Effects of remifentanil on monophasic action potential and transmural dispersion of repolarization in rabbit myocardium

AIM: To study the effect of remifentanil on monophasic action potential and transmural dispersion of repolarization (TDR) in the 3-layer myocardium of isolated rabbit hearts. METHODS: Adult rabbits (n=18, 2.0 ~ 2.5 kg) were used to isolate the hearts for preparing Langendorff perfusion model. The hearts were randomly divided into 3 groups after perfusion with K-H solution for 15 min: the perfusion in control group (C group) continued for 60 min; the hearts in remifentanil group (R group) were perfused with 12 μg/L remifentanil K-H solution for 60 min; the hearts in remifentanil+aminophylline group (RA group) were given 60-min perfusion of 12 μg/L K-H remifentanil+30 mg/L aminophylline. The HR and 3 layers of myocardial monophasic action potential (MAP) in the left ventricular anterior wall were recorded at time points after balanced infusion for 15 min (T₀), and continued perfusion for 15 min (T₁), 30 min (T₂) and 60 min (T₃). The monophasic action potential duration of repolarization at 90% (MAPD₉₀) and the transmural dispersion of repolarization (TDR) were calculated. The early afterdepolarization, delay afterdepolarization and arrhythmia were also observed. RESULTS: In R group, slower HR and prolonger MAPD₉₀ and TDR at T₁~T₃ were observed as compared with those at T₀ (P<0.05). R group showed slower HR and longer MAPD₉₀ and TDR than C group and RA group (P<0.05). CONCLUSION: Remifentanil slows the HR, extends the MAPD₉₀ and increases the TDR, thus being prone to induce reentry. Aminophylline makes HR faster and MAPD₉₀ shorter, thereby reducing the TDR.     

鱼肠道弧菌免疫检测方法的研究

采用灭活的鱼肠道弧菌作为抗原,制备兔抗血清,建立了鱼肠道弧菌的间接荧光抗体检测技术(indirect im-munofluorescence assay test,IFAT)、Western-blotting免疫印迹检测技术。IFAT结果显示阳性信号覆盖整个菌体,明亮,清晰,荧光信号呈长弧形且两端钝圆,与鱼肠道弧菌形状一致。Western-blotting结果显示共10条蛋白带发生免疫反应,其中6条蛋白带结果较清晰,显示出较强的特异性免疫反应,阴性对照组无条带。IFAT和Western-blotting结果说明所建立检测方法能够准确、快速的检测出鱼肠道弧菌。     

大白菜游离小孢子培养技术高效体系的研究

以大白菜吉红82、#534DH 群体及#438 群体为试材，研究了利用细胞破碎仪机械提取小孢子与人工挤压提取小孢子对大白菜游离小孢子活力及提取量的影响。结果表明：机械提取替代人工挤压可在短时间内同时提取多份样品，并可快速调整小孢子的培养浓度。收集40 个适期花蕾于5 mL 离心管内，3 000 r·min-1 破碎10 s，将提取的小孢子定容于25 mL NLN-13 培养基，可确保小孢子浓度在适宜培养范围内，并成功诱导胚胎发生。     

适用于转录组测序的大白菜小孢子总RNA提取方法筛选

为了找到适用于转录组测序要求的大白菜小孢子总RNA 提取方法，以出胚率较高的吉红82 为试材，以热激诱导处理后的小孢子为研究对象，比较分析了TRIzol 法、改良CTAB 法及超纯试剂盒法3 种方法的RNA 提取效果。结果表明：TRIzol法（磨样）和改良CTAB 法提取的RNA 浓度较高，但完整性较差；超纯RNA 提取试剂盒法提取的RNA 质量最高，且完整性好，能够满足转录组测序的要求。     

Ultrastructural impairment of islet microvascular endothelial cells in STZ-induced type 1 diabetic mice

AIM: To investigate the ultrastructural changes of islet microvascular endothelial cells in STZ-induced type 1 diabetic mice. METHODS: BALB/c mice were randomly divided into diabetic group and control group. The expression of insulin and platelet-endothelial cell adhesion molecule-1 (CD31) in islet microvessels was detected by immunohistochemical staining. The ultrastructural changes of islet β cells and islet microvessels were observed under transmission electron microscope. RESULTS: Compared with control group, the number of islet β cells, ratio of β cells/α cells, average number of secretory granules in β cells and insulin expression area per islet in diabetic group were significantly decreased (P<0.01). Besides, diabetic group had fewer microvessels with lower expression of CD31 (P<0.01). Mitochondria in islet microvascular endothelial cells and pericytes in diabetic group were swelling. The basement membrane of islet microvessels became thicker in diabetic group (P<0.01). CONCLUSION: Islet microvascular endothelial cells were impaired in type 1 diabetic mice.     

