In a feeding trial at the Institute of Animal Nutrition of the Federal Research Institute of Animal Health (FLI) over 12 weeks with 20 cows of the German Holstein Breed the influence of different copper and zinc contents in the ration on their concentration in blood serum, liver and hair was tested. All animals received a diet based on maize- and grass silage ad libitum. The animals were divided in two groups with 10 cows each; group A received a concentrate according to their milk yield with a copper and zinc content as recommended (GfE 2001), whereas group B was offered a concentrate with roughly the double amount of copper and zinc. At the beginning and at the end of the trial a sample of blood, pigmented hair and a liver bioptate was taken from all animals to evaluate the incorporation of copper and zinc in these tissues. In serum and pigmented hair the copper concentrations did not differ between the two groups [13.4 for Group A and 12.5 micromol/L for Group B in serum respectively 6.8 (Group A) and 7.4 mg/kg DM (Group B) in pigmented hair]. Only the copper concentration in the liver was influenced by the different feeding. The higher copper content for group B resulted in a significantly higher copper concentration in the liver (506 mg/kg DM compared to 383 mg/kg DM). The liver is the best indicator organ for a sufficient copper supply. An increase in the zinc content in the ration resulted neither in higher zinc concentrations in serum (15.1 in Group B in comparison to 13.4 micromol/L for Group A) nor in higher zinc concentrations in liver (140 for Group B and 112 mg/kg DM for Group A) and pigmented hair (130 in Group A and 123 mg/kg DM in Group B). There is a significant correlation between copper intake and copper concentration in the liver (r = 0.46), whereas the correlation between zinc intake and zinc concentration in the liver is only tendencially (r = 0.23). The three tested samples serum, liver and cow hair are not qualified to reflect exactly a sufficient zinc supply.  相似文献

In 360 samples of colostrum and 36 samples of blood of warmblood mares, the concentration of immunoglobulin G (IgG) was evaluated in the post partal period with an ELISA and the results were compared to values obtained with 2 field methods--refractometry and colostrometry. A significant correlation (p < 0.0001) was determined between ELISA and colostrometry (r = +0.88) and between ELISA and refractometry (r = +0.93). So both field-methods seem suitable for evaluation of the colostral IgG-concentration in mares. Further the kinetic of the IgG concentration in colostrum, the volume of colostrum and the total amount of IgG was measured in the 12 hours post partum (p.p.) in each half udder of 36 mares of different parity. Immediately p.p. primiparous mares have a greater mean concentration of IgG (68 mg/ml) than multiparous mares (51 mg/ml). However, multiparous mares have a mean colostral volume of 1020 ml whereas, in primiparous mares, a mean volume of 527 ml was determined within the first three hours p.p. As a result of this the total amount of IgG was lower in primiparous (31.5 g) than in multiparous mares (48.5 g). A significant decrease of IgG concentration was measured in multiparous mares in the 1.5 hours following partum versus 3 hours in primiparous mares. The mean IgG concentration in the blood serum of the 36 mares immediately p.p. was 13.4 +/- 3.6 mg/ml. No significant correlation was observed between values of IgG concentration in the blood and in the colostrum of the mares.  相似文献

Canine idiopathic pulmonary fibrosis is a chronic, progressive interstitial lung disease particularly prevalent in West Highland White Terriers. In the present prospective pilot study, we evaluated the feasibility of modified VetMousetrap? device in high resolution CT to detect idiopathic pulmonary fibrosis in West Highland White Terriers. Twelve awake West Highland White Terriers with canine idiopathic pulmonary fibrosis and 24 clinically healthy West Highland White Terriers were scanned using a helical dual slice scanner utilizing VetMousetrap? device without or with minimal chemical restraint with butorphanol. Three evaluators blindly assessed the images for image quality and the presence of canine idiopathic pulmonary fibrosis related imaging findings such as ground glass opacity and reticular opacities. Additionally, the attenuation of the lung was quantified with ImageJ software using histogram analysis of density over the lung fields. Computed tomography was successfully completed and motion artifact ranked in statistical analysis barely noticeable to mild in all dogs. The agreement between imaging findings and clinical status was very good with overall κ value 0.91 and percentage of agreement of 94%. There was also very good intraobserver (κrange = 0.79‐0.91) and interobserver agreement (κ = 0.94). Moderate to severe ground glass opacity was present in all affected dogs. In the ImageJ analysis, a significant difference in lung attenuation between the study groups was observed. We conclude that modified VetMousetrap? device is applicable in diagnosing canine idiopathic pulmonary fibrosis in awake West Highland White Terriers avoiding anesthetic risk in these often severely hypoxic patients.  相似文献

