Twenty-five cats with clinical signs of upper respiratory tract disease and five cats without upper respiratory signs underwent echolaryngography. Direct inspection of the larynx under general anesthesia was undertaken in all cats and used as the 'gold standard' for the diagnosis of laryngeal diseases. The aims were to: (a) establish which anatomic structures of the larynx are visible ultrasonographically in cats without upper respiratory tract disease, (b) establish which laryngeal abnormalities can be detected and accurately localised using ultrasonography and (c) evaluate in which conditions the technique may provide complementary information or an alternative method of investigation. The ultrasound investigation accurately indicated the presence and location of cysts and masses. Abnormal laryngeal movements were also detected, although it was difficult to confirm whether these were unilateral or bilateral. Vocal cord thickening was seen but the underlying cause could not be established with this technique.  相似文献

Element concentrations in tree rings can be used to monitor changes in environmental quality. With regard to the detection of incipient soil acidification, the manganese concentration in soils and plants is a significant marker for the switch of acid buffering in soils mainly with the exchange of base cations or with the dissolution of aluminium oxides. This is a site-specific non-linear event, indicating the onset of Al3+ dominance in the soil solution, were damages to vegetation due to acid stress become possible. This turning point is also a marker for the attainment of pH 4.2 in soils, the critical threshold used for critical load calculations. On a plot of the German environmental monitoring in forests the element concentrations in tree rings of 60-year-old spruces reveal a distinct decline in the Mn concentration, beginning in the late 1960s ending in the late 1970s. With this information it was possible to assume a base saturation in the soil of about 15–20% in the late 1960s, and to model the development of the base saturation of the site. A decline from 17.5 to 6% within one decade could be related to the deposition. This is in accordance with the base saturation of 6.5%, measured in 1993 for this site, but also for adjacent spruce sites on the same geological substrate. The knowledge of the time span were this site-specific non-linear event occurred is essential for the reconstruction of the soil chemistry of a site. Moreover, it enables the assignment of observations like ‘forest damages’ to the onset of changes in environmental quality.  相似文献

Paratuberculosis or Johne's Disease, caused by Mycobacterium avium subspecies paratuberculosis (MAP), is a notifiable disease in Germany which produces enormous economical losses in dairy farms. At present,there is no confirmed data about the actual number of infected livestock herds in Germany. A countrywide monitoring program to evaluate the prevalence in dairy herds would only be economically feasible on the basis of bulk milk testing. In this study, we evaluated two ELISA test kits (SVANOVIR Ptb-ELISA, IDEXX-M.pt. Milk test kit) for the detection of antibodies against MAP in bulk milk. First, the Paratuberculosis-status of the herd derived from the history of the farm was used as a gold standard. Paratuberculosis-negative farms were tested negative with each test, but paratuberculosis-positive or Paratuberculosis-serologically-positive farms were detected only in one case (Svanovir) or three cases (IDEXX), respectively. Even if inconclusive results are counted as positive, 82.9 % (Svanovir) or 80 % (IDEXX) of the paratuberculosis-positive or serologically paratuberculosis positive farms were not detected. Nevertheless, a re-validation of both ELISAs by means of ROC and TG-ROC analyses was attempted by searching for ideal cut-offs, optimised for bulk milk. If a high specificity was selected, no acceptable sensitivity could be reached.The best results were obtained using a sensitivity of 32.3 % at a specificity of 100 % (Svanovir). With a small change of the cut-off value, the sensitivity increased to still 57 %, but this reduced the specificity to 67 %. Similar results were obtained with the IDEXX-ELISA. We then evaluated the Svanovir-ELISA for the detection of bulk milk samples on the basis of the current paratuberculosis prevalence within 69 dairy herds from Rhineland-Palatinate using individual milk samples.When the bulk milk samples were tested in two different laboratories using the same ELISA, considerable differences in the results became evident. Nearly all samples were tested with a higher relative test result in one laboratory, which often led to differences in the classification of the prevalence levels.The estimated within-herd seroprevalences ranged between 0 % and 37 %.There was little agreement between the historical paratuberculosis herd status and the within-herd prevalence in milk serum, as reflected in a kappa-index of 0.146.To determine the sensitivity and specificity of the bulk milk ELISA by ROC and TG-ROC analysis, 116 bulk milk samples were used that had been obtained from the 69 dairy herds participating in the study. The optimal ratio of sensitivity (81 %) and specificity (77 %) relative to a "gold standard" was obtained when the cut-off was set at the 10 % level. These values for sensitivity and specificity were better than those obtained in an evaluation of the same ELISA in which the historical Paratuberculosis herd-status was used as a "gold standard." The results of this study question the suitability of the available ELISAs for bulk milk testing.Taking into account that the Svanovir-ELISA for individual milk samples has a sensitivity of 60 96% relative to the blood serum variant of the test, and that the latter has also a limited sensitivity due to the pathogenesis of paratuberculosis, the available test systems examined in this Study do not seem to be suitable for herd diagnosis by using bulk milk samples.  相似文献

