Resveratrol, a well-known phytoalexin and antioxidant, is produced by the action of stilbene synthase (STS) in some plant species. Hop (Humulus lupulus L.) plants of the Tettnang variety were transformed with a gene encoding for STS from grapevine. Under the control of the constitutive 35S cauliflower mosaic virus promoter, expression of the transgene resulted in accumulation of resveratrol and high levels of its glycosylated derivatives in leaves and inflorescences. Piceid, the predominant derivative, reached a concentration of up to 560 microg/g of fresh weight (f.w.) in hop cones, whereas no stilbenes were detected in nontransformed controls (wild-type). In transgenic plants the amounts of alpha- and beta-acids, naringenin chalcone, and prenylated flavonoids did not change significantly when compared with nontransformed plants. Transgenic plants showed normal morphology and flower development as did the nontransformed controls. The results clearly show that in hop constitutive expression of sts interferes neither with plant development nor with the biosynthesis of secondary metabolites relevant for the brewing industry. Since resveratrol is a well-known phytoalexin and antioxidant, sts transgenic hop plants could display enhanced pathogen resistance against microbial pathogens, exhibit new beneficial properties for health, and open new venues for metabolic engineering.  相似文献

This study describes the identification of six allyl esters in a garlic cheese preparation and in a commercial cream cheese. The extracts were prepared by liquid/liquid extraction and concentrated by the SAFE process. The identification of the allyl esters of acetic, butyric, hexanoic, heptanoic, octanoic, and decanoic acids is based on the correlation of their mass spectrometric data and chromatographic retention time data obtained from the extracts with those of authentic standards. In addition to the gas chromatography (GC)/mass spectrometry analysis, the flavor ingredients were characterized by GC sniffing by a trained flavorist. OBJECTIVE: To describe perceptions of weight of adolescents and associated factors. DESIGN: Cross-sectional survey in 2002. SETTING: Eight public middle schools in two distinct socio-economic areas of Jiangsu Province, China. SUBJECTS: Eight hundred and twenty-four adolescents (aged 12-14 years) and 628 of their parents. METHODS: Self-administered questionnaires for students and their parents were used to collect the data. Height and weight of the students were measured. RESULTS: One-third of the girls perceived themselves as overweight, while only 8.9% were actually overweight or obese according to the World Health Organization definition. On the other hand, 15.0% of the girls considered themselves underweight, while 5.6% were underweight according to the definition. Among the boys, one-fifth of those defined as overweight perceived their weight as normal. Furthermore, 23.9% perceived themselves as underweight when in fact only 4.9% were classified as underweight according to the definition. About one-quarter of the students dieted in the past year. Adolescents who perceived themselves to be overweight dieted and skipped breakfast more often. They also had lower intake frequencies of animal foods, Western foods and milk/yoghurt. No disparity was observed in the intake frequency of fruits and vegetables between the groups of different weight perception. Parents' weight norms seemed to favour overweight in both genders, but especially among boys. CONCLUSIONS: Both overweight and underweight concerns were common among girls, while a higher body weight was favoured by normal-weight boys and their parents. Distorted weight perceptions in adolescents, as well as dieting, are problems that need appropriate intervention.  相似文献

The passage of a 14C-labeled carbamate, 2-(2-chloro-1-methoxy-' ethoxy)phenyl N-methylcarbamate, or its labeled metabolites through the alimentary system of the grey garden slug Deroceras reticulatum was examined by autoradiographic studies and scintillation counting. It was demonstrated that, in a first step, the molluscicide penetrated the cells of the oesophagus and the crop. It was quickly transported by the hemolymph to the periphery of the body and re-entered the cells of the digestive tract and the mid-gut gland in a second step from the hemolymph side. The crypt cells of the mid-gut gland are discussed as cells involved in detoxification, and connective tissue cells as the major storage sites of the labeled material. Excretion in feces and secretion in mucus are thought to be the routes of 14C elimination.  相似文献

Quality assessment of thermally modified spruce (Picea abies (L.) Karst) and beech (Fagus sylvatica L.) wood and of the corresponding reference samples was carried out by means of non-destructive FT-NIR spectroscopic measurements and PLS regression. Oven-dry and basic density as well as MOE and MOR determined by 3-point bending tests were evaluated. The focus was put on specimens produced from material that had been thermally modified in an industrial scale kiln. Modelling results range from poor to very good. The results of the spectra taken from the spruce samples resulted in better prediction results than the spectra of the beech samples. This could be due to different proveniences or variation in the industrial modification process. The results indicate that FT-NIR surface measurements of sound thermally modified wood samples could be applied to evaluate several characteristics before and after the modification process. The method could be used for screening during pre-sorting of thermally modified wood.  相似文献

