Background

Patulin, a toxic mold metabolite, has been found as natural contaminant of processed fruits, most notably apples, apple juices and other apple-based products. A number of adverse health effects in humans and animals are associated with patulin intoxication. The current study was performed to analyse possible toxic effects of patulin on femoral bone microstructure in adult rabbits in detail. Fourteen clinically healthy four-month-old rabbits of both sexes (6 males and 8 females) were included in the study. Animals from the experimental groups (group E♂, n = 3; group E♀, n = 4) were injected intramuscularly with patulin at dose 10 μg/kg body weight two times a week for 28 days. The dose of patulin was estimated based on the maximum permitted level of patulin for apple products intended for infants and young children. Three males and four females without patulin administration served as controls (groups C♂ and C♀). Cortical bone thickness and qualitative and quantitative histological characteristics of compact bone tissue were investigated.

Results

Intramuscular applications of patulin significantly increased the thickness of cortical bone in both sexes of rabbits. In patulin-exposed males, an absence of primary vascular longitudinal bone tissue near the endosteal border was observed, which could be associated with intensive bone remodeling. Femoral diaphyses of females displayed a lower number of secondary osteons in the middle part of the substantia compacta, and occurrence of the osteons near the periosteum. This could indicate alterations in bone turnover. Histomorphometrical evaluations showed significantly increased sizes of the primary osteons’ vascular canals (P < 0.05) in males exposed to patulin possibly due to mycotoxin-induced increased levels of testosterone.

Conclusions

This study demonstrates significant impact of intramuscular application of patulin on bone microstructure in adult rabbits. Moreover, we have found that the effects of patulin on qualitative and quantitative histological characteristics of compact bone are sex-dependent.  相似文献

This paper aims to characterise soils of 12 wetlands, of which ten are Ramsar sites, in the ecocomplex of wetlands of the Hauts Plateaux region in Northeast Algeria. Soil samples from every site were collected following the four cardinal directions, along a transect covering the peripheral vegetation belts, and from two depths of the surface horizon. Each soil sample was analysed to determine electrical conductivity, pH, total carbonates, gypsum, chlorides, bicarbonates, sulfates and the particle size. The soil texture and chemical facies (Cl–SO4–HCO3) of each site were identified and discussed. Changes in physicochemical parameters were tested according to the spatial features of sites (orientations, vegetation transects and sample depth). A great heterogeneity was found between soils of sampled sites. Indeed, soil physicochemical characteristics differed from one site to another and between belts of the natural vegetation within the same site. Overall, the study wetlands were characterised by salty to very salty soils (electrical conductivity = 3·46 ± 2·44 dS m−1), of neutral to alkaline pH (6·9–8·1), moderately calcareous (CaCO3 ranged between 15·7% and 33·7%) and little to extremely gypsiferous (gypsium varied from 2·1% to 39·4%). The dominant soil texture classes were medium textures (loam, sandy loam or silty clay loam). Chemically, chlorides (18·5 ± 16·3 Meq/100 g) and/or sulfates (16·5 ± 12·5 Meq/100 g) dominated soil solutions of these environments but with slight bicarbonate contents (0·6 ± 2·6 Meq/100 g). Moreover, there were poor correlations between physicochemical parameters, which indicates interactions between certain parameters under the effect of specific habitat conditions. Haematological parameters of 2‐year‐old carp (Cyprinus carpio L.) were assessed to study the protective effect of chloride on the health of fish exposed to elevated nitrite concentrations. Four groups of carp were exposed to different concentrations of nitrite and chloride for 96 h (group E1: 67 mg L?1 NO2?, 11 mg L?1 Cl?; group E2: 67 mg L?1 NO2?, 100 mg L?1 Cl?; group E3: 0 mg L?1 O2?, 100 mg L?1 Cl? and group C: 0 mg L?1 NO2?, 11 mg L?1 Cl?). The main haematological response of carp to an acute exposure to nitrite (group E1) was a significant decrease (P<0.05) in haemoglobin concentrations (53.40±6.61 g L?1), haematocrit (0.21±0.02 LL?1), erythrocyte count (1.13±0.12 TL?1), leucocyte count (7.1±4.19 GL?1) and lymphocyte count (5.28±2.51 GL?1), and a significant increase in methaemoglobin concentration (90.50±4.38%, P<0.01) and mean corpuscular haemoglobin concentration (0.27±0.2 LL?1, P<0.05). At higher chloride concentrations (group E2), a lower nitrite toxicity was observed. In group E2 carp, methaemoglobin made up 38.32±13.30%. Erythrocytes in carp exposed to nitrite showed qualitative changes. Compared with the control group C, group E1 carp showed a significantly higher number (P<0.05) of elongated erythrocytes, with the nucleus located at one cell pole (0.519±0.388 TL?1). All erythrocytes of group E1 carp had remarkably clear cytoplasms compared with the cytoplasm in the control group C. The biochemical values found were comparable with those found in controls. The main histological lesions were found in the gills of carp exposed to nitrite and consisted of hyperplasia and an elevated number of chloride cells.  相似文献

