The storage of unfertilized oocytes, either immature, maturing or mature, is still unsatisfactory. Here we describe an approach in which germinal vesicles isolated as karyoplasts from immature oocytes are vitrified by open the pulled straws (OPS) method in evacuated porcine zonae pellucidae. After thawing, their survival was almost absolute. Moreover, when thawed GV-karyoplasts were fused to immature oocyte cytoplasts the maturation of reconstructed cells resulted in the production of secondary oocytes--metaphase II. 相似文献

Moss surveys provide spatially dense data on environmental concentrations of heavy metals and nitrogen which, together with other biomonitoring and modelling data, can be used for indicating deposition to terrestrial ecosystems and related effects across time and areas of different spatial extension.Context
For enhancing the spatial resolution of measuring and mapping atmospheric deposition by technical devices and by modelling, moss is used complementarily as bio-monitor.Aims
This paper investigated whether nitrogen and heavy metal concentrations derived by biomonitoring of atmospheric deposition are statistically meaningful in terms of compliance with minimum sample size across several spatial levels (objective 1), whether this is also true in terms of geostatistical criteria such as spatial auto-correlation and, by this, estimated values for unsampled locations (objective 2) and whether moss indicates atmospheric deposition in a similar way as modelled deposition, tree foliage and natural surface soil at the European and country level, and whether they indicate site-specific variance due to canopy drip (objective 3).Methods
Data from modelling and biomonitoring atmospheric deposition were statistically analysed by means of minimum sample size calculation, by geostatistics as well as by bivariate correlation analyses and by multivariate correlation analyses using the Classification and Regression Tree approach and the Random Forests method.Results
It was found that the compliance of measurements with the minimum sample size varies by spatial scale and element measured. For unsampled locations, estimation could be derived. Statistically significant correlations between concentrations of heavy metals and nitrogen in moss and modelled atmospheric deposition, and concentrations in leaves, needles and soil were found. Significant influence of canopy drip on nitrogen concentration in moss was proven.Conclusion
Moss surveys should complement modelled atmospheric deposition data as well as other biomonitoring approaches and offer a great potential for various terrestrial monitoring programmes dealing with exposure and effects.24.

The effects of temperature on anatomical and chemical characteristics of Quercus cerris cork were examined. Cork samples were subjected to isothermal air heating between 150 °C and 400 °C and analyzed for mass loss, cellular structure and chemical composition.The thermal decomposition of Q. cerris cork is similar to that of Q. suber cork. Cork is thermally stable below 200 °C and after that degradation depended on temperature and heating time with increasing mass loss, i.e. 3% at 200 °C 10 min and 46% at 350 °C 60 min. With temperature and starting at 200 °C, cells expanded, cell wall thickness was reduced and corrugations were lost.Extractives degraded at lower temperatures, although aliphatic extractives were found to be more stable. Suberin from Q. cerris was more heat resistant than Q. suber suberin, while lignin showed similar resistance.These results provide a basis for studies on the production of Q. cerris bark expanded cork agglomerates for insulation purposes. 相似文献

Hydropower plants on the lower river Sava, Slovenia, were developed without sealing the underground upstream. As a consequence, without the countermeasures of elevating and recultivating, the agricultural land on the river banks would be inundated because of the water‐table increase of the river. To remedy this, the fields were elevated and recultivated. The goal of this study was to assess soil quality and production potential after land raising and recultivation and to answer the question whether it is possible to recover soil quality and crop yield after large‐scale mass manipulation, such as land raising. After recultivation and after the second year of land reuse for the two cultures grass–clover mixture and corn on two sites, Middle Pijavsko and Lower Pijavsko, soil physical and chemical characteristics and crop yields were evaluated. Mixing of topsoil with the second horizon during removal and during backfill with filling material resulted in uneven soil fertility, plant growth and decreased yield. Driving on the refilled second layer with heavy machinery caused soil compaction in the Middle Pijavsko in spite of favourable soil texture (loam, 38·7% sand). On the areas with very high sand content (over 55%) and low clay content (10%), soil compaction was not as severe or persistent. Recultivation measures restored the agricultural land almost to the production potential prior to powerplant construction. The factors affecting the adoption of modern varieties(MVs) of rice and impact on poverty in Odisha, India were discussed. A total of 363 households from Cuttack and Sambalpur districts of Odisha via multistage sampling technique participated in the survey. The Cragg's Double hurdle model was used to model the determinants of adoption and intensity of adoption of MVs of rice, and the propensity score matching was used to analyze the impact of adoption on poverty. The results showed that age, education, risk aversion, land size, yield, perception of MVs as high yielding, resistant to diseases and availability of MVs positively influenced the decision to adopt. However, variables such as household size, experience of a farmer, off-farm job participation, amount of credit received, cost of seeds, insecticides and fertilizers negatively influenced the adoption of MVs. Intensity of adoption of MVs was negatively influenced by experience of a farmer, cost of fertilizer and marketability of MVs, and positively affected by household size, risk aversion, land size, cost of insecticides, perception of MVs as high yielding and availability of MV seeds. Poverty incidence, gap and severity were high among non-adopters to adopters of MVs. After matching adopters and non-adopters of MV groups using four different algorithms of nearest neighbour matching, stratification matching, radius matching and kernel matching, the impact of MV adoption resulted in higher per capita monthly household expenditure by about US$ 52.82 to US$ 63.17. 相似文献

Microalgae are well known for their biotechnological potential, namely with regard to bioactive lipidic components—especially carotenoids and polyunsaturated fatty acids (PUFA), well-known for therapeutic applications based on their antioxidant capacity. The aim of this work was to evaluate the influence of four distinct food-grade solvents upon extractability of specific lipidic components, and on the antioxidant capacity exhibited against both synthetic (2,2-diphenyl-1-picrylhydrazyl (DPPH•) and 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS+•)) and biological reactive species (O2•− and •NO−). A eukaryotic microalga (Scenedesmus obliquus (M2-1)) and a prokaryotic one (Gloeothece sp.) were used as case studies. Concerning total antioxidant capacity, the hexane:isopropanol (3:2) and acetone extracts of Sc. obliquus (M2-1) were the most effective against DPPH• and ABTS+•, respectively. Gloeothece sp. ethanol extracts were the most interesting scavengers of O2•−, probably due the high content of linolenic acid. On the other hand, acetone and hexane:isopropanol (3:2) extracts were the most interesting ones in •NO− assay. Acetone extract exhibited the best results for the ABTS assay, likely associated to its content of carotenoids, in both microalgae. Otherwise, ethanol stood out in PUFA extraction. Therefore, profiles of lipidic components extracted are critical for evaluating the antioxidant performance—which appears to hinge, in particular, on the balance between carotenoids and PUFAs. 相似文献

Maritime pine (Pinus pinaster Ait.) is an important timber and pulpwood species covering about 4 million ha in Portugal, Spain and France. This work studied the cross-sectional distribution of heartwood and sapwood along the stem using 3D modelling on 20 trees. The target was to assess the potential of this species for the industrial production of heartwood sawn products. The maritime pine stems were characterized by wide sapwood of 10 cm, in average, and the presence of heartwood at all height levels with constant diameter up to 35 % tree height, decreasing afterwards. In the radial section, the heartwood follows a circumferential-like shape which changes along the stem. Pith eccentricity index values were low and homogeneous regarding to stem and higher for heartwood. Heartwood cross-sectional shape was constant up to 45 % of total height of the tree and more elongated upwards. The constant heartwood diameter, low taper and pith eccentricity index, and an approximate circular shape up to 10 m height indicated these species’ potential to produce solid wood products made out of heartwood. 相似文献
