Summary Since the macroscopical and microscopical examination of bull semen does not in all cases appear to give conclusive indications concerning the fertilizing capacity, an investigation of some biochemical parameters was undertaken. It was shown that the following biochemical examination could furnish some more information about the quality of the semen: 1. Determination of the phenylalanine- α-ketoglutarate transaminase activity both in the seminal plasma and in the whole semen. A high transaminase activity in the plasma points to leakage out of the spermatozoa, thus indicating an affection of the spermatozoa. In that case larger amounts of basic amino acids and of leucine were often found in the seminal plasma than would normally be observed. 2. Gas chromatographic examination of the steroids occurring both in the seminal plasma and in the spermatozoa. In a number of infertile bulls small peaks of progesterone were found together with those of other, unidentified compounds. 3. Investigation of the carbohydrate metabolism in the semen. In a number of fertile bulls the following deviations were found: a. the presence of quite a lot of fructose in the spermatozoa; b. the presence of glucuronic acid and / of other foreign compounds in the spermatozoa or in the plasma; c. an elevated pH in the seminal plasma some 2 to 3 hours after ejaculation; d. a low phenylalanine-α -ketoglutarate transaminase activity in the whole semen (the spermatozoa had been disrupted by freezing). 4. Investigation of the presence of reducing aldehydes (glycolaldehyde and glyceraldehyde) inside the spermatozoa. The determinations mentioned under 1, 3c and 3d can easily be carried out in the laboratories of each District Animal Health Service and of the A.I. stations; the same is true of the occurrence of amino acids in the seminal plasma and of fructose and aldehydes inside the spermatozoa if facilities are available for electrophoresis and chromatography.  相似文献

The first merogony of Eimeria bovis takes place in lymphatic endothelial cells of the ileum, resulting in the formation of macromeronts up to 250 mum. In this study, we investigated the host cell cytoskeleton (actin filaments, microtubules, spectrin, vimentin intermediate filaments) associated with parasitic development in vitro by confocal laser scanning microscopy (CLSM) using primary bovine umbilical vein endothelial cells (BUVEC) and bovine spleen lymphatic endothelial cells (BSLEC) as host cells. No prominent changes in the host cell cytoskeleton were detected 1-3 days after E. bovis sporozoite invasion. With ongoing meront maturation a significant increase in microtubules and actin filaments close to the parasitophorous vacuole (PV) was found. Mature macromeronts within the PV were completely enclosed by these cytoskeletal elements. Our findings suggest, that in order to guarantee the survival of the host cell on the enlargement of macromeronts, E. bovis needs not only to augment but also to rearrange its cytoskeletal system.  相似文献

Water scarcity and severe environmental degradation are causing water managers in the Fergana Valley, Uzbekistan to re-evaluate irrigation water use. Crop models could play an important role in helping farmers decide which systems (crops and irrigation technologies) are feasible. CROPGRO is a physiologically robust agronomic model, although the current version does not consider the effects of soil salinity on crop water use or growth. CROPGRO was modified to include a salinity response function and was tested for gypsiferous soils. A qualitative analysis of the model indicated the model performed as expected under a range of atmospheric, irrigation and crop tolerance scenarios. Model simulations compared very favourably for common bean (Phaseolus vulgaris) to results obtained in the greenhouse for yield and seasonal crop evapotranspiration with values of the Willmott agreement index (i) of 0.98 for both variables evaluated at different levels of salinity and deficit irrigation. Final biomass predictions were less satisfactory, although the modified model performed as well as the original model. The modified model was successfully tested with field data on common bean from an experiment in the Fergana Valley (i of 0.75 for ET and 0.74 for final yield), although the sensitivity of the model to a soil fertility function and relative nodule number made it difficult to assess the model performance.  相似文献

Maize cultivars (Zea mays L.) were evaluated for their aluminum (Al) sensitivity using intact plants and excised root tips exposed to 25 μM Al in nutrient solution of low ionic strength and pH 4.3. Aluminum supply increased callose formation and Al concentrations in root tips of intact plants as well as in excised root tips. Using intact plants, differences in Al sensitivity among cultivars could be characterized by Al‐induced callose formation, Al‐induced inhibition of root elongation, as well as Al contents in root tips as parameters. Significant correlations between Al‐induced callose formation and Al contents in root tips (r2 = 0.64**) and inhibition of root elongation (r2 = 0.80***) were found. Excised root tips did not show a significant Al‐induced inhibition of root elongation. While average Al‐induced callose formation was similar for root tips of intact plants and excised root tips, mean Al contents in excised root tips were up to 1.5‐fold higher than in root tips of intact plants after 24 h of Al treatment. Aluminum‐induced callose formation as found in excised root tips did neither correspond to Al‐induced callose formation nor to inhibition of root elongation of intact plants. The addition of 10 mM glucose to the incubation medium led to a significant increase in the elongation of excised root tips and a 2‐3‐fold increase in Al‐induced callose formation. Staining with triphenyl‐tetrazolium‐chloride (TTC) revealed increased viability of these root segments. However, these effects of glucose supply did not improve the characterization of the cultivars for Al resistance. The results presented suggest that Al exclusion mechanisms expressed in root tips of intact plants might be non‐operational in excised root tips. Therefore, the characterization of maize germplasm for Al resistance using excised root tips appears not to be reliable.  相似文献

