The development of sustainable methods for sea urchin juvenile production is currently constrained by high mortality rates during larval growth and the high costs of larval rearing systems management. With the aim of developing a method for the production of juveniles of the purple sea urchin Paracentrotus lividus in a medium‐scale recirculating system, the present study focused on the effects of high stocking densities on larval growth. Plutei larvae were reared at three different densities (up to 7 ind/ml) in a semi‐static culture system. Larval survival and metamorphosis success were evaluated in order to identify the most effective density range. The highest metamorphosis rates (80%–95%) were obtained at 4 and 7 larvae/ml. These results are comparable with (and in some cases higher than) those reported for the same species at much lower larval densities. In conclusion, the rearing conditions tested here show for the first time that a significantly higher (4 ind/ml) stocking density than those of traditional P. lividus rearing methods (based on large volumes and low densities) can be adopted, thus supporting the feasibility of an increase in the final output of competent larvae with no increase in rearing volumes. 相似文献

Exotic trees are often planted to recover degraded lands. Inoculation with mycorrhizal fungi can improve their survival. Plant growth is partly dependent on the strain used, but little attention has been paid to the selection of mycorrhizal fungi. The aim of this study was to determine whether the growth of Casuarina equisetifolia L. (Johnson) is affected by two different mycorrhizal inocula generated using fungal spores retrieved from an Australian site (allochthonous soil) and a Senegalese site (native soil) under C. equisetifolia trees. Comparative experiments were conducted with plants in a Senegalese soil, previously sterilized or not, and grown in a greenhouse. At harvest, parameters related to plant growth and mycorrhization were evaluated and soil bacterial communities were compared. Tree growth was significantly influenced by both types of inoculants. In unsterilized soil, plants inoculated with the native inoculant were taller than plants inoculated with the allochthonous inoculant and control plants. The frequency of mycorrhization with both inoculants was higher in unsterilized soil. The strongest effects of the mycorhizosphere on the soil microbiome were obtained with the allochthonous inoculum, and analysis of the taxonomic composition revealed mycorrhizal communities specific to each inoculum. These results suggest that the development of C. equisetifolia and its root bacterial community are dependent on the composition of the mycorrhizal inoculum. The functional consequences of this rhizosphere effect in terms of soil fertility should be further studied to better guide reforestation operations. 相似文献

Information on the effects of distance from the seashore on soil biotic communities is limited and restricted to soil arthropods and microarthropods. The aim of this study, conducted in a sand-dune area, was to distinguish the changes of a ground-active arthropod community along a 4-km-transect from the seashore going inland. Pitfall traps were set up in open spaces at four locations (L1, through L2, L3, and L4 going inland) through the year. The abiotic gradient from the seashore going inland was found to be mostly dependent on seasonality. Group richness was found to be remarkably greater at the L2, L3, and L4 locations compared with the L1 location in winter, and at the L2 and L3 locations compared with the L1 location in spring. The Fisher α index was 0.32?±?0.21, 3.51?±?0.87, 5.04?±?0.70, and 4.86?±?0.54 in winter and 3.29?±?0.55, 13.79?±?1.49, 10.12?±?1.24, and 5.06?±?0.62 in spring at the L1, L2, L3, and L4 locations, respectively. Regarding abundance and group richness, the trophic relationships between predators, phytophages, omnivores, and saprophages were remarkably affected by both location and seasonality. The dominant groups and dominant index distribution of ground-active arthropod communities were found to follow a similar pattern. Soil organic carbon content was found to be the key factor driving the ground-active arthropod community structure along a coastal gradient going inland in winter and spring. We conclude that the distribution, trophic composition, and diversity index are affected by both location and seasonality, while total abundance is consistently affected by location across all three seasons. 相似文献

