The mechanisms involved in prion neurotoxicity are unclear, and therapies preventing accumulation of PrPSc, the disease-associated form of prion protein (PrP), do not significantly prolong survival in mice with central nervous system prion infection. We found that depleting endogenous neuronal PrPc in mice with established neuroinvasive prion infection reversed early spongiform change and prevented neuronal loss and progression to clinical disease. This occurred despite the accumulation of extraneuronal PrPSc to levels seen in terminally ill wild-type animals. Thus, the propagation of nonneuronal PrPSc is not pathogenic, but arresting the continued conversion of PrPc to PrPSc within neurons during scrapie infection prevents prion neurotoxicity.  相似文献

Several authors have suggested that edible plants could avoid herbivory by mimicking olfactory cues of toxic plants. However, very few studies have been carried out to test this hypothesis. The aims of the present study were to identify the volatiles of three clover species and to test whether a species lacking chemical defences, such as red clover, could avoid being grazed by rabbits by mimicking the volatiles of the cyanogenic white clover. Two main volatiles were identified in all three clover species, and a further two volatiles were present in white clover only. Rabbits presented with a choice between white clover, red clover and red clover sprayed with white clover extract ate significantly more red clover than white or white-flavoured red clover. The results suggest that the volatiles of toxic plants could be used and exploited as a source of natural, safe and effective repellents to control the impact of pest herbivores on plants.  相似文献

Free radicals in olive oils were identified and quantified by EPR, by means of the spin-trapping technique making use of alpha-phenylbutylnitrone (PBN) as spin trap. The radical species were identified as PBN-trapped hydroxyl radicals (PBN-*OH) in the water microdroplets inside the fat medium. The largest radical concentration was 12.5 microM identical with 100%. The following were the relative concentrations of the radicals under different conditions: (1) Two oils, produced by continuous centrifugation, aged for 1 year, showed a 25-30% increase in the radicals compared to nonaged oils; 1-year-old oil, produced by pressure, did not differ from the nonaged oil. (2) Radical production was markedly reduced by N(2) bubbling; it was increased by heating, whereas it showed a biphasic pattern by air bubbling over time. (3) Radical concentration as a function of the UV irradiation time increased up to a maximum, after which it decreased and finally remained constant. The phenolic and oxygen contents were related to the radical content. This study demonstrates that the EPR technique is suitably applied to the detection of free radicals in olive oil and that storage, handling, and stress conditions of the oils significantly influence the radical concentration.  相似文献

The aim of this research was to validate the results obtained previously by purge and trap (PT) and to investigate the ability of solid phase microextraction (SPME), a more rapid and less expensive technique, to discriminate drinking milk subjected to different heat treatments (i.e., pasteurization, ultrahigh temperature, "in-bottle" sterilization) and produced at different factories. The data obtained by both methods were processed by multivariate statistical analysis. PT and SPME showed comparable repeatability, although with different performances for the yield of extraction, and allowed the three milk categories to be distinguished. Within the chemical class of methyl ketones, 2-heptanone was found to be the most discriminating compound, and the possibility of using the concentration of this volatile as a marker for heat treatment was investigated.  相似文献

Southern populations of the European minnow fromremote oligotrophic mountain lakes along an Alpinenorth-south transect accumulated more p,p DDE andPCBs than northern populations. As these semi-volatileorganochlorines predominantly evaporate in warmcountries, higher rates of condensation (deposition)are assumed to occur in the southern slopes of theAlps. The higher accumulation of lead and cadmium insouthern population is rather attributed to geogenicand specific environmental impacts than to atmosphericdeposition. Increasing liver ratios of [glutathionedisulfide]/[glutathione], a potential indicator foroxidative stress, from north to south reflect thegenerally higher toxic load at remote sites at thesouthern edge of the Alps. However, histopathologicalchanges in the liver did not correlate withaccumulated toxicants indicating that deposition doesnot lead to severe lesions but induces specificmechanisms for detoxification.  相似文献

