Vaccination is the most cost effective control measure for Johne’s disease caused by Mycobacterium avium subspecies paratuberculosis (MAP) but currently available whole cell killed formulations have limited efficacy and are incompatible with the diagnosis of bovine tuberculosis by tuberculin skin test. We have evaluated the utility of a viral delivery regimen of non-replicative human Adenovirus 5 and Modified Vaccinia virus Ankara recombinant for early entry MAP specific antigens (HAV) to show protection against challenge in a calf model and extensively screened for differential immunological markers associated with protection. We have shown that HAV vaccination was well tolerated, could be detected using a differentiation of infected and vaccinated animals (DIVA) test, showed no cross-reactivity with tuberculin and provided a degree of protection against challenge evidenced by a lack of faecal shedding in vaccinated animals that persisted throughout the 7 month infection period. Calves given HAV vaccination had significant priming and boosting of MAP derived antigen (PPD-J) specific CD4+, CD8+ IFN-γ producing T-cell populations and, upon challenge, developed early specific Th17 related immune responses, enhanced IFN-γ responses and retained a high MAP killing capacity in blood. During later phases post MAP challenge, PPD-J antigen specific IFN-γ and Th17 responses in HAV vaccinated animals corresponded with improvements in peripheral bacteraemia. By contrast a lack of IFN-γ, induction of FoxP3+ T cells and increased IL-1β and IL-10 secretion were indicative of progressive infection in Sham vaccinated animals. We conclude that HAV vaccination shows excellent promise as a new tool for improving control of MAP infection in cattle.

Powdery mildew is a foliar disease of rubber trees. It is one of the most important leaf diseases in rubber plantations worldwide. The causal agent of this powdery mildew, previously known as Oidium heveae, has been identified under various names. To precisely identify the causal agents of this disease, 57 conidial samples were collected from 2013 to 2017 in Hainan, Yunnan and Guangdong provinces, the main rubber tree planting regions in China. Based on morphological observations using light and scanning electron microscopy, pathogenicity testing, and nrDNA ITS and 28S sequences, Erysiphe quercicola is suggested as the only causal agent of rubber tree powdery mildew in China.  相似文献

A major challenge in developing agroforestry approaches that utilize tree-leaf biomass for provision of N to crops is to ensure synchrony between the N released from decomposing prunings and N demand by crops. A study was conducted in the subhumid highlands of Kenya to assess the rate of decomposition and mineralization of soil-incorporated Calliandra calothyrsus Meissner (calliandra) and Leucaena leucocephala (Lam.) de Wit (leucaena) tree biomass and maize roots (Zea mays L.) both in an alley cropping and a sole cropping system. The amount of mineralized N peaked four weeks after planting (WAP) maize in all the treatments during both seasons of 1995. Cumulative mineralized N at week 20 ranged from 114 to 364 kg N ha−1 season−1, the absolute control treatment giving the lowest and the prunings-incorporated treatments giving the highest amounts in the two seasons. Total N uptake by maize, ranging from 42 to 157 kg ha−1 season−1, was lowest in the 'alley-cropped, prunings-removed' treatments, and highest in the 'non alley-cropped-prunings-incorporated' treatments. The apparent N recovery rate by maize was highest in the fertilizer applied treatments in the two seasons. Decomposition rate constants (kD) ranged from 0.07 to 0.21 week−1, and the rates among the different plant residues were as follows: leucaena < calliandra < maize roots. Nitrogen release rate constants (kN), ranging from 0.04 to 0.25 week−1, followed a similar pattern as the rate of decomposition with leucaena releasing the highest amount of N followed by calliandra and lastly by maize roots. Soil nutrient depletion as a result of continuous cultivation of soils without adequate addition of external inputs is a major challenge in the highlands of Kenya. An experiment was set up in Meru South District, Kenya in 2000 to investigate the effects of different soil-incorporated organic (manure, Tithonia diversifolia, Calliandra calothyrsus, Leucaena leucocephala) and mineral fertilizer inputs on maize yield, and soil chemical properties over seven seasons. On average, tithonia treatments (with or without half recommended rate of mineral fertilizer) gave the highest grain yield (5.5 and 5.4 Mg ha−1 respectively) while the control treatment gave the lowest yield (1.5 Mg ha−1). After 2 years of trial implementation, total soil carbon and nitrogen contents were improved with the application of organic residues, and manure in particular improved soil calcium content. Results of the economic analysis indicated that on average across the seven seasons, tithonia with half recommended rate of mineral fertilizer treatment recorded the highest net benefit (USD 787 ha−1) while the control recorded the lowest (USD 272 ha−1). However, returns to labor or benefit-cost ratios were in most cases not significantly improved when organic materials were used.  相似文献

