The aim of this work was to determine the fiber, sugar, and organic acid contents in Spanish commercial presentations of table olives and characterize them by means of a multivariate analysis. The selection of variables was carried out on the basis of a canonical analysis and their classification, according to processing styles and cultivars, through a linear discriminant analysis. Values of dietary fiber in table olives ranged from 2 to 5 g/100 g edible portion (e.p.). Some stuffing materials (almond, hot red pepper, and hazelnut) or the addition of capers produced a significant increase in the total dietary fiber in green olives. Glucose, fructose, and mannitol were usually found in the ranges of 0-55, 0-70, and 0-107 mg/100 g e.p., respectively. Succinic acid was detected only in green and directly brined olives (0-40 mg/100 g e.p.), while lactic and acetic acids were used within the ranges of 0-681 and 5-492.8 mg/100 g e.p., respectively. A multivariate analysis showed that fiber, mannitol, and succinic, lactic, and acetic acids can be used to discriminate between processing styles (95.5% correct assignations) and cultivars (61.20%). The use of indicators in soil monitoring schemes to detect changes in soil quality is receiving increased attention, particularly
the application of soil biological methods. However, to date, the ability to compare information from different laboratories
applying soil microbiological techniques in broad-scale monitoring has rarely been taken into account. This study aimed to
assess the consistency and repeatability of two techniques that are being evaluated for use as microbiological indicators
of soil quality: multi-enzyme activity assay and multiple substrate-induced respiration (MSIR). Data were tested for intrinsic
(within-assay plate) variation, inter-laboratory repeatability (geometric mean regression and correlation coefficient) and
land-use discrimination (principal components analysis). Intrinsic variation was large for both assays suggesting that high
replicate numbers are required. Inter-laboratory repeatability showed diverging patterns for the enzyme assay and MSIR. Discrimination
of soils was significant for both techniques with relatively consistent patterns; however, combined laboratory discrimination
analyses for each technique showed inconsistent correspondence between the laboratories. These issues could be addressed through
the adoption of reliable analytical standards for biological methods along with adequate replication. However, until the former
is addressed, dispersed analyses are not currently advisable for monitoring schemes. 相似文献
An imaging report is a vital communication tool between a radiologist and clinician. In a field where in‐person communication may not be readily available, it is imperative that the report clearly relays pertinent clinical information in a timely manner. The purpose of this observational study was to describe and compare opinions and expectations of small animal general practitioners, veterinary specialists, and veterinary radiologists regarding the imaging report. Online surveys were distributed, and data were collected from 202 veterinary clinicians and 123 veterinary radiologists. The majority (89%) of clinicians were satisfied with their imaging reports and stated that they read the radiology report as soon as it was available (92%). Just less than half (48%) of clinicians indicated it was standard of care that a board‐certified veterinary radiologist read all imaging studies. Radiologists and clinicians agreed that a clinical history (98% and 94%, respectively) and clinical question (82% and 68%, respectively) were needed to generate a good radiology report. Fifty‐five percent to 70% of clinicians prefer red bulleted reports, which included incidental findings (96%); while radiologists slightly favored prose reporting (37–46%). Clinicians found it helpful when additional imaging (86%), medical (71%), and surgical recommendations (73%) were made. About one‐third of specialists who had been in practice for >11 years thought they were better able to interpret imaging for their own specialty than the radiologist. Clinicians voiced discontentment with reports that were not completed in a timely manner or did not give a prioritized differential list. Further studies are warranted to provide a more in‐depth evaluation of veterinary radiology reporting structure and style. 相似文献
1. Turkey eggs were incubated at 36.5, 37.5 and 38.5°C. The age of mortality, the incidence of malpositions and the incidence of morphological abnormalities were recorded from all unhatched eggs.
2. Eggs incubated at 36.5°C hatched later than eggs incubated at 37.5°C but did not differ in age of mortality or incidence of malpositions and abnormalities.
3. Eggs incubated at 38.5°C hatched significantly less well than eggs incubated at 37.5°C and showed significant differences in time of embryo mortality. Overheated embryos had a mortality peak between 15 and 20 d of incubation and an increased mortality after 24 d of incubation.

The effects of temperature and inoculum density of Fusarium oxysporum f. sp. ciceris race 5 on suppression of Fusarium wilt in chickpea (Cicer arietinum) cv. PV 61 by seed and soil treatments with rhizobacteria isolated from the chickpea rhizosphere were studied in a model system. Disease development over a range of temperatures (20, 25, and 30 degrees C) and inoculum densities (25 to 1,000 chlamydospores per gram of soil) was described by the Gompertz model. The Gompertz relative rate of disease progress and final amount of disease increased exponentially and monomolecularly, respectively, with increasing inoculum densities. Disease development was greater at 25 degrees C compared with 20 and 30 degrees C. At 20 and 30 degrees C, disease development was greater at 250 to 1,000 chlamydospores per gram of soil compared with 25 to 100 chlamydospores per gram of soil. At 25 degrees C, increasing inoculum densities of the pathogen did not influence disease. Nineteen Bacillus, Paenibacillus, Pseudomonas, and Stenotrophomonas spp. out of 23 bacterial isolates tested inhibited F. oxysporum f. sp. ciceris in vitro. Pseudomonas fluorescens RGAF 19 and RG 26, which did not inhibit the pathogen, showed the greatest Fusarium wilt suppression. Disease was suppressed only at 20 or 30 degrees C and at inoculum densities below 250 chlamydospores per gram of soil. Bacterial treatments increased the time to initial symptoms, reduced the Gompertz relative rate of disease progress, and reduced the overall amount of disease developed. 相似文献
Bats are hosts of diverse coronaviruses (CoVs) known to potentially cross the host–species barrier. For analysing coronavirus diversity in a bat species‐rich country, a total of 421 anal swabs/faecal samples from Costa Rican bats were screened for CoV RNA‐dependent RNA polymerase (RdRp) gene sequences by a pancoronavirus PCR. Six families, 24 genera and 41 species of bats were analysed. The detection rate for CoV was 1%. Individuals (n = 4) from four different species of frugivorous (Artibeus jamaicensis, Carollia perspicillata and Carollia castanea) and nectivorous (Glossophaga soricina) bats were positive for coronavirus‐derived nucleic acids. Analysis of 440 nt. RdRp sequences allocated all Costa Rican bat CoVs to the α‐CoV group. Several CoVs sequences clustered near previously described CoVs from the same species of bat, but were phylogenetically distant from the human CoV sequences identified to date, suggesting no recent spillover events. The Glossophaga soricina CoV sequence is sufficiently dissimilar (26% homology to the closest known bat CoVs) to represent a unique coronavirus not clustering near other CoVs found in the same bat species so far, implying an even higher CoV diversity than previously suspected. 相似文献