不同单酶制剂混合检测对比研究

试验以木聚糖酶、纤维素酶、淀粉酶和酸性蛋白酶为对象,首先研究了酸性蛋白酶分别和其他3种单酶进行混合后酶活测定值变化情况,结果表明,酸性蛋白酶对其他3种单酶的检测结果没有影响;试验第二部分研究了4种单酶整体混合在一起后各单酶测定值的变化情况,结果表明,木聚糖酶测定值出现很大偏高,其他3种单酶测定结果未出现显著偏差;进行进一步论证试验表明,淀粉酶可能是使木聚糖酶测定结果偏高的原因。     

不同灌溉方式下冬小麦穗部性状与产量关系的试验研究

采用田间试验,以豫麦69为试验材料,研究了不同水分处理下常规灌溉和一体化垄作沟灌的冬小麦产量与穗部性状的相互关系。结果表明,同常规灌溉方式相比,一体化垄作沟灌方式下,冬小麦的穗粒数、籽粒质量及产量分别增加了5.5356%、7.5489%、7.7454%,但穗数减少了0.4302%。常规灌溉和一体化垄作沟灌方式下,产量与穗数、穗粒数以及籽粒质量均正相关,但一体化垄作沟灌方式下的相关系数较常规灌溉方式大。一体化垄作灌溉有利于改善农田小气候、发挥作物的边行优势和提高小麦产量。     

去花对菊芋数量学性状的影响

以白皮菊芋为试材,通过对现蕾期菊芋去除1/4花、1/3花、1/2花、全部去除和不去花等5个处理,测定叶片、小花、分枝和块茎数量等相关指标,探讨了去花对菊芋数量学性状的影响规律,为提高菊芋块茎生物产量提供理论依据。结果表明:去花1/3、1/2和全株去花提高了单株菊芋块茎数量,降低单株块茎质量,1/4去花对菊芋单株块茎数量和单个块茎质量影响不显著;去花处理降低了二级分枝和一级分枝的数量;去花处理增加了单株叶片的数量和单片叶片的质量;随着去花数量的增加,单株小花数量呈现"降-升-降"的变化规律,单个小花质量呈现"降-升"的变化趋势。     

土壤压砂保水处理含水率对西瓜生产潜力的影响

以西瓜为试材,针对压砂保墒问题,选择压砂时期和灌溉定额作为研究因素,采用对比试验方法研究了压砂保墒效果及其种植西瓜的生产潜力。结果表明:压砂覆盖能够有效保持土壤水分,上年雨季前压砂与雨季后压砂相比较,播种初期前者处理土壤质量含水率可达10.8%以上,而后者仅为8.9%;补水条件下前者比后者处理西瓜纵径大2.8cm,西瓜横径大3.1cm,未补水条件下前者比后者处理西瓜纵径大3.9cm,西瓜横径大2.7cm;补水条件下,前者处理西瓜产量为12 301.0kg·hm~(-2),后者处理西瓜产量为7 549.1kg·hm~(-2),未补水条件下,前者处理西瓜产量为6 723.3kg·hm~(~(-2)),后者处理西瓜产量为3 829.5kg·hm~(-2),可见,压砂保水能够明显提高西瓜生产潜力。     

The Application of Pre-splitting Blasting to Excavation on High Slope in Soft Rock Section

Based on analyzing about blasting mode of slope in the construction of road, the paper shows that pre splitting blasting is mostly suitable in the section of severely weathering soft rock with developed joints and fractures, introduced the laying out about the hole of pre splitting blasting that can apply in the high slope of the expressway in the section, and designed the parameter. The practice shows that it gained favourable effect by this method.