The aim of this study was to evaluate the contribution of three plasma antioxidants (albumin, uric acid, SH groups) to the plasma total peroxyl radical-trapping antioxidant capacity (TRAP) in 2 and 4 h of intestinal reperfusion in rats. TRAP increased significantly both after 2 and 4 h of reperfusion. Neither albumin nor SH groups contributed significantly to this increase. TRAP was strongly influenced by the increase in uric acid concentration and also probably by the cell destruction caused by oxidative stress. Since the TRAP increase was accompanied by an increase in the level of thiobarbituric acid reactive substances (TBARS, a marker of lipid peroxidation), we can conclude that even such a large increase in TRAP is not sufficient to prevent the progression of lipid peroxidation and oxidative stress.  相似文献

Drug entry into the body of a helminth is a key factor in the efficacy of anthelmintics. The present project was designed to study the ex vivo uptake and efflux of the benzimidazole anthelmintic flubendazole (FLU) in four strains of H. contortus: the ISE strain (fully susceptible to anthelmintics), the ISE-S strain (resistant to ivermectin), the BR strain (resistant to benzimidazoles) and the WR strain (multi-resistant). The transport of FLU between dead and living nematodes was also compared as well as the effect of verapamil, an inhibitor of the main efflux ABCB1 transporter (P-glycoprotein), on FLU accumulation in nematodes. The obtained results showed that FLU is able to effectively enter H. contortus adults due to high FLU lipophilicity. Passive diffusion is probably the only mechanism in both FLU import and efflux from nematodes. No differences in FLU transport were found among four H. contortus strains with different sensitivity to anthelmintics. No active FLU efflux from H. contortus and no effect of verapamil were observed, indicating that H. contortus cannot protect itself against FLU by the active removal of this anthelmintic from its body.  相似文献

Adenosine is a well described anti-inflammatory modulator of immune responses. The aim of the present study was to describe the role of common adenosine agonist 5'-N-ethylcarboxamidoadenosine (NECA) in cytokine production by main porcine T cell subpopulations. TNF-α, IFN-γ, IL-2 and IL-10 were detected by multicolor flow cytometry together with cell surface markers CD3, CD4 and CD8. It was found that NECA inhibits (in a dose-dependent manner) production of pro-inflammatory TNF-α and Th1-associated cytokines IFN-γ, IL-2 in all concanavalin A-stimulated T cell subpopulations. Moreover, production of IL-10 was potentiated in all T cell subpopulations tested. These corresponded well with the fact that all T cell subsets expressed mRNA for adenosine receptor (AR) subtypes to comparable extents. Contrary to concanavalin A-stimulated cells, NECA had a moderate effect on PMA-stimulated T cells, suggesting that AR in pigs acts via signaling pathways not associated with protein-kinase C. Non-selective antagonist CGS15943 as well as allosteric modulator SCH202676 failed to reverse the effect of NECA in pigs. In conclusion, NECA has an anti-inflammatory effect on porcine T cell subpopulations.  相似文献

Lymphocyte subsets isolated from germ-free piglets experimentally infected with swine influenza virus (SIV), porcine reproductive and respiratory syndrome virus (PRRSV) or porcine circovirus type 2 (PCV2) were studied and the profile of these subsets among these three infections was monitored. Germ-free piglets were used since their response could be directly correlated to the viral infection. Because SIV infections are resolved even by colostrum-deprived neonates whereas PRRSV and PCV2 infections are not, SIV was used as a benchmark for an effectively resolved viral infection. PRRSV caused a large increase in the proportion of lymphocytes at the site of infection and rapid differentiation of B cells leading to a high level of Ig-producing cells but a severe reduction in CD2CD21+ primed B cells. Unlike SIV and PCV2, PRRSV also caused an increase in terminally differentiated subset of CD2+CD8α+ γδ cells and polyclonal expansion of major Vβ families suggesting that non-specific helper T cells drive swift B cell activation. Distinct from infections with SIV and PRRSV, PCV2 infection led to the: (a) prevalence of MHC-II+ T cytotoxic cells, (b) restriction of the T helper compartment in the respiratory tract, (c) generation of a high proportion of FoxP3+ T cells in the blood and (d) selective expansion of IgA and IgE suggesting this virus elicits a mucosal immune response. Our findings suggest that PRRSV and PCV2 may negatively modulate the host immune system by different mechanisms which may explain their persistence.
