We investigated the prevalence of an anatomic variant of the proximal femur, termed the broomstick-like femoral head and neck formation, and its influence on the Féderation Cynologique Internationale (FCI) hip dysplasia score in 294 German Shepherd dogs. One-hundred and eighty (61%) of the 294 dogs in our study had this anatomic variant. The calculated area of the femoral heads in dogs with a broomstick-like conformation was 4.5 +/- 0.6 cm2 on the hip-extended view. In dogs with a normal femoral head, the calculated area of the femoral head was significantly larger at 4.8 +/- 0.6 cm2 (P < 0.05). In the frog-leg view, there was no significant difference in femoral head area between dogs with the broomstick-like conformation and normal dogs. There was no difference in the antetorsion angle between dogs with broomstick-like conformation and normal dogs. There was also no difference in the distraction ratio between the two phenotypes. The official FCI hip score was similar in dogs with and without the broomstick-like conformation. The average heritability of the broomstick-like conformation was 0.3 +/- 0.1, suggesting heritable influence. We conclude that the broomstick-like conformation is a common finding in the German shepherd dog and has genetic base. The broomstick-like conformation does not appear to be associated with the presence of canine hip dysplasia and it can therefore be assumed to be a normal anatomic variant.  相似文献

Because essential amino acids are metabolized in the liver, liver diseases may impair their catabolism. In this study, serum L-phenylalanine concentrations in 28 dogs with liver diseases were compared with those of 28 healthy dogs and 13 dogs with nonhepatic diseases. Dogs with liver diseases had significantly increased L-phenylalanine serum concentrations compared to healthy dogs (P<0.001) and to those with nonhepatic diseases (P<0.01). There were no significant differences among the L-phenylalanine serum concentrations of dogs with different degrees of liver diseases. The sensitivity and specificity of L-phenylalanine to fasting bile acids were comparable.  相似文献

The National Resistance Monitoring of the Federal Office of Consumer Protection and Food Safety (BVL) is to determine the prevalence of resistance of bacterial pathogens from animals using a valid database. From 2004 to 2006, a total of 1,472 Pasteurella multocida strains isolated from pigs with acute respiratory tract diseases was submitted to the BVL and examined. Of these, 1,11 (75.5 %) were included in the study and tested using 24 different antimicrobial substances. The results showed that the resistance level is generally low, with the exception of the substances tetracycline, trimethoprim, and the combination trimethoprim/sulfamethoxazole. It also became clear that resistance data need to be evaluated separately for each of the animal production categories, so that a realistic figure of the current resistance level can be presented. This knowledge provides information about the resistance situation in Germany, and helps deduce the necessary management measures that must be taken to minimize resistance to antibiotics. Furthermore, it provides valuable information that can form the basis for empirical therapy, so that the National Resistance Monitoring makes an important contribution to the safety of food derived from animals and consequently aids the improvement of consumer protection.  相似文献

The histomorphological findings and immunohistochemical expression of matrix metalloproteinases (MMPs-1, 2, 9 and 14) and their tissue inhibitors (TIMPs-2, 3 and 4) are reported in the parietal (pTV) and septal leaflets (sTV) of the tricuspid valves in normal dogs and dogs with chronic valvular disease (CVD). The layers of the normal sTV were not as well defined as in the pTV and the spongiosa of the sTV contained abundant mucopolysaccharides (MPS) and adipocytes. In CVD, there was expansion of the spongiosa of the pTV due to deposition of MPS, leading to formation of nodules along the free edge. In CVD, there was fibrosis of the atrialis of the sTV and formation of nodular deposits of MPS in the spongiosa and ventricularis, mainly affecting the proximal and middle parts of the leaflet. In dogs with normal pTV and sTV, MMPs-1 and 14 and TIMPs-2, 3 and 4 were expressed, while MMPs-2 and 9 were absent. In mild CVD, expression of MMPs-2, 9 and 14 were increased in the pTV, whereas small foci within the spongiosa contained MMP-9 and TIMP-3 positive cells. In advanced CVD, MMP-14 also was increased in the pTV. In mild CVD, there was increased expression of MMPs-1 and 2 and TIMP-2, but decreased expression of TIMP-4, in the sTV. Small foci with expression of MMP-14 and TIMPs-2, 3 and 4 were also present in the sTV in mild CVD. In advanced CVD, there was increased expression of MMPs-2 and 9 and TIMP-2 in the sTV. In CVD there is upregulation of various MMPs in the pTV, whereas there is a complex alteration in expression of MMPs and TIMPs in the sTV.  相似文献

