In this study, different types of tissue sampling for PCR-based diagnosis and follow-up of canine visceral leishmaniosis were compared. Skin, whole blood and lymph node samples were collected from 95 naturally infected dogs living in South Italy, where the disease is endemic. Twenty-nine of these 95 dogs, treated with meglumine administered concurrently with allopurinol for 30 days, and then with allopurinol alone, were monitored during a period of 2 years. The DNA extracted from the clinical specimens was amplified by PCR using as target DNA a 116-bp fragment in the constant region of the kinetoplast DNA minicircle. PCR analysis was more sensitive than indirect immunofluorescence antibody test in detecting Leishmania infection in symptomatic dogs: 99% of lymph node samples resulted positive, whereas 94% of blood samples and 95% of skin samples gave a positive result. PCR analysis of samples from dogs followed up 2 years showed that: (1) all subjects resulted positive in at least one of the three types of samples; (2) all time the dogs had a relapse, PCR resulted positive in all three types of samples; (3) when dogs were apparently healthy, PCR analysis was positive on skin and lymph node samples, but not always on blood samples. Since lymph node sampling is invasive and sometimes difficult in healthy asymptomatic dogs, our results suggest that, independently from the presence or not of cutaneous lesions, skin biopsy represents a good substratum for PCR-based diagnosis and follow-up of canine visceral leishmaniosis.  相似文献

Alpine bluegrass (Poa alpina L.) is common in mountain areas, where it represents a valuable forage resource and is widely used for ecological restoration of disturbed landscapes. This study assessed the variation of germplasm from the Rhaetian Alps, Italy. Both seed production and morphology were examined for potential use in restoration projects and for conservation and selection purposes of local genetic resources. The effects of the ex situ growing site were also assessed in two altitude‐contrasting sites. Individual populations harboured great phenotypic diversity for all traits, including seed yield, while variation among valleys of origin was less manifest. Noticeable site‐specific population yield responses were observed, suggesting that the environment adopted for seed production may steer the choice of materials to be multiplied. The longer vegetative period in the lowland site, combined with environment‐tailored agronomic practices, contributed to better plant growth, seed yield and survival compared to the mountain site, suggesting that seed production of this Alpine species could be feasible in a lowland environment when adequate inputs were provided. One valley (Valchiavenna) displayed high average levels of seed yield and plant vigour across growing sites, indicating its possible value as a source of germplasm suitable for resowing at high altitude.  相似文献

The diversity of nutritional and culinary traits in 21 populations of common bean from the Basilicata region was studied for three consecutive harvests (1995‐97). The results were compared with the six commercial cultivars widely grown in the region. Some populations deserve particular attention since they had large seeds, high protein content, low trypsin inhibitor levels and short cooking time. The cluster analysis showed that 18 populations clustered in two main groups and that the bush populations were clearly distinguishable from the climbing ones. When phaseolin type and some plant morphological traits were added to the nutritional and culinary traits to classify these populations, it was observed that they resembled the race Peru (Andean gene pool).  相似文献

Knowledge of the kinetics and pathways of hydrolytic degradation is crucial to the prediction of the fate and transport mechanism of chemicals. This work first describes the kinetics of the chemical hydrolysis of imazosulfuron, a new sulfonylurea herbicide, and evaluates the results to propose a degradation pathway. The hydrolysis of imazosulfuron has been studied in aqueous buffers both within the pH range 1.9-12.3 at ambient temperature (thermostated at 25 +/- 2 degrees C) and at pH 3.6 within the temperature range of 15-55 degrees C. The hydrolysis rate of imazosulfuron was characterized by a first-order kinetics, pH- and temperature-dependent, and accelerated by acidic conditions and higher temperatures. The calculated half-lives at pH 4.5 and 5.9 were 36.5 and 578 days, respectively. At pH 6.6, 7.4, 9.2, and 12.3 no significant change in imazosulfuron concentration was observed after 150 days. Half-lives were much lower at pH <4 (= imazosulfuron pK(a)), at which they ranged from 3.3 to 6.3 days. Moreover, a change in temperature from 15 to 25 degrees C in acidic conditions (pH 3.6) decreased the half-life of imazosulfuron by a factor of approximately 4.0; in any case, a 3-5-fold increase in the rate of hydrolysis was found for each 10 degrees C increase in temperature. In acidic conditions the only hydrolysis products were the two molecules resulting from the cleavage of the sulfonylurea bridge.  相似文献

Blueberries ( Vaccinium corymbosum L.) are rich sources of phenolics and anthocyanins (ACNs). We investigated the absorption of ACNs after consumption of one portion (300 g) of minimally processed blueberry purée (P) obtained from blanched (BL) or unblanched (NB) berries. A repeated-measures, crossover design study was conducted on healthy human volunteers. Blood was drawn between baseline and 24 h after BL-P or NB-P consumption, while urine were collected from the day before the experiment up to 48 h. Total plasma ACN content was not significantly different, while phenolics content was higher in BL-P with respect to NB-P. The maximum ACN absorption in plasma was observed after 1.5 h from the intake of the purées and was significantly higher (p ≤ 0.05) after the intake of BL-P. Both products increased the excretion of hippuric acid in urine. In conclusion, blanching had no significant effect on total ACN content and enhanced their absorption from minimally processed purées.  相似文献

The objective of the present work was to determine the effect of Lactobacillus murinus strain LbP2 on canine fecal immunoglobulin A (IgA) levels. Seven dogs were orally treated with a 3-mL suspension of L. murinus LbP2 containing 5 × 109 colony-forming units on alternate days for 2 wk. Six dogs were treated with 3 mL of phosphate-buffered saline as placebo. Fecal samples were taken from the rectal ampulla on days 0 and 16, and the total canine fecal IgA concentration was determined with an immunoperoxidase assay kit. The IgA levels of individual dogs were compared with the nonparametric Wilcoxon test. Differences were considered significant when the P-value was less than 0.05. An increase in the total fecal IgA concentration was observed in the 7 dogs after treatment with L. murinus LbP2 (P = 0.01796). No differences were detected between the initial total fecal IgA values and those obtained at the end of placebo treatment. Thus, after oral administration L. murinus LbP2 showed potential immunomodulatory effects, an important property to assess in a microorganism being considered for use as a probiotic.  相似文献

Phosphorus availability is a major limiting factor for yield of most crop species. The objective of this study was to compare the solubilization of three sources of phosphorus (P) by different fungal isolates and to determine the possible mechanisms involved in the process. Talaromyces flavus (S73), T. flavus var flavus (TM), Talaromyces helicus (L7b) and T. helicus (N24), Penicillium janthinellum (PJ), and Penicillium purpurogenum (POP), fungal strains isolated from the rhizosphere of crops, are known to be biocontrol agents against pathogenic fungi. The P solubilization efficiency of these fungal strains in liquid media supplemented either with tricalcium phosphate (Ca3(PO4)2; PC), aluminum phosphate (AlPO4; AP), or phosphorite (PP) depended on the source of P and the fungal species. The type and concentration of organic acids produced by each species varied according to the source of available P. In the medium supplemented with PC, the highest proportion was that of gluconic acid, whereas in the media supplemented with the other P sources, the highest proportion was that of citric and valeric acids. This suggests that the release of these organic compounds in the rhizosphere by these microorganisms may be important in the solubilization of various inorganic P compounds. Results also support the hypothesis that the simultaneous production of different organic acids by fungi may enhance their potential for solubilizing insoluble phosphate.  相似文献