South African juvenile dusky kob Argyrosomus japonicus are more abundant in turbid estuaries than in clear marine‐dominated estuaries. Turbidity can reduce light penetration into the water and create an environment different from that experienced by fish under culture conditions in mechanically filtered clear water. In order to optimize rearing conditions of this species, the effects of light intensity (23–315 lx) and feeding method (restricted ration vs. feeding to apparent satiation) on growth and food conversion ratio (FCR) of juvenile A. japonicus were assessed in a 56‐day growth trial. Fish weight increased from 7.2±1.6 to 41.9±10.2 g fish?1 at a growth rate of 3.25% body weight day?1. Light intensity did not significantly affect growth or FCR. Feeding method did not significantly affect growth rate, but average FCR was significantly better in treatments fed a ration of 3.6% body weight day?1 than in treatments fed to apparent satiation. Therefore, a light intensity range of 23–315 lx can be used to culture dusky kob juveniles. The better FCR in fish fed a restricted ration suggests that a ration of 3.6% body mass day?1 allowed good growth of juvenile dusky kob.  相似文献

In the semi-arid Brazilian Northeast, the exploitation of alluvial aquifers for irrigation and domestic supply to rural communities over the last 10 years has upset the traditional mechanisms of water resources management. In the Forquilha watershed (221 km2; 5°17″S, 39°30″W), the two main water resources are reservoirs (with a capacity exceeding 0.9-6.7 × 106 m3), used for domestic water supply only, and an alluvial aquifer (2.3 × 106 m3), used for irrigation and domestic water supply. From 1998 to 2006, the irrigated area with alluvial groundwater increased from 0 to 75 ha, and the fraction of population supplied through domestic water networks, using reservoirs and the aquifer, increased from 1% to 70%. Based on physical and socioeconomic issues, three main water territories have been defined (“Aquifer”, “Reservoirs”, and “Disperse Habitat”). Considering the next 30 years with a realistic population growth, three hypotheses regarding irrigated area (i.e., 0, 75, or 150 ha), and several possible water-management scenarios, hydrological balance models were built and used to simulate the different impacts on water resource availability and salinity. Simulation results showed that, in all cases, releases from the upstream main reservoir are necessary to keep reservoir salinity below 0.7 g L−1 and for guaranteeing domestic needs in the whole watershed. As a consequence, a management approach that takes into account the interrelations among the three territories is necessary. Moreover, the simulations showed that the area of irrigated fields cannot exceed the current extent (75 ha), or serious restrictions on water availability and salinity will take place. Moreover, important socioeconomic problems are expected, including a high cost of palliative water supply with tank trucks from external sources.  相似文献

MyD88 is a key downstream adapter for most Toll-like receptors (TLRs) and interleukin-1 receptors (IL-1Rs). MyD88 deficiency in mice leads to susceptibility to a broad range of pathogens in experimental settings of infection. We describe a distinct situation in a natural setting of human infection. Nine children with autosomal recessive MyD88 deficiency suffered from life-threatening, often recurrent pyogenic bacterial infections, including invasive pneumococcal disease. However, these patients were otherwise healthy, with normal resistance to other microbes. Their clinical status improved with age, but not due to any cellular leakiness in MyD88 deficiency. The MyD88-dependent TLRs and IL-1Rs are therefore essential for protective immunity to a small number of pyogenic bacteria, but redundant for host defense to most natural infections.  相似文献

Nitrogen efficiency is a complex trait. Identification of secondary plant traits correlating with N efficiency would facilitate the breeding for N‐efficient cultivars. Sixteen tropical maize cultivars differing in grain yield at low N supply (N efficiency) under field conditions in Zimbabwe exhibited a significant negative correlation between N efficiency and leaf senescence during grain filling. The same cultivars were studied for leaf senescence under N deficiency in a short‐term nutrient‐solution experiment. Leaf chlorophyll contents as estimated by SPAD values and photosynthesis rates were used as measures for leaf senescence. Cultivars differed both in SPAD values and photosynthesis rates of the older leaves during N deprivation. Significant negative correlations were found between SPAD values, photosynthesis rates in the nutrient‐solution experiment, and leaf‐senescence scores in the field experiments, and positive correlations were found between photosynthesis rates and grain yield under low‐N conditions in the field. Relationships between physiological root parameters, which were also investigated in the nutrient‐solution experiment, and N uptake or grain yield of the cultivars in the field could not be established. It is concluded, that the assessment of the capacity of a genotype to maintain a higher photosynthetic capacity of older leaves during N deficiency–induced senescence at the seedling stage is a suitable selection parameter for the N efficiency of tropical maize cultivars.  相似文献

Turkey poults were fed a vitamin D-deficient diet and examined for clinical signs and structural changes of bone and parathyroid glands. Vitamin D-deficient poults developed ricketic changes during days 10 to 14. Control poults (deficient diet plus vitamin D) did not develop rickets. In deficient poults, lengths of proliferating-prehypertrophied zones of growth plates increased significantly in the proximal tibiotarsus but were only slightly elongated in the distal tibiotarsus. Unmineralized hypertrophic chondrocyte zones increased in length rapidly in conjunction with a decrease in the length of mineralized hypertrophic degenerative zones; this occurred more rapidly in proximal than in distal tibiotarsus. Other ricketic changes included decreases in bone ash, total femoral bone ash (calcium, phosphorus, magnesium), bone length, and body weight. Plasma alkaline phosphatase was increased, calcium was normal, and phosphorus was normal or elevated. Parathyroids were hyperplastic and had foci of degeneration. Vitamin D3 metabolites 25OHD3, 1,25(OH)2D3, and 24,25(OH)2D3 were rapidly depleted. Increase in bone ash Ca/P ratios in deficient poults suggests that phosphorus may be selectively released from ricketic bone. Low 25OHD3 and 1,25(OH)2D3 of control poults early in the experiment suggests that 1,400 IU of vitamin D3/kg of feed may not be an adequate level of vitamin D3 for growing turkey poults.  相似文献