为了研究物料下落过程中随机性粉尘的产生特性,在实验室开发并设计了一套自由下落装置.通过实验主要测定空气湿度和质量流量对粉尘产生率的影响.结果表明,空气湿度对粉尘排放有影响,且对粗细粒子的影响程度不同;当空气湿度达到一定数值时,粉尘产生率已很小,此后再增加空气湿度,粉尘产生率基本不变.随着质量流量的增加,粉尘产生率逐渐减小,但粉尘浓度会增加,对工作场所工人的身体健康及环境带来严重危害.根据实验数据建立了空气湿度和质量流量与粉尘产生率之间的数学模型,经验证,相关性很好,实测值与理论计算值基本一致,可以定量评价不同质量流量下不同空气湿度的粉尘产生率.  相似文献

A monitoring study on the current sensitivity situation of wheat powdery mildew (Blumeria graminis f.sp. tritici) towards metrafenone was performed in 2009 with 2509 isolates. Airborne isolates were randomly collected in different regions of the most important European cereal growing regions. The sensitivities of the majority of isolates were comparable to the baseline sensitivity, which was determined in 2000 before market launch of the compound. Ninety-two isolates showed sensitivities outside the baseline. Eighty-four of them were classified as moderately adapted and could still be inhibited with registered rates of metrafenone in glasshouse tests under various preventive or curative conditions. Eight isolates were identified to be resistant, which were not fully inhibited at registered rates. Resistance management strategies and further extensive monitoring studies are indicated for a sustainable use of metrafenone against wheat powdery mildew.  相似文献

Formation of the nitroso derivatives of 14C-labeled carbaryl, carbofuran, and p-chlorophenyl methylcarbamate (PCMC), and their subsequent stabilities were investigated using low levels of carbamate (0.5 μmol) and aqueous HCl concentrations encompassing those considered maximum for the gastric contents of humans (pH 1–2) and of rats (pH 3–4). Reacting the carbamates with excess sodium nitrite for 10 min at 37°C in a pH 1.0 HCl solution gave nitrosocarbamate yields of 42 to 64%, while only trace amounts were formed at pH 2 and above. The nitrosocarbamates were most stable at pH 3–5 with half-lives ranging from 114 to 470 min. Stability of all three nitroso derivatives was considerably less at pH 1.5 (t12, 25–34 min), but at pH 7 the stability varied: nitroso-PCMC t12, 6 min; nitrosocarbofuran t12, 70 min; nitrosocarbaryl t12, 139 min. Denitrosation to the parent carbamate was the predominant degradation pathway at pH 1.5, but at pH 3–7 degradation was primarily by hydrolysis of the carbamate ester linkage. Each of the nitrosocarbamates was directly mutagenic in the S. typhimurium assay system. Since the data show that nitrosation of residue levels of carbamate pesticides occurs readily at pH 1 but not at pH 2 and above, it is critical that gastric contents of any animal model used for assessing nitrosocarbamate formation have a pH approaching 1 as may occur in the human stomach.  相似文献

Vascular endothelial growth factor (VEGF) is a major regulator of angiogenesis and a potential autocrine growth factor for neoplastic cells in various malignancies. In the present study, we have investigated expression of VEGF and VEGF receptors in canine mastocytomas and the canine mastocytoma cell line C2. As assessed by immunostaining of tissue sections and cytospin slides, primary neoplastic mast cells (MC) and C2 cells were found to express the VEGF protein. In Northern blot and RT-PCR experiments, C2 cells expressed VEGF mRNA in a constitutive manner. VEGF mRNA expression in C2 cells was counteracted by LY294002 and rapamycin, suggesting involvement of the PI3-kinase/mTOR pathway. Moreover, C2 cells were found to express VEGF receptor-1 (Flt-1) and VEGF receptor-2 (KDR). However, recombinant VEGF failed to promote (3)H-thymidine uptake in C2 cells, and a neutralizing anti-VEGF antibody (bevacizumab) failed to downregulate spontaneous proliferation in these cells. In addition, rapamycin decreased the expression of VEGF in C2 cells at the mRNA and protein level without suppressing their proliferation. Together, canine mastocytoma cells express VEGF as well as VEGF receptors. However, despite co-expression of VEGF and its receptors, VEGF is not utilized as an autocrine growth regulator by canine mastocytoma cells.  相似文献