Osmotic stress as well as silicon (Si) improve the resistance of barley (Hordeum vulgare cv. Ingrid) against barley powdery mildew (Blumeria graminis f. sp. hordei Speer). Nothing is known about interactions, particularly whether Si is necessary for pathogen resistance induced by osmotic stress. In this paper, we show that Si nutrition was not necessary for osmotic‐stress‐induced pathogen resistance. Si‐mediated resistance could, however, be enhanced by osmotic stress and vice versa. Even at maximum Si‐mediated resistance, further enhancement of pathogen resistance by osmotic stress was possible. The fungus was controlled by the formation of effective papillae in both treatments. The combined effect of Si and osmotic stress was as strong as the calculated addition of the Si and the osmotic‐stress effect. Our data clearly show that the effect of osmotic stress and Si is not competitive but additive. A synergistic action of both treatments cannot be supported by our data. It is assumed that the basal pathogen resistance of barley is enhanced by Si due to strengthening of papillae in addition to the increased formation of effective papillae induced by osmotic stress. Therefore, the addition of Si increases pathogen resistance equally at all investigated NaCl concentrations. A function of Si in pathogen defense exceeding the strengthening of papillae is not supported by our data.  相似文献

The change of aggregate size classes caused by rainfall was examined using six different silly soils. The surface elevation profile lines were measured with a non-contact laser scanner with a measurement resolution of O.1 mm. The profile length ratio RZ and, based on the Fourier-analysis of the elevation profile line, the partial roughness were determined. The results demonstrate that in contrast to the profile length ratio RZ, the evaluation of partial roughness is an adequate method to determine the proportion of aggregate size fractions on total surface roughness. Furthermore, the results show that — independent of the soil texture and history — the smallest aggregate size classes are the most labile fractions. These small and labile aggregate size fractions are involved in the soil erosion process, especially in surface sealing. The extent of surface sealing, however, is influenced by soil texture, too.  相似文献

Due to high nitrogen deposition in central Europe, the C : N ratio of litter and the forest floor has narrowed in the past. This may cause changes in the chemical composition of the soil organic matter. Here we investigate the composition of organic matter in Oh and A horizons of 15 Norway spruce soils with a wide range of C : N ratios. Samples are analyzed with solid‐state 13C nuclear magnetic resonance (NMR) spectroscopy, along with chemolytic analyses of lignin, polysaccharides, and amino acid‐N. The data are investigated for functional relationships between C, N contents and C : N ratios by structural analysis. With increasing N content, the concentration of lignin decreases in the Oh horizons, but increases in the A horizons. A negative effect of N on lignin degradation is observed in the mineral soil, but not in the humus layer. In the A horizons non‐phenolic aromatic C compounds accumulate, especially at low N values. At high N levels, N is preferentially incorporated into the amino acid fraction and only to a smaller extent into the non‐hydrolyzable N fraction. High total N concentrations are associated with a higher relative contribution of organic matter of microbial origin.  相似文献

Since 2016, annually occurring species-specific die-offs of brown trout (Salmo trutta fario) occurred in the Thur River, situated in the Eastern part of Switzerland. These events lead to drastically reduced population densities in the impacted river regions. Clinical signs in brown trout and mortality were restricted to few weeks in August/September. To characterize the syndrome and to find possible causes, from end of March to November 2018, one-year-old brown trout (Salmo trutta fario) and rainbow trout (Oncorhynchus mykiss) were exposed to water from Thur River, fish were sampled regularly and screened for infectious agents, including viral metagenomics, and pathology was described. Starting approximately four months post-exposure, brown trout showed severe lymphohistiocytic pancarditis and necrotizing and haemorrhagic hepatitis. These lesions were recorded until the end of the experiment in November. Rainbow trout were not affected at any point in time. No infectious agents could be identified so far as cause of disease, especially no viral aetiology. Even if pathogenesis and pathology point in the direction of an infectious agent, a causative relationship could not be confirmed and aetiology remains unclear.  相似文献

Phosphorus (P) is essential for sustainable forest growth, yet the impact of anthropogenic impacts on P leaching losses from forest soils is hardly known. We conducted an irrigation experiment with 128 mesocosms from three forest sites representing a gradient of resin extractable P of the A‐horizon. On each site we selected a Fagus sylvatica and a Picea abies managed subsite. We simulated ambient rain (AR), anthropogenic nitrogen input (NI) of 100 kg (ha · a)?1 and forest liming (FL) with a dolomite input of 0.3 Mg (ha · a)?1. Soil solution was extracted from the organic layer, 10 cm depth and 20 cm depth of the mesocosms, and analyzed for molybdate reactive phosphorus (MRP) and molybdate unreactive phosphorus (MUP). Additionally, we separated colloids from the soil solution using Asymmetric Field Flow Fractionation for assessing the colloidal fraction of total element concentrations. NI increased MRP and MUP concentrations for all plots with one exception, while FL decreased MRP and MUP with the exception of another plot. While the irrigation treatments had little impact on the P‐richest site, MRP and MUP concentrations changed strongly at the poorer sites. The colloidal fraction of P in the soil solution equaled 38–47% of the total P load. Nitrogen input and liming also affected the Fe, Al, Ca, and Corg contents of the colloidal fraction.  相似文献
