The potential of apple pomace for lactic acid production by simultaneous saccharification and fermentation (SSF) was evaluated. The effects of the cellulase to solid ratio (CSR), the liquor to solid ratio (LSR), and the beta-glucosidase to cellulase ratio (BCR) on the kinetics of lactic acid generation were assessed, and a set of mathematical models was developed to reproduce and predict the lactic acid concentration of fermentation broths. Operating at low cellulase and cellobiase charges (1 FPU/g and 0.25 IU/FPU, respectively) and short reaction times (10 h), SSF media containing 27.8 g of lactic acid/L were obtained with a volumetric productivity of 2.78 g/Lh. Material balances showed that the SSF processing of 100 kg of dry apple pomace results in the production of 36.6 kg of lactic acid, 18.3 kg of oligomeric carbohydrates (which can be used as ingredients for functional foods), 8.4 kg of microbial biomass, and 8 kg uronic acids. 相似文献

This work analyzes the direct effect of soil management practices on soil microbial communities, which may affect soil productivity and sustainability. The experimental design consisted of two tillage treatments: reduced tillage (RT) and zero tillage (ZT), and three crop rotation treatments: continuous soybean (SS), corn–soybean (CS), and soybean–corn (SC). Soil samples were taken at soybean planting and harvest. The following quantifications were performed: soil microbial populations by soil dilution plate technique on selective and semi-selective culture media; microbial respiration and microbial biomass by chloroform fumigation-extraction; microbial activity by fluorescein diacetate hydrolysis; and fatty acid methyl ester (FAME) profiles. Soil chemical parameters were also quantified. Soil organic matter content was significantly lower in RT and SS sequence crops, whereas soil pH and total N were significantly higher in CS and SC sequence crops. Trichoderma and Gliocladium populations were lower under RTSS and ZTSS treatments. Except in a few cases, soil microbial respiration, biomass and activity were higher under zero tillage than under reduced tillage, both at planting and harvest sampling times. Multivariate analyses of FAMEs clearly separated both RT and ZT management practices at each sampling time; however, separation of sequence crops was less evident. In our experiments ZT treatment had highest proportion of 10Me 16:0, an actinomycetes biomarker, and 16:1ω9 and 18:1ω7, two fatty acids associated with organic matter content and substrate availability. In contrast, RT treatment had highest content of branched biomarkers (i15:0 and i16:0) and of cy19:0, fatty acids associated with cell stasis and/or stress. As cultural practices can influence soil microbial populations, it is important to analyze the effect that they produce on biological parameters, with the aim of conserving soil richness over time. Cytogenetic analyses of the river puffer, Takifugu obscurus, the tiger puffer, Takifugu rubripes and hybrids produced between female T. obscurus and male T. rubripes and their hybrid triploids (produced by cold shock treatment at 4°C) were performed. T. obscurus had 2n = 44 chromosomes and 1.84 ± 0.019 pg DNA/cell, T. rubripes had 2n = 44 and 2.64 ± 0.015, the hybrids had 2n = 44 and 2.15 ± 0.010 and the hybrid triploids had 3n = 66 and 3.22 ± 0.010. The erythrocyte values of the hybrids were more similar to those for T. obscurus, whereas the hepatocyte, midgut and proximal tubule kidney cell values of the hybrids fell down between those for the parental species (p < .05). The erythrocyte, proximal tubule, hepatocyte and midgut epithelial cell sizes for the hybrid triploids were 1.5‐fold larger than those for the hybrids (p < .05). The thickness of retina and each layer in the hybrid triploids were 1.1‐fold larger than those of the hybrids (p < .05) and did not differ significantly among T. obscurus, T. rubripes and the hybrids (p > .05); however, the hybrid triploids had fewer cell nucleus outer layers than the hybrids (p < .05). Gonad development in the hybrids and hybrid triploids was less matured than in T. obscurus and T. rubripes. The metaphase nucleolus organizer regions (NORs) and the gill cells of T. obscurus, T. rubripes and the hybrids contained two satellite telocentrics, whereas the hybrid triploids contained three satellite telocentrics. 相似文献

We have prepared a series of polypropylene/exfoliated graphene (PP/EG) nanocomposite films via efficient meltcompounding and
compression, and investigated their morphology, structures, thermal transition behavior, thermal stability, electrical and
mechanical properties as a function of EG content. For the purpose, EG, which is composed of disordered graphene platelets
as reinforcing nanoscale fillers, is prepared by the oxidation/exfoliation process of natural graphite flakes. SEM images
and X-ray diffraction data confirm that the graphene platelets of EG are well dispersed in PP matrix for the nanocomposites
with EG contents less than 1.0 wt%. It is found that thermo-oxidative degradation of PP/EG nanocomposites is noticeably retarded
with the increasing of EG content. Electrical resistivity of the nanocomposite films was dramatically changed from ∼1016 to ∼106 Ω·cm by forming electrical percolation threshold at an certain EG content between 1 and 3 wt%. Tensile drawing experiments
demonstrate that yielding strength and initial modulus of PP/EG nanocomposite films are highly improved with the increment

