In order to develop a specific tool differentiating the African field strains of Mycoplasma mycoides subsp. mycoides SC from other potentially less virulent strains, including the vaccine strains, we have developed a PCR followed by a restriction enzyme analysis (PCR–REA). This approach also differentiates the African field strains from the Australian strains and the type strain PG1. The genomic marker detected by the PCR–REA is based on a single nucleotide change in the bgl gene that codes for 6-phospho-β-glucosidase (Bgl), an enzyme that is involved in sugar metabolism.  相似文献

OBJECTIVE: To use electromyography (EMG) to measure physiologic activity of the longissimus dorsi muscles of horses during trotting on a treadmill. ANIMALS: 15 adult horses (5 to 20 years old that weighed 450 to 700 kg) that did not have clinical signs of back pain. PROCEDURE: Data were recorded for each horse during trotting on a treadmill at speeds of 2.6 to 4.4 m/s. Surface electromyography was recorded bilaterally from the longissimus dorsi muscles at the levels of T12, T16, and L3. RESULTS: In each motion cycle, 2 EMG maxima were found at the end of the diagonal stance phases. The EMG activity peaked slightly later at L3 than at T12 and T16. Maximum EMG amplitudes were highest at T12 and decreased caudally, with mean +/- SD values of 4.51 +/- 1.20 mV at T12, 3.00 +/- 0.83 mV at T16, and 1.78 +/- 0.67 mV at L3. Mean minimum EMG activity was 1.30 +/- 0.63 mV at T12, 0.83 +/- 0.35 mV at T16, and 0.80 +/- 0.39 mV at L3. The relative amplitudes (ie, [maximum - minimum]/maximum) were 67 +/- 11% at T12, 66 +/- 8% at T16, and 71 +/- 8% at L3. CONCLUSIONS AND CLINICAL RELEVANCE: Activity of the longissimus dorsi muscles is mainly responsible for stabilization of the vertebral column against dynamic forces. The difference between minimum and maximum activity may allow application of this method as a clinical tool. Data reported here can serve as reference values for comparison with values from clinically affected horses.  相似文献

In Switzerland, the incidence of equine botulism and acute pasture myodystrophy have remarkably increased in the last five years. Equine fodder-borne botulism in Europe is most likely caused by Clostridium botulinum types C and D that produce the toxins BoNT/C and BoNT/D. Horses showing signs suggestive of botulism (muscle weakness and tremors, reduced tongue tone, slow chewing, salivation and difficulties swallowing, drooping eyelids, mydriasis), especially patients that have fed on suspect fodder (mostly haylage), must be treated with anti-serum as soon as possible. They also need intensive care, which is often difficult to provide and always expensive in the face of a guarded to poor prognosis. Therefore, prevention (high standards of forage quality and vaccination) is all the more important. Pasture myodystrophy is an acute disease with signs of rhabdomyolysis and lethality rate over 90%. It affects grazing horses under frosty, windy and rainy conditions. Preliminary results indicate that Clostridium sordellii and Clostridium bifermentans producing lethal toxin may play a role in pasture myodystrophy. Our efforts concentrate on developing a new subunit vaccine for equine botulism and understanding the ethiology and pathogenesis of pasture myodystrophy with the goal of improving prevention against these highly fatal diseases that present a significant risk to our horse population.  相似文献

Strains of [Actinobacillus] rossii, [Pasteurella] mairii and [Pasteurella] aerogenes can be isolated from abortion in swine. The RTX toxin Pax has previously been found only in those [P.] aerogenes strains isolated from abortion. Nothing is known about RTX toxins in field isolates of the other two species. To gain insight into the distribution of selected RTX toxin genes and their association with abortion, PCR screening for the pax, apxII and apxIII operons on 21 [A.] rossii and seven [P.] mairii isolates was done. Since species can be phenotypically misidentified, the study was backed up by a phylogenetic analysis of all strains based on 16S rRNA, rpoB and infB genes. The pax gene was detected in all [P.] mairii but not in [A.] rossii strains. No apx genes were found in [P.] mairii but different gene combinations for apx were detected in [A.] rossii strains. Most of these strains were positive for apxIII, either alone or in combination with apxII. Whereas pax was found to be associated to strains from abortion no such indication could be found with apx in [A.] rossii strains. Phylogenetically [A.] rossii strains formed a heterogeneous cluster separated from Actinobacillus sensu stricto. [P.] mairii strains clustered with [P.] aerogenes but forming a separate branch. The fact that [P.] aerogenes, [P.] mairii and [A.] rossii can phylogenetically clearly be identified and might contain distinct RTX toxin genes allows their proper diagnosis and will further help to investigate their role as pathogens.  相似文献
