Recently, transgenic potato plants were created showing underexpression of the 20R isoform of the 14-3-3 protein. The transgenic plants grown in tissue culture showed a significant increase in nitrate reductase activity and a decrease in nitrate level. The transgenic line with the lowest 14-3-3 quantity was field-trialed (1997-2000) and analyzed. The reduction in the 14-3-3 protein level consistently resulted in a starch content increase and in an increase in the ratio of soluble sugars to starch in the tubers, although the latter was only barely visible. The determination of amino acid composition in the tubers showed a significant increase in methionine, proline, and arginine content and a slight but consistent increase in hydrophobic amino acid and lysine content in the cells of the transgenic potato plants. We also observed an increase in the crude protein content, from 19 to 22.1% of the control value in consecutive years. It is proposed that all of these changes might have resulted from the downregulation of nitrate reductase and sucrose phosphate synthase activities by 14-3-3, although other potential mechanisms cannot be excluded (e.g., an increase in enzyme protein level). 14-3-3-repressed transgenic plants showed a significant increase in calcium content in their tubers. It is thus proposed that a function of the isolated 14-3-3 isoform is in the control of amino acid synthesis and calcium metabolism. However, the mechanism of this control is as yet unknown.  相似文献

In the conditions of long-term experiment carried out in the experimental field of Warsaw Agricultural University - SGGW at _ yczyn, ion concentration in the soil solution were examined as well as the relationships shaped between ions in the soil solution due to long-term diversified mineral and organic fertilization. Soil solution was obtained by the vacuum method and the concentration of particular ions in the soil solution was determined by the ICP method. Mineral and organic fertilization and liming significantly affected chemical composition of the soil solution and the relationships between particular ions in the soil solution. The lowest concentration of ions in the soil solution was observed in the control plots (not fertilized). Mineral and organic fertilization as well as liming caused the increase of ion concentration in the soil solution. In the plots with mineral fertilization alone K + and NH + 4 ions predominated in the cation structure in the soil solution. In the case of liming the share of Ca 2+ ions increased. Independently from the system of fertilization used, CI m and HCO m 3 ions predominated in the anion structure. The application of organic fertilization affected to the highest degree the concentration of aluminium ion in the soil solution significantly limiting the share of this ion in the cation structure.  相似文献

Cytoskeletal dynamics play a crucial role in pathogen recognition and cell defence during the initial interactions between an invader and plant host. The aim of the work reported here was to characterize how Heterobasidion annosum s.s., Heterobasidion parviporum, and Heterobasidion abietinum affect the microtubules and microfilaments of Pinus sylvestris root cells 12‐, 24‐, 48‐, and 96‐h post‐inoculation. Inoculation of P. sylvestris with H. parviporum or H. abietinum, which have a lower specificity for P. sylvestris than H. annosum s.s, resulted in greater reorganization of host microtubules during the early stages of interaction than inoculation with the more specific H. annosum s.s. In some infected cells, spots of actin aggregates were observed. Disruption of cytoskeletal components by the application of specific cytoskeletal inhibitors facilitated the entry of the H. parviporum and H. abietinum into roots. These results suggest that the P. sylvestris cytoskeleton plays a role in the host response in the initial stages of the host–pathogen interaction.  相似文献

Juvenile European pikeperch, Sander lucioperca, were fed commercial feed (group C) or experimental feed supplemented with NuPro® nucleotide‐rich Saccharomyces cerevisiae yeast protein (extract obtained through a cell wall removal process) in doses of 20, 40 or 60 g kg?1 feed (groups N2, N4, N6) for 8 weeks. Growth, non‐specific immunity parameters, histological structure of the liver and intestine, proximate whole‐body composition and blood biochemical parameters were assayed. It was noted that brewer’s yeast extract has immunomodulatory proprieties. NuPro® in doses of 40 and 60 g kg?1 feed strongly stimulated non‐specific (innate) cellular and humoral immunity in pikeperch. The experimental feed did not have a significant impact on pikeperch growth (P > 0.05). The proximate composition of the fish bodies and the hepatosomatic and viscerosomatic (VSI) indices were also not affected, which indicated that the tested diets had no negative impact on the metabolism or deposition of nutrients in fish tissues. The lower levels of transaminases AST and ALT, which were noted in the groups with the two highest doses of NuPro® (P < 0.05), might indicate improved liver function. It was also demonstrated that the brewer’s yeast extract stimulates the absorption activity of intestinal epithelial cells.  相似文献

