Homogenates of HeLa cells contain neuraminidase activity. This enzyme is particle-bound, and it has a pH optimum of 4.2. Hydrocortisone-regulated cells contain two to three times as much neuraminidase as the corresponding controls. The hydrocortisone treatment also causes an increase in the cell content of beta-glucuronidase and acid deoxyribonuclease.  相似文献

Nonresonant manipulation of nuclear spins can probe large volumes of sample situated in inhomogeneous fields outside a magnet, a geometry suitable for mobile sensors for the inspection of roads, buildings, and geological formations. However, the interference by Earth's magnetic field causes rapid decay of the signal within a few milliseconds for protons and is detrimental to this method. Here we describe a technique to suppress the effects of Earth's field by using adiabatic rotations and sudden switching of the applied fields. We observed hundreds of spin echo signals lasting for more than 600 milliseconds and accurately measured the relaxation times of a liquid sample.  相似文献

Individuals are predicted to behave more altruistically and less competitively toward their relatives, because they share a relatively high proportion of their genes (e.g., one-half for siblings and one-eighth for cousins). Consequently, by helping a relative reproduce, an individual passes its genes to the next generation, increasing their Darwinian fitness. This idea, termed kin selection, has been applied to a wide range of phenomena in systems ranging from replicating molecules to humans. Nevertheless, competition between relatives can reduce, and even totally negate, the kin-selected benefits of altruism toward relatives. Recent theoretical work has clarified the processes and selective forces underlying this effect and has demonstrated the generality of the effect of competition between relatives.  相似文献

Charcoal and sodium nitrate, the active ingredients in pesticide gas cartridges, are quantified via carbon analysis and ion chromatography, respectively. Linearity was excellent (R(2) > 0.995) over a range consisting of 50-150% of the target concentration for both ingredients. The coefficient of variation for the replicate analyses of gas cartridges over multiple days was <4% for both analytes. Using the results from the analysis of two batches of gas cartridges, theoretical populations were modeled and used to determine practical sampling strategies to support a quality control program for a gas cartridge manufacturing operation. This modeling indicates that the analysis of three cartridges from each of five different lots would produce mean values for both active ingredients that are within 5% of the true mean >99% of the time.  相似文献

Understanding the impacts of manure amendments on soil microorganisms can provide valuable insight into nutrient availability and potential crop and environmental effects. Soil microbial community characteristics, including microbial populations and activity, substrate utilization (SU) profiles, and fatty acid methyl ester (FAME) profiles, were compared in three soils amended or not amended with dairy or swine manure at two temperatures (18 and 25°C) and two soil water regimes (constant and fluctuating) in laboratory incubation assays. Soil type was the dominant factor determining microbial community characteristics, resulting in distinct differences among all three soil types and some differing effects of manure amendments. Both dairy and swine manures generally increased bacterial populations, substrate diversity, and FAME biomarkers for gram-negative organisms in all soils. Microbial activity was increased by both manures in an Illinois soil but only by dairy manure in two Maine soils. Dairy manure had greater effects than swine manure on SU and FAME parameters such as increased activity, utilization of carbohydrates and amino acids, substrate richness and diversity, and fungal FAME biomarkers. Temperature and water regime effects were relatively minor compared with soil type and amendment, but both significantly affected some microbial responses to manure amendments. Overall, microbial characteristics were more highly correlated with soil physical factors and soil and amendment C content than with N levels. These results indicate the importance of soil type, developmental history, and environmental factors on microbial community characteristics, which may effect nutrient availability from manure amendments and should be considered in amendment evaluations.Mention of trade names or commercial products in this article is solely for the purpose of providing specific information and does not imply recommendation or endorsement by the US Department of Agriculture  相似文献

Limb‐sparing for distal radial osteosarcoma has a high rate of complications. Using personalized three‐dimensional (3D)‐printed implants might improve outcome. The goals of this study were to optimize use of patient‐specific, 3D‐printed endoprostheses for limb‐sparing in dogs in the clinical environment and to report the outcome. This was a pilot study where five client‐owned dogs were enrolled. Computed tomography (CT) of the thoracic limbs was performed, which was used to create patient‐specific endoprostheses and cutting guides, and repeated on the day of surgery. Intra‐arterial (IA) carboplatin was introduced in the clinical management. Limb‐sparing was performed. Outcome measures were time required to produce the endoprosthesis and cutting guide, fit between cutting guide and endoprosthesis with host bones, gait analysis, size of the tumour, percent tumour necrosis, complications, disease‐free interval (DFI) and survival time (ST). Four dogs received IA carboplatin. Excessive tumour growth between planning CT and surgery did not occur in any dog. The interval between the CT and surgery ranged from 14 to 70 days. Fit between the cutting‐guide and endoprosthesis with the host bones was good to excellent. At least one complication occurred in all dogs. Two dogs were euthanized with STs of 192 and 531 days. The other dogs were alive with a follow up of 534 to 575 days. IA chemotherapy is a promising strategy to minimize the risk of excessive tumour growth while waiting for the endoprosthesis and cutting‐guide to be made. The design of the cutting‐guide was critical for best fit of the endoprosthesis with host bones.  相似文献