Objective The goal of this work is to study the dark adaptation curve of the normal horse electroretinogram (ERG). Procedures The electroretinographic responses were recorded from six healthy female ponies using a contact lens electrode and a mini‐Ganzfeld electroretinographic unit. The horses were sedated intravenously with detomidine, an auriculopalpebral nerve block was then performed, and the pupil was fully dilated. The ERG was recorded in response to a low intensity light stimulus (30 mcd.s/m2) that was given at times (T) T = 5, 10, 15, 20, 25, 30, 40, 50, and 60 min of dark adaptation. Off‐line analysis of the ERG was then performed. Results Mean b‐wave amplitude of the full‐field ERG increased continuously from 5 to 25 min of dark adaptation. The b‐wave amplitude peaked at T = 25, however, there was no statistical significance between T = 20 and T = 25. The b‐wave amplitude then remained elevated with no significant changes until the end of the study at T = 60 (P > 0.49). The b‐wave implicit time increased continuously between T = 5 and T = 20, then gradually decreased until T = 60. No distinct a‐wave was observed during the testing time. Conclusions Evaluation of horse rod function or combined rod/cone function by means of full‐field ERG should be performed after a minimum 20 min of dark adaptation. 相似文献

In 2000, one-year-old seedlings of pyrenean oak (Quercus pyrenaica Willd.) and sessile oak (Quercus petraea [Matt.] Liebl) were planted in a thinned and an unthinned plot in a pinewood (Pinus sylvestris), and in a nearby clearing. In summer 2002 and 2003, water relations and gas exchange parameters were measured to address the impact of drought on the seedlings. Chlorophyll a fluorescence was also measured to explore leaf photochemistry and a possible non-stomatal limitation to photosynthesis (A). Reduction in stomatal conductance (g) in response to the decrease of predawn water potential (Ψpd) resulted the main cause affecting net carbon uptake. Water potential at midday (Ψmd) was similar in both species but Quercus petraea was more sensitive to soil water deployment occurred along summer, showing slightly lower Ψpd because worse recover of water potential during night. Rate of photosynthesis was higher in Q.␣pyrenaica probably in relation to its greater leaf mass per area (LMA) and nitrogen content per leaf area (Na). Mortality was highest in the clearing and lowest in the thinned pinewood. Throughout the summer, soil moisture was higher in the thinned area, possibly because of the reduction in tree transpiring surface and interception of rainfall. As a tool to address selected issues of virus biology, we constructed a recombinant cDNA clone of bovine viral diarrhea virus (BVDV) expressing Gaussia luciferase (Gluc) reporter gene. A full-length genomic cDNA clone of a non-cytopathic BVDV isolate was assembled by recombination in yeast Saccharomyces cerevisiae. The Gluc gene was inserted between the Npro and Core protein coding regions by recombination. The cDNA transcribed in vitro was infectious upon transfection of MDBK cells, resulting in reporter gene expression and productive virus replication. The rescued viruses were stable for 15 passages in cell culture, maintaining the replication kinetics, focus size and morphology similar to those of the parental virus. Expression and correct processing of the reporter protein were also maintained, as demonstrated by Gluc activity. These results demonstrate that genes up to 555 bp are simply assembled by a single step in yeast recombination and are stably expressed by this cDNA clone. 相似文献

- Bivalves are important components of freshwater ecosystems; however, they are also one of the most threatened animal groups, especially members of the order Unionida. The main threats to freshwater bivalves are habitat modification and invasive species. Protected areas are a common way to minimize impacts and preserve native species, but they are rarely designated with a focus on freshwater invertebrates.
- The main goal of this article was to describe the distribution of freshwater bivalves in the state of Rio de Janeiro, and the relationship between these species and bioclimatic areas, land use and protected areas.
- Five native and two invasive bivalve species have been reported in Rio de Janeiro. They occur mainly in bioclimatic areas related to the Paraíba do Sul River and Campista Lowlands. The few records of bivalves inside protected areas are in areas of sustainable use that offer a lower level of protection, and frequently in sympatry with invasive bivalves.
- Thus, the established protected areas in Rio de Janeiro are not adequately effective for freshwater bivalve conservation, and some species remain under threat even within them. Freshwater bivalves thus remain largely unprotected in the state.
- The development of new protected areas and management plans should consider other faunal groups that are usually ignored, such as freshwater bivalves, to achieve more inclusive and effective protection.