Nitrogen fixation in biological soil crusts from southeast Utah,USA   总被引:20,自引:0,他引:20  
Biological soil crusts can be the dominant source of N for arid land ecosystems. We measured potential N fixation rates biweekly for 2 years, using three types of soil crusts: (1) crusts whose directly counted cells were >98% Microcoleus vaginatus (light crusts); (2) crusts dominated by M. vaginatus, but with 20% or more of the directly counted cells represented by Nostoc commune and Scytonema myochrous (dark crusts); and (3) the soil lichen Collema sp. At all observation times, Collema had higher nitrogenase activity (NA) than dark crusts, which had higher NA than light crusts, indicating that species composition is critical when estimating N inputs. In addition, all three types of crusts generally responded in a similar fashion to climate conditions. Without precipitation within a week of collection, no NA was recorded, regardless of other conditions being favorable. Low (<1°C) and high (>26°C) temperatures precluded NA, even if soils were moist. If rain or snow melt had occurred 3 or less days before collection, NA levels were highly correlated with daily average temperatures of the previous 3 days (r2=0.93 for Collema crusts; r2=0.86 for dark crusts and r2=0.83 for light crusts) for temperatures between 1°C and 26°C. If a precipitation event followed a long dry period, NA levels were lower than if collection followed a time when soils were wet for extended periods (e.g., winter). Using a combination of data from a recording weather datalogger, time-domain reflectometry, manual dry-down curves, and N fixation rates at different temperatures, annual N input from the different crust types was estimated. Annual N input from dark crusts found at relatively undisturbed sites was estimated at 9 kg ha-1 year-1. With 20% cover of the N-fixing soil lichen Collema, inputs are estimated at 13 kg ha-1 year-1. N input from light crusts, generally indicating soil surface disturbance, was estimated at 1.4 kg ha-1 year-1. The rates in light crusts are expected to be highly variable, as disturbance history will determine cyanobacterial biomass and therefore N fixation rates.  相似文献

OBJECTIVE: The purpose of the present study was to examine the prevalence of fast-food purchases for family meals and the associations with sociodemographic variables, dietary intake, home food environment, and weight status in adolescents and their parents. DESIGN: This study is a cross-sectional evaluation of parent interviews and adolescent surveys from Project EAT (Eating Among Teens). SUBJECTS: Subjects included 902 middle-school and high-school adolescents (53% female, 47% male) and their parents (89% female, 11% male). The adolescent population was ethnically diverse: 29% white, 24% black, 21% Asian American, 14% Hispanic and 12% other. RESULTS: Results showed that parents who reported purchasing fast food for family meals at least 3 times per week were significantly more likely than parents who reported purchasing fewer fast-food family meals to report the availability of soda pop and chips in the home. Adolescents in homes with fewer than 3 fast-food family meals per week were significantly more likely than adolescents in homes with more fast-food family meals to report having vegetables and milk served with meals at home. Fast-food purchases for family meals were positively associated with the intake of fast foods and salty snack foods for both parents and adolescents; and weight status among parents. Fast-food purchases for family meals were negatively associated with parental vegetable intake. CONCLUSIONS: Fast-food purchases may be helpful for busy families, but families need to be educated on the effects of fast food for family meals and how to choose healthier, convenient family meals.  相似文献

An attempt was made to evaluate gluten structural changes in refined and whole wheat pasta from hard white winter wheat to elucidate the impact of whole wheat components on the formation and structure of the gluten network in pasta. Attenuated total reflectance–FTIR spectroscopy was used to track gluten secondary structure through most of the major steps in pasta processing: raw material, mixing, drying, and cooking. Protein solubility, accessible thiols, and SDS‐PAGE data were also collected to provide additional information on the nature of protein interactions and network composition. Few secondary structural differences were observed between refined and whole wheat flours from hard white wheat. However, mixing induced a significant shift to β‐sheet structures in refined dough that was not equally matched by whole wheat dough. Drying under both high temperature, short time (HT) and low temperature, long time (LT) conditions resulted in a reversion to structural distributions similar to those for flour in both pastas. However, greater protein denaturation in HT samples was indicated by lower protein solubility also in the presence of denaturants and disulfide reducing agents. Cooking generated a substantial increase in β‐sheet structures for both pasta systems. This structure was greatest in refined and LT samples. Thiol accessibility data indicate the presence of a highly aggregated, compact gluten network in refined pasta, mostly driven by hydrophobic association. Conversely, the network in whole wheat pasta was more loosely associated and dependent on disulfide bonding, both of which fit well with the secondary structural data.  相似文献

Donors and aid agencies are now looking to strategies of pro‐poor economic growth to raise living standards in developing countries. I critically examine how particular conceptualisations of economy and economic development shape the broader context within which pro‐poor growth approaches are implemented by governments and development agencies. While pro‐poor growth may expand a national economy, surplus will be distributed in a community and among individuals according to culturally specific logics. To exemplify this point I discuss the community economy of Bomolo on the remote, rural island of Flores, Indonesia. The Ngadha people of Bomolo are identified by the Indonesian state, religious organisations and development agencies as lacking and in need of economic development. Yet in Bomolo, gambling, although illegal and antithetical to development, is fantastically popular, and large amounts of cash surplus are spent on this popular pastime. I argue for more attention to the issue of surplus – the outcome of pro‐poor growth. In particular, I propose that surplus distribution is a crucial yet underdeveloped component of the growth equation.  相似文献
