Depriving rats of rapid eye movement (REM) sleep was shown to lower their thresholds and raise their response rates for rewarding brain stimulation. Conversely, allowing rats to self-stimulate while they were being deprived of this sleep form reduced the amount of REM rebound during recovery from deprivation. These results demonstrate a reciprocal relation between rewarding brain stimulation and REM sleep. 相似文献

The cytokine interleukin-10 (IL-10) has shown promise in clinical trials for treatment of inflammatory bowel disease (IBD). Using two mouse models, we show that the therapeutic dose of IL-10 can be reduced by localized delivery of a bacterium genetically engineered to secrete the cytokine. Intragastric administration of IL-10-secreting Lactococcus lactis caused a 50% reduction in colitis in mice treated with dextran sulfate sodium and prevented the onset of colitis in IL-10(-/-) mice. This approach may lead to better methods for cost-effective and long-term management of IBD in humans. 相似文献

将Imt1(Inositol O-methyltransferase,肌醇甲基转移酶)构建成酵母表达载体pBD#-3,在酵母双突变体gpd#+(-)(glycerol 3-phosphate-dehydrogenase,3-磷酸甘油脱氢酶)中高效表达,使gpd#+(-)突变株对氯化钠的耐性由2.5%提高到5.0%。生化分析表明,在每mg干酵母细胞内积累高达2.2nmol的Ononitol(芒柄醇)含量。Western blot分析证明了Imt1基因的表达。这一研究为植物耐盐基因工程开辟了一条新途径。 相似文献

The OSIRIS cameras (optical, spectroscopic, and infrared remote imaging system) onboard the European Space Agency's Rosetta spacecraft observed comet 9P/Tempel 1 for 17 days continuously around the time of NASA's Deep Impact mission. The cyanide-to-water production ratio was slightly enhanced in the impact cloud, compared with that of normal comet activity. Dust particles were flowing outward in the coma at >160 meters per second, accelerated by the gas. The slope of the brightness increase showed a dip about 200 seconds after the impact. Dust Afrho values before and long after the impact confirm the slight decrease of cometary activity. The dust-to-water mass ratio was much larger than 1. 相似文献

Chimeras of two isoprenoid synthases catalyze all four coupling reactions in isoprenoid biosynthesis
The carbon skeletons of over 55,000 naturally occurring isoprenoid compounds are constructed from four fundamental coupling reactions: chain elongation, cyclopropanation, branching, and cyclobutanation. Enzymes that catalyze chain elongation and cyclopropanation are well studied, whereas those that catalyze branching and cyclobutanation are unknown. We have catalyzed the four reactions with chimeric proteins generated by replacing segments of a chain-elongation enzyme with corresponding sequences from a cyclopropanation enzyme. Stereochemical and mechanistic considerations suggest that the four coupling enzymes could have evolved from a common ancestor through relatively small changes in the catalytic site. 相似文献

In Caenorhabditis elegans, an effective RNA interference (RNAi) response requires the production of secondary short interfering RNAs (siRNAs) by RNA-directed RNA polymerases (RdRPs). We cloned secondary siRNAs from transgenic C. elegans lines expressing a single 22-nucleotide primary siRNA. Several secondary siRNAs start a few nucleotides downstream of the primary siRNA, indicating that non-RISC (RNA-induced silencing complex)-cleaved mRNAs are substrates for secondary siRNA production. In lines expressing primary siRNAs with single-nucleotide mismatches, secondary siRNAs do not carry the mismatch but contain the nucleotide complementary to the mRNA. We infer that RdRPs perform unprimed RNA synthesis. Secondary siRNAs are only of antisense polarity, carry 5' di- or triphosphates, and are only in the minority associated with RDE-1, the RNAi-specific Argonaute protein. Therefore, secondary siRNAs represent a distinct class of small RNAs. Their biogenesis depends on RdRPs, and we propose that each secondary siRNA is an individual RdRP product. 相似文献
