The fine structure is described of the merogonic stages and gametocytes of a Plasmodium tropiduri Arag?o et Neiva, 1909-like parasite infecting the teiid lizard Kentropyx calcarata Spix from North Brazil. The trophozoites are bordered by two membranes, and with growth a pellicle is formed by the addition of an inner, thick double layer and fragmented membrane. The same type of inner membrane occurs in the pellicle of the merozoites differentiating from the meronts. Merozoites contained a large electron-dense body, sometimes seen to be embraced by a tubular mitochondrion with a dense matrix. Micro- and macrogametocytes are bounded by a double membrane, closely apposed by the detached wall of the parasitophorous vacuole. Both contain osmiophilic bodies. The microgametocyte contains an electron-dense aggregate, and the macrogametocyte has a large mitochondrion and a complex of tubuli and cisternae. How can neural activity propagate through cortical networks built with weak, stochastic synapses? We find precise repetitions of spontaneous patterns of synaptic inputs in neocortical neurons in vivo and in vitro. These patterns repeat after minutes, maintaining millisecond accuracy. Calcium imaging of slices reveals reactivation of sequences of cells during the occurrence of repeated intracellular synaptic patterns. The spontaneous activity drifts with time, engaging different cells. Sequences of active neurons have distinct spatial structures and are repeated in the same order over tens of seconds, revealing modular temporal dynamics. Coating iris bulbs with a preparation ofTrichoderma harzianum was highly effective under greenhouse conditions in reducing incidence of diseases caused byRhizoctonia solani andSclerotium rolfsii. In field experiments with irises for bulb production, the incidence ofR. solani in plants and bulbs was effectively reduced (up to 93%), and the yield increased (by 35–41%), by applyingT. harzianum either as a bulb coating or broadcast application (biological treatment), treating soil with pentachloronitrobenzene (PCNB; quintozene) (chemical treatment), or solarizing the soil by mulching it with transparent polyethylene sheets (physical treatment) prior to planting. Fusarium wilt is an economically important disease of tomatoes, caused by the soil-born fungus Fusarium oxysporum f. sp. lycopersici. There are three host-specific races of this pathogen. The dominant tomato gene I-2 confers resistance to race 2. The I-2 fusarium resistance gene was mapped genetically to chromosome 11 of tomato, between the RFLP markers TG105 and TG36, 0.4 centiMorgan (cM) from TG105. A mean value of 43 kb for each cM was assigned in the vicinity of I-2. We have generated new RFLP markers in the region by chromosome walking from TG105 towards I-2 on lambda clones, and by subcloning a 350 kb long YAC clone (YAC 8) that contains TG105. These RFLP markers were mapped physically on YAC 8 by PFGE. The location of I-2 relative to these markers was genetically estimated using a recombinant inbred (RI) segregating population. The order of the markers according to the RI population is inconsistent with their order on the physical map. A cDNA clone, D14, that was isolated by YAC 8, turned out to be 53% similar to xanthine dehydrogenase from mammals and flies. Antibodies raised against a part of the protein encoded by D14 recognize cross reacting material of MW 80 kD, that is highly enriched in nodules of legumes, and seems to be induced by various environmental and pathogenic stress conditions. 相似文献

Glycoalkaloids are secondary metabolites characterised by an undesirable taste, and known to be toxic when consumed in large
quantities. Their content in potato tubers should therefore be selected against and DNA markers can significantly facilitate
such a process. This study was designed to describe the genetic control of the total content of glycoalkaloids (TGA) in the
potato tuber and to enhance the development of DNA markers. A segregating population was constructed from tetraploid potatoclones,
which were characterised by divergent TGA-content. The population had a high-TGA parent, LT7, and a low-TGA parent, NT8. We
used 342 Inter-Simple Sequence Repeat (ISSR) single primers or two such primer combinations and 36 Cleaved Amplified Polymorphic
Sequence (CAPS)primer pairs, digested with various restriction enzymes, to amplify random PCR products. Parental clones of
both populations were used to identify polymorphic PCR products for further linkage analyses with TGA-content. In these analyses,
one single ISSR marker was found to be significantly associated with TGA-content. A multiple regression analysis was also
carried out using a ‘stepwise’ procedure. In this regression analysis TGA-content was the dependent variable whereas the polymorphic
PCR products and all possible two-way interactions among them were the independent variables. The resulting best model consisted
of an interaction between two loci in addition to a single locus effect. This interaction suggests that the expression of
TGA-content is partially modulated by two interacting loci.

The fungus Aspergillus
tubingensis (strain OY907) was isolated from the Mediterranean marine sponge Ircinia variabilis. Extracellular extracts produced by this strain were found to inhibit the growth of several fungi. Among the secreted extract components, a novel anhydride metabolite, tubingenoic anhydride A (1) as well as the known 2-carboxymethyl-3-hexylmaleic acid anhydride, asperic acid, and campyrone A and C were purified and their structure elucidated. Compound 1 and 2-carboxymethyl-3-hexylmaleic acid anhydride inhibited Neurospora crassa growth (MIC = 330 and 207 μM, respectively) and affected hyphal morphology. We produced a N. crassa mutant exhibiting tolerance to 1 and found that a yet-uncharacterized gene, designated mas-1, whose product is a cytosolic protein, confers sensitivity to this compound. The ∆mas-1 strain showed increased tolerance to sublethal concentrations of the chitin synthase inhibitor polyoxin D, when compared to the wild type. In addition, the expression of chitin synthase genes was highly elevated in the ∆mas-1 strain, suggesting the gene product is involved in cell wall biosynthesis and the novel anhydride interferes with its function. 相似文献

Urban air pollution and smoke from fires have been modeled to reduce cloud formation by absorbing sunlight, thereby cooling the surface and heating the atmosphere. Satellite data over the Amazon region during the biomass burning season showed that scattered cumulus cloud cover was reduced from 38%in clean conditions to 0%for heavy smoke (optical depth of 1.3). This response to the smoke radiative effect reverses the regional smoke instantaneous forcing of climate from -28 watts per square meter in cloud-free conditions to +8 watts per square meter once the reduction of cloud cover is accounted for. 相似文献

In field studies, mean stands of crops of snapbean, field corn, pea, soybean, and squash in New York were somewhat greater from seeds treated with conidia of Trichoderma harzianum applied in a Methocel slurry than from non-treated controls; however, stands were generally poorer than those from captan-treated seed. The biocontrol agent and captan had little effect on kidney bean and sweet corn since these crops are only slightly susceptible to seedling disease caused by Pythium spp. which were present in the field. In Colorado fields also infested with Pythium spp., stands of sugar beet from T. harzianum-treated seed (Pelgel slurry) tended to be greater than those from non-treated seed and equal to those from maneb-treated seed, but differences were not significant. In Colorado, Rhizoctonia solani, cause of root rot, was also present in the soil. A combined analysis of variance of treatments across 2 years showed that a preplanting-incorporated in-row application of T. harzianum in a wheat-bran carrier or a maneb seed treatment slightly, but significantly, reduced the severity of Rhizoctonia root rot in sugar beet as compared with the non-treated control. Seed treatments with T. harzianum had little or no effect. In plots where the agent was applied to the soil, numbers of Trichoderma propagules increased 600-fold by harvest; where maneb seed treatment was used, they increased only fivefold. Densities of the agent remained the same or decreased in plots where Trichoderma was used as a seed treatment. 相似文献