This paper analyzes changes in the length of coat on selected body areas in horses and ponies kept under different husbandry (stable) conditions during the winter‐spring period. The study included 12 Ma?polski geldings and 12 geldings of Felin ponies aged 10–15 years. Horses were kept in two stables (six horses and six ponies in each stable). The type of performance, husbandry conditions and feeding of the studied animals were comparable. As of December 1, samples of hair coat from the scapula, sternum, back and abdomen areas of both body sides were collected seven times. The lengths of 20 randomly selected hair fibers were measured. Daily measurements of air temperature in the stables were also taken. An analysis of variance (ANOVA) was performed using the following factors: the body part from where the coat was sampled, the subsequent examination and the stable as well as the interaction between these factors. The significance of differences between means was determined with a t‐Tukey test. The relations between air temperature in the stable and hair length were calculated using Pearson's correlation. It was found that air temperature in the stable impacts the length of winter coat in horses and ponies. The effect of this factor is more pronounced in ponies; as in the stables with lower temperatures it produces a longer hair coat which is more evenly distributed over the body in comparison with horses. Keeping horses and ponies in stables with a low air temperature accelerates coat shedding by approximately 25 days. Coat shedding begins from the scapula area.  相似文献

Horses intended for leisure riding do not undergo any selection and most often retired sports horses or defective horses are chosen, as a low selling price determines their purchase by a leisure riding center. Unfortunately, horses bought at low prices usually have low utility value, are difficult to handle, require a special or individual approach and do not provide satisfaction in riding. However, neither modern horse breeding nor scientific research address the need to breed horses for leisure activities. There is no clear definition of a model leisure horse and criteria or information for its selection are not readily available in scientific publications. A wide spectrum of research methods may be used to evaluate various performance traits in horses intended for leisure activities. The fact that the population of recreational horses and their riders outnumber sporting horses should attract the special attention of scientific research. Their utility traits need to be determined with modern technology and methods in the same way they are for sporting horses. Such a system of evaluation would be very helpful for riders.  相似文献

Sulfachlorpyrazine (SCP) is currently used to treat coccidian infections in turkeys; however, there is no information available about the withdrawal period necessary for the turkey to be safe for human consumption. A high performance liquid chromatography method with ultraviolet‐visible light detection was adapted and validated for the determination of SCP in turkey tissues. The procedure is based on isolation of the (SCP sodium) compound from edible turkey tissues (muscles, liver, kidneys, and fat with skin) with satisfactory recovery (72.80 ± 1.40) and specificity. The residue depletion of SCP in turkeys was conducted after a dose of 50 mg/kg body weight/day had been administrated orally for 3 days. After treatment has been discontinued residue concentrations were detected in tissues on the 7th day. The highest SCP concentrations were measured in muscles. Based on the results presented in this study, it could be assumed that a withdrawal period of 21 days, before medicated turkeys could be slaughtered, would be sufficient to ensure consumer safety.  相似文献

Effect of the lectin PHA on the feeding behavior of the grain aphid   总被引:1,自引:0,他引:1  
The electrical penetration graph (EPG) method was used to quantify the effect of the lectin PHA (phytohemagglutinin) on the feeding behavior of the grain aphid Sitobion avenae F (Hemiptera: Aphididae). Addition of PHA to an artificial diet affected aphid probing behavior. In general, increasing concentrations of PHA in sucrose-agarose gels significantly reduced the number of aphid probes and extended their duration. Aphids feeding on the gels with lectin (without concentration 50 μg · cm−3) had a prolonged activity (EPG pattern C) that was corresponded to prolonged penetration of the epidermis and mesophyll for aphids feeding on plants. Such insects also showed a significant reduction in salivation and passive ingestion from gels and a reduction in the total number of gel penetrations. At concentrations ≤250 μg · cm−3, PHA reduced feeding behavior on gels, and the EPG data were interpreted to correspond with reduced salivation into phloem sieve elements (EPG pattern E1) and reduced ingestion of phloem sap (EPG pattern E2). PHA also delayed the onset of the first E1 and E2 patterns. At higher concentrations (≥500 μg · cm−3), PHA completely stopped salivation and passive ingestion from gels. Similarly, higher concentration of PHA strongly reduced activity on gels that corresponded to activity in xylem (EPG pattern G); high PHA delayed the onset of the first G pattern and reduced total time of pattern G activity.  相似文献