Anomalies involving arterial branches in the lungs are one of the causes of hemoptysis in humans and dogs. Congenital and acquired patterns of bronchoesophageal artery hypertrophy have been reported in humans based on CT characteristics. The purpose of this retrospective study was to describe clinical, echocardiographic, and multidetector computed tomography features of bronchoesophageal artery hypertrophy and systemic‐to‐pulmonary arterial communications in a sample of 14 dogs. Two main vascular patterns were identified in dogs that resembled congenital and acquired conditions reported in humans. Pattern 1 appeared as an aberrant origin of the right bronchoesophageal artery, normal origin of the left one, and enlargement of both the bronchial and esophageal branches that formed a dense network terminating in a pulmonary artery through an orifice. Pattern 2 appeared as a normal origin of both right and left bronchoesophageal arteries, with an enlarged and tortuous course along the bronchi to the periphery of the lung, where they communicated with subsegmental pulmonary arteries. Dogs having Pattern 1 also had paraesophageal and esophageal varices, with the latter being confirmed by videoendoscopy examination. Authors conclude that dogs with Pattern 1 should be differentiated from dogs with other congenital vascular systemic‐to‐pulmonary connections. Dogs having Pattern 2 should be evaluated for underlying pleural or pulmonary diseases. Bronchoesophageal artery hypertrophy can be accompanied by esophageal venous engorgement and should be included in the differential diagnosis for esophageal and paraesophageal varices in dogs.  相似文献

Cytokinin activity was detected by means of the soybean callus test in the scales of Iris hollandica Hoog cultivar ‘Prof. Blaauw’ during the cold treatment for flower formation. A component of the biological activity extracted from the bulb scales coeluted with zeatin riboside when subjected to high-performance liquid chromatography. Fusarium oxysporum is a ubiquitous soilborne ascomycete responsible for vascular wilt in many plant species worldwide. This species comprises more than 120 putative host-specific formae speciales capable of causing marked economic losses. In summer 2009, wilt symptoms, including chlorosis and poor development of the root system, were observed on cultivars of chicory (Cichorium intybus) in northern Italy. The causal agent isolated from symptomatic tissues in this case was identified as F. oxysporum on the basis of both morphological features and molecular analyses. In this work, we attempted to characterize the isolates of F. oxysporum from C. intybus by both biological and molecular approaches. Pathogenicity trials performed on five species of the Asteraceae family with isolates of F. oxysporum from C. intybus indicated that the pathogen has a unique host range, infecting chicory only. Neither lettuce nor endive, lawn daisy or Paris daisy developed the disease. Five cultivars within C. intybus species were tested, and the cv. ??Clio?? was the most susceptible. Phylogenetic analyses relative to the ribosomal intergenic spacer (IGS) and translation elongation factor 1-alpha (EF1-??) assigned isolates pathogenic to chicory to a single cluster, distinct from other pathogenic F. oxysporum. In light of these findings, we propose to designate this organism as Fusarium oxysporum f.sp. cichorii.  相似文献

Oxylipins are a large and diverse family of fatty acid derivatives exhibiting different levels of oxidation of the carbon chain. They are involved in many biological functions in mammals, plants and diatoms. In this last group of organisms, they are suggested to play a role in the reproductive failure of copepod predators, showing clear pro-apoptotic effects on newborn nauplii. In this work, these compounds were tested for the ability to induce mitotic arrest in sea urchin embryos. We show for the first time that oxylipins have an increased efficacy in their corresponding methylated form. Natural oxylipins were also used as an inspiration for the rational design and synthesis of stable chemical analogs with apoptotic activity against tumor cell lines. This approach led to the synthesis of the linear C15-ketol (22) that was shown to induce apoptosis in human leukemia U-937 cells. These results are proof of the concept of the use of eco-physiological considerations as a platform to guide the search for novel drug candidates.  相似文献