Sperm quality tests on fish are classically used for evaluating cryopreservation procedures, and they are also promising to assess aquatic toxicity and biomarkers of xenobiotic effects on reproduction. Osmotic shock from the storage medium is one of the main factors affecting sperm quality during evaluation. Thus, the objective of this study was to evaluate the effects of different osmolalities (240–460 mOsm/kg) for at least 4 days on the sperm quality parameters of the viviparous fish Jenynsia multidentata. The level of significance was (P < 0.05). The plasma osmolality of J. multidentata is 326 ± 3.9 mOsm/kg. The motility of fresh semen was higher in osmolalities of 280 and 300 mOsm/kg but did not differ between osmolalities from 240 to 320 mOsm/kg. Above 380 mOsm/kg, the motility observed was 0 %. Over the time period studied motility increased with increasing osmolality, and the most constant and long-lasting rates were between 300 and 320 mOsm/kg. On the 4th day of evaluation, higher membrane integrity rates were observed between 280 and 360 mOsm/kg, higher mitochondrial membrane potential was observed between 300 and 460 mOsm/kg, and higher DNA integrity rates were observed between 260 and 380 mOsm/kg. Moreover, osmolalities ≥460 and ≤240 resulted in the lowest motility and DNA integrity levels. Over 4 days, the plasma membrane integrity was significantly lower at ≤260 and ≥400 mOsm/kg, and the mitochondrial membrane potential was significantly lower only in osmolalities ≤240 mOsm/kg. Therefore, we conclude that for sperm quality preservation in J. multidentata, an osmolality of 300–320 mOsm/kg of the most suitable diluent is necessary. Furthermore, we conclude that the storage of sperm in a hyposmotic (<260 mOsm/kg) or hyperosmotic (>400 mOsm/kg) solution affects not only motility but also other sperm quality parameters. 相似文献

No-tillage systems lead to physical, chemical and biological changes in soil. Soil fertility is responsive to changes in tillage as it depends on nutrient status, soil water content and biological characteristics. This work aimed to determine long term changes in phosphorus forms and availability in the profile of two tropical soils under conventional and no-till systems, and to discuss the significance of these changes on plant growth and demand for P fertilizers. Undisturbed soil cores with 20 cm in diameter were collected to a depth of 40 cm, accommodated in PVC tubes and taken to a greenhouse, where the experiment was conducted. Two soils were collected in Central Brazil, in areas under Cerrado. Both soils had been cropped for at least 10 years under conventional tillage and no-till. In the greenhouse, pots received phosphorus fertilization or not at 43.7 kg ha−1, and soybean was grown for 60 days, when soil P fractions were determined. Labile P fractions in the soil profile were not affected by management systems, and there was no accumulation of available P under no-till. A large amount of P added as fertilizer was adsorbed in soil and remained in moderately labile fractions, mainly on uppermost soil layers. Therefore, the phosphate fertilizer has promoted P accumulation on less available fractions in soil, remaining P on the soil after crop harvest. Interferon-gamma (IFN-gamma) detection assay is being applied as an ancillary test to tuberculin tests in the diagnosis of bovine tuberculosis to detect the maximum number of infected animals. Among possible factors influencing the performance of tuberculosis-diagnostic tests, paratuberculosis, a widespread disease in Spain and other European countries, has been pointed out as a cause of false positive reactions. Still, its effect on the sensitivity of these tests in cattle has yet to be fully characterized. The impact of paratuberculosis in the apparent sensitivity of IFN-gamma assay was studied in a bullfighting cattle herd with a mixed tuberculosis-paratuberculosis infection, using culture of Mycobacterium bovis and Mycobacterium avium paratuberculosis as the gold standard to determine the infection status of every animal. A total of 218 animals were slaughtered and sampled for bacteriology after blood sampling. IFN-gamma assay showed a lower apparent sensitivity in animals with a mixed infection (50%) compared to all animals suffering tuberculosis (78.3%). This finding indicates that the presence of paratuberculosis in tuberculosis-infected herds could imply a serious impairment in the sensitivity of IFN-gamma detection test. 相似文献