BACKGROUND: Resistance to photosystem II inhibitors—triazines (atrazine) and triazinones (metamitron, metribuzin)—in Chenopodium album L. is caused by the serine 264 to glycine mutation in the D1 protein. This mutation has been detected in C. album collections from Belgium with unsatisfactory metamitron efficacy in the field and was confirmed in greenhouse resistance bioassays. Incomplete herbicide efficacy in practice can also be caused by reduced uptake due to environmental conditions. Hence, for reliable differentiation and resistance identification, a rapid method for mutation detection in the target gene psbA is required. RESULTS: Dose–response curves obtained in herbicide greenhouse assays with metamitron‐resistant and ‐susceptible reference biotypes showed that a dose of 2 L ha?1 metamitron was suitable for discrimination. A psbA PCR‐RFLP was developed, based on the presence of a FspBI restriction enzyme recognition site, covering D1 codon 264 in susceptible genotypes. A paper‐based DNA extraction allowed direct processing of leaf samples already in the field. In order to detect the mutation even in mixed seed samples, a nested PCR‐RFLP was also developed. CONCLUSION: The method allows exhaustive surveys screening C. album leaf or seed samples for the occurrence of the D1 Ser264Gly mutation to confirm or disprove metamitron resistance in the case of unsatisfactory control. When diagnosing disease of the stifle in dogs ultrasonography is a good addition to clinical and radiological examination. Radiology can evaluate the bony aspects of the joint and their relationship to each other. In contrast, sonography allows visualization of the soft tissue. For most evaluations the 7.5 MHz linear scanner is suited best. Normal stifles of 58 dogs of different breeds were evaluated using a standardized examination procedure. This procedure had been derived from that used in humans. The stifle is divided into several regions which are examined and evaluated. These are the suprapatellar, infrapatellar, lateral, caudal and medial region. One hundred twenty seven patients which had problems associated with the stifle joint were examined sonographically after a clinical and radiographic exam. Osteochondrosis dissecans, ruptured cranial cruciate ligament, meniscal damage, arthritis, tumor, post surgical conditions, injuries of the patella, patellar ligament or tibial tuberosity and luxating patella were examined sonographically and the findings recorded.  相似文献

Infectious haematopoietic necrosis (IHN) and viral haemorrhagic septicaemia (VHS) are OIE‐listed and notifiable viral fish diseases which are controlled by eradication and surveillance programmes globally. The present study provides improved RT‐qPCR procedures based on recently described OIE protocols. Improvements comprise the design of a new TaqMan® probe, replacing a TaqMan® MGB probe that turned out to show impaired binding. Reason for this is SNPs detected in the nucleoprotein N gene sequences of IHNV strains targeted by the RT‐qPCR. Furthermore, the IHNV and VHSV RT‐qPCR assays were realized as one‐step and one‐run procedures supplemented by an endogenous control system. The IHNV and VHSV RT‐qPCR assays are characterized by a technical sensitivity of 19 and 190 gene equivalents (cRNA) and an analytical sensitivity of 2–7 and 13 TCID50/ml, respectively. For verification purposes, 105 IHNV and 165 VHSV isolates and several non‐targeted viral and bacterial pathogens were included and returned adequate results. However, in field samples divergent results left 14 samples of 154 undetected for IHNV and one sample of 127 for VHSV using cell culture. The study shows that RT‐qPCR assays ensure facilitated and reliable testing on IHNV and VHSV in eradication and surveillance programmes.  相似文献

Purpose

The present study focuses on a compositional characterization of the humic acid (HA) fraction of several wetland soils using solid-state 13C NMR spectroscopy. The data were analysed using the molecular mixing model (MMM), based on an empirical approach by Nelson and Baldock. The compositional data from HAs obtained with this model were used to obtain a wider assessment of the process of humification from comparison of total soil wetland organic matter composition and HA composition.

Materials and methods

Twenty samples of humic acids (HAs) isolated from a Mediterranean semiarid wetland (‘Tablas de Daimiel’, central Spain) were studied using elemental analysis and cross polarization magic angle spinning (CPMAS)13C nuclear magnetic resonance (NMR) spectroscopy. The NMR data were analysed with the molecular mixing model (MMM) considering up to six generic components (carbohydrate, protein, lignin, lipid, char and ‘carbonyl’). HAs are considered a conceptual mixture of these model components, and the MMM determines the proportions of the characteristic biomolecules contributing to HA composition.

Results and discussion

The composition of the HAs under study depends on local factors such as site vegetation and occurrence of fire. Correlations between the proportions of the six generic components and further comparison with those determined for the unfractionated OM (whole sample, WS), gave information on HA origin and humification mechanisms. In particular, the proportions of char and carbohydrate (R 2 0.637) and contents of lignin and protein (R 2 0.471) in the HAs were negatively correlated (P < 0.05). Significant correlations (R 2 0.439) also existed for char contents in whole sample (WS) compared to HA, and for carbohydrates in WS compared to HA (R 2 0.558). Char proportion grew in HA with respect to the WS, and carbohydrates dropped to a half on average in HA compared to WS.

Conclusions

Two different humification mechanisms could be identified for no-fire and fire areas. In the former, HA-char was preserved selectively from char in the sample, whereas in the latter, char was newly formed by fire effect.

Applications of hyperspectral remote sensing data to derive relevant properties for precision agriculture are described. Green leaf area index, fraction of senescent material and grain yield are retrieved from the hyperspectral data. Two sensors were used to obtain these data; the airborne visible/infrared imaging spectrometer AVIS and the space-borne compact high-resolution imaging spectrometer CHRIS; they show the applicability of the methods to different spatial scales. In addition, the bi-directional observation capability of the CHRIS sensor is used to derive information about the average leaf angle of the canopies which are used to link canopy structure with phenological development. Derivation of the canopy properties, green leaf area index and fraction of senescent material was done with the radiative transfer model, SLC (soil–leaf–canopy). The results were used as input into the crop growth model PROMET-V to calculate grain yield. Two years of data from the German research project preagro are presented.  相似文献