OBJECTIVE: To evaluate the feasibility of ultrasound guided vascular access in pigs by comparing central venous and arterial cannulation techniques. ANIMALS: Twenty-two healthy female Pietrain pigs, 14-18 weeks old and weighing 51.1 +/- 4.3 kg (mean +/- SD). STUDY DESIGN: Comparative animal trial. MATERIALS AND METHODS: After induction of general anaesthesia, cannulation of the external jugular vein and internal carotid artery was attempted using real-time ultrasound guidance. The quality of the ultrasound picture was assessed on an analogue scale from 1 (excellent) to 5 (insufficient). Vessel size, cannulation success rate, number of puncture attempts and time from first puncture attempt until insertion of the Seldinger wire were recorded. RESULTS: Cannulation was successful in all but one animal in which a cut-down technique was performed. The arteries were significantly smaller than the veins (p < 0.001) resulting in a significantly prolonged cannulation time (p = 0.032) for insertion of arterial catheters without differences in success rate. In 89% of attempted cannulations, the Seldinger wire was inserted within 5 minutes. CONCLUSIONS AND CLINICAL RELEVANCE: In anaesthetized pigs undergoing instrumentation for biomedical research, ultrasound-guided vascular access is a simple and rapid alternative to surgical cut-down. In veterinary anaesthesia, the technique might be useful in sedated or anesthetized pigs in which arterial or central venous access is required.  相似文献

In this study, we determined whether deficient dietary amino acid (AA) concentrations influence the precaecal (pc) AA digestibility when determined using the regression approach. We mixed two basal diets. Basal diet 1 was deficient in essential AAs, whereas adequate AA concentrations were ensured in basal diet 2 by adding free AAs. Rapeseed cake and full‐fat soya beans as test ingredients were included in the basal diets at levels of 100 and 200, and 150 and 300 g/kg, respectively, at the expense of maize starch. Each diet was tested with six replicates of 10 broiler chickens each. The feed intake of the chickens that were fed diets based on basal diet 2 was similar, whereas the feed intake of the chickens that were fed diets based on basal diet 1 differed considerably. The numerical differences in pc AA digestibility determined with basal diet 1 or 2 ranged from 2.6 percentage points to 20.8 percentage points in rapeseed cake and from 0.5 percentage points to 15.2 percentage points in soya beans. Across all measured AAs, the average differences were 10.1 percentage points and 5.4 percentage points in rapeseed cake and soya beans, respectively. The differences in the estimated pc AA digestibility between the basal diets were probably caused by different basal endogenous AA losses in the digesta between treatments as a consequence of different feed intake. Adequate AA concentrations and test ingredient levels that are specifically adjusted to avoid a negative effect on feed intake are recommended for future studies.  相似文献

During recent decades, forest ecosystems have been exposed to high levels of atmospheric pollution, and it has been argued that this affects the composition and activity of decomposer communities and, subsequently, ecosystem functioning. To investigate the effects of atmospheric pollution on protozoa and microflora, a new experimental design was used. Undisturbed soil columns, originating from six coniferous forests across Europe and representing different stages of soil acidification, were transferred to two Scots pine forests (Fontainebleau and Wekerom) with different levels of N and S deposition (NH4 +-N=4.90 and 42.50?kg ha–1 year–1; SO4 S=10.90 and 30.40?kg ha–1 year–1, respectively). The number of protozoa, microbial biomass C and microbial activity were estimated in the organic layer (Of) of the transferred soils at the two host sites after 21 months of incubation. The experiment aimed at answering two questions: (1) Do changes in environmental conditions, studied by transferring soils from one site to another, affect protozoa and microbial communities and, if so, (2) how important are changes in both N and S deposition in explaining the effects of soil transfer on protozoa and microbial communities? The interaction between protozoa and microbial communities was addressed with regard to these changes in environmental conditions. No effect of enhanced N or S deposition on protozoan numbers and microbial biomass C, basal respiration and caloric quotient was revealed. Reciprocal transfer of various soil columns resulted in lower abundance and activity of protozoa and microbes. This reduction could not be explained by differences in N and S deposition, but by differences in microclimate and adaptation. In some cases, protozoa correlated with pH, C/N ratio, P and S content and leached mineral N.  相似文献

Illegal, unregulated and unreported (IUU) fishing is a major problem in many of the world's fisheries. The stocks most severely impacted centre on those characterised by high economic value, such as abalone, as well as long lived and slow growing species such as Patagonian toothfish (Dissostichus eleginoides). Effective management of these stocks as well as assessment of the impacts of IUU fishing on the resources is impeded by the technical difficulties associated with determining the magnitude of the IUU catches. The South African abalone Haliotis midae fishery rates as an extreme example of extraordinarily high levels of illegal and unreported (IU) catch. To assess the level and trends in IU catches, we used a combination of approaches that included collation of confiscation records from law enforcement, development of a novel index (the confiscations per unit policing effort—CPUPE), estimation of illegal catches using a spatial and age-structured assessment model, and cross-checking of model outputs through comparison with trade data on abalone imports in destination countries. The model-predicted 2008 IU estimate was 860 tonnes, more than 10 times the total allowable catch (TAC) for that year, and implied that, on average, 14% of all IU catches are confiscated. Associated management responses included the listing of H. midae on Appendix III of CITES (the Convention on International Trade in Endangered Species of Wild Fauna and Flora), and a temporary closure of the commercial fishery. We summarise both technical and management lessons to be learnt from this integrated approach to assess and verify the magnitude of IU fishing.  相似文献

Yeast is a potential alternative to fish meal in diets for farmed fish, yet replacing more than 50 % of fish meal results in reduced fish growth. In a 4-week experiment, 15 rainbow trout (Oncorhynchus mykiss) were cannulated and fed three diets each week: 30 % fish meal as a control (FM); 60 % replacement of fish meal protein, on a digestible basis, with Saccharomyces cerevisiae (SC); and 60 % replacement with Wickerhamomyces anomalus and S. cerevisiae mix (WA). Blood was collected at 0, 3, 6, 12 and 24 h after feeding. In the final week, fish were exposed to a 1-min netting stressor to evaluate possible diet–stress interactions. Significant increases in pH, TCO2, HCO3 and base excess were found after fish were fed the SC and WA diets compared with FM, which elevated blood alkaline tides. Yeast ingredients had lower buffering capacity and ash content than fish meal, which explained the increase in alkaline tides. In addition, fish fed the WA diet had significantly reduced erythrocyte area and fish fed SC and WA diets had increased mean corpuscular haemoglobin levels, indicating haemolytic anaemia. Higher levels of nucleic acid in yeast-based diets and potentially higher production of reactive oxygen species were suspected of damaging haemoglobin, which require replacement by smaller immature erythrocytes. Acute stress caused the expected rise in cortisol and glucose levels, but no interaction with diet was found. These results show that replacing 60 % of fish meal protein with yeasts can induce haemolytic anaemia in rainbow trout, which may limit yeast inclusion in diets for farmed fish.  相似文献
