Partial sequencing of the γ-gliadin gene of 62 spelt and 14 soft wheat cultivars was performed. Fifty-six of the 62 spelt cultivars and 13 of the 14 soft wheat cultivars were shown to exhibit the typical spelt or soft wheat γ-gliadin sequence, respectively. Exceptions were ascribed to crossbreeding of soft wheat and spelt. Using the typical soft wheat γ-gliadin sequence, two alternative DNA-based analytical methods were developed for the detection and quantification of spelt flour "adulteration" with soft wheat. A simple and fast detection of soft wheat in spelt flours could be achieved by restriction fragment length (RFLP) analysis. In combination with lab-on-a-chip capillary gel electrophoresis (LOC-CE) the soft wheat proportion could be estimated. Heteroduplex formation served as additional confirmation for the presence of spelt besides soft wheat. Hence, RFLP-LOC-CE constitutes a perfect analysis tool for the quality control of cereal seeds and pure cultivars. A precise quantification of soft wheat "adulterations" in spelt flour down to 1% could be achieved by the developed real-time PCR method. The calibration parameters of the real-time PCR assay fulfilled the minimum performance requirements of the European Network of GMO (genetically modified organisms) Laboratories (ENGL).  相似文献

A 3‐year field experiment was carried out to determine the significance of root‐growth characteristics contributing to N‐uptake efficiency of two oilseed rape (Brassica napus L.) cultivars differing in N efficiency. Two N treatments were applied, and the core and minirhizotron techniques were used to study root‐length density and number of living roots, respectively. Fertilizer‐N supply increased shoot dry matter, grain yield, total N uptake, and total soil Nmin contents particularly in the top soil. Although significant differences occurred in all parameters between years, the interactions between years and cultivars were mostly not significant. Compared to cv. Capitol, the N‐efficient cv. Apex was characterized by a higher grain yield at N0 and a higher N uptake during reproductive growth. This genotype also had a higher root‐length density and more living fine roots particularly in the topsoil layer. Root growth of this genotype was especially high from beginning of shooting to beginning of flowering, while shoot growth and N uptake during vegetative growth were comparatively low. Our results suggest that N‐efficient cultivars can be characterized by a high investment in root growth during the vegetative stage with a comparatively slow shoot growth and N‐uptake rate until beginning of flowering, which, however, continues during reproductive growth. High root production only during reproductive growth seems to be less effective to achieve high N efficiency, because this may lead to a shortage of assimilates for seed filling. High root‐length density at vegetative stages may thus be advantageous for N uptake and reproductive growth and could be a useful morphological character for the selection and breeding of N‐efficient cultivars.  相似文献

Suitable predictors of degradability are sought to support the identification of biochars with large potential to increase C sequestration in soils. We determined the biodegradation of 9 chars from hydrothermal carbonization and pyrolysis in two agricultural soils. The 200‐ and 115‐day degradation correlated strongly with the O:C‐ and slightly with the H:C‐atomic ratio of 9 and 14 biochars, respectively. Highest temperature treatment and ash content did not show similar correlations.  相似文献

A colonic separation mechanism (CSM) is the prerequisite for the digestive strategy of coprophagy. Two different CSM are known in small herbivores, the 'wash-back' CSM of lagomorphs and the 'mucous-trap' CSM of rodents. Differences between these groups in their digestive pattern when fed exclusively hay were investigated in six rabbits (Oryctolagus cuniculus) and six guinea pigs (Cavia porcellus). Intake, digestibility (by total faecal collection), solute and particle mean retention times (MRT, using Co-EDTA and Cr-mordanted fibres) were measured. Rabbits selected less fibrous parts of the hay than guinea pigs, leaving orts with higher content of neutral detergent fibre [NDF; 721 ± 21 vs. 642 ± 31 g/kg dry matter (DM) in guinea pigs]. They also expressed a lower NDF digestibility (0.44 ± 0.10 vs. 0.55 ± 0.05 of total), a similar particle MRT (15 ± 3 vs. 18 ± 6 h), a longer solute MRT (51 ± 9 vs. 16 ± 4 h), and a lower calculated dry matter gut fill (19.6 ± 4.7 vs. 29.7 ± 4.1 g DM/kg body mass) than guinea pigs (p < 0.05 for each variable). These results support the assumption that the 'wash-back' CSM, exhibited in the rabbits, is more efficient in extracting bacterial matter from the colonic digesta plug than the 'mucous-trap' CSM found in the guinea pigs. Related to metabolic body mass, rabbits therefore need a less capacious colon for their CSM where a more efficient bacteria wash-out is reflected in the lower fibre digestibility. A lighter digestive tract could contribute to a peculiarity of lagomorphs: their ability to run faster than other similar-sized mammals.  相似文献

Veterinarians are often directly involved in clinical studies or requested for information to help interpret their results. Therefore, it is reasonable to examine the reservoir of study methods. This article transfers methodological considerations from clinical research into veterinary medicine. The study question determines the appropriate study method. Recently a ten-step procedure was suggested for selection of appropriate study designs in humans. Based on this approach, a pragmatic study design was adapted to the conditions prevailing in interventional studies in dogs and cats with disturbed behaviour. The different concepts for clinical studies are introduced. Whether or not the design and the evaluation of pragmatic studies in dogs and cats with disturbed behaviour has been maintained and the prerequisites have thereby been fulfilled so that the obtained results are suitable to be applied under everyday conditions can be tested in eight steps. Using the pragmatic design the superiority of complex interventions can be investigated. The results of pragmatic studies help to substantiate a value judgement, i. e., the recommendation or rejection of a specific therapeutic intervention for a defined disease entity in a specific therapeutic setting. The goal of pragmatic studies is to obtain results appropriate for use in everyday situations. In conclusion, the suggested procedure is useful for the selection of the appropriate study designs for specific questions. This procedure is also suitable to test whether the conclusions of published study results coincide with the chosen methods.  相似文献

Some invasive wildlife species have the potential to act as additional host and vector species for parasitic and other infectious diseases. We used the raccoon dog (Nyctereutes procyonides), a carnivore species that has its origin in Asia, as an example to demonstrate biological and ecological prerequisites which enables an invasive species to occupy a new habitat permanently. Studies conducted during the last ten years identified a total of 23 endoparasites, two ectoparasites, six bacterial or protozoan species and five viruses, found in the Nyctereutes procyonoides ussuriensis subspecies in its newly occupied range or in N. procyonoides koreensis in its original range. Results of studies in Finland and Germany furthermore showed that biological characteristics of the raccoon dog make this carnivore an appropriate host or vector for a variety of parasites and infectious diseases. This may result in a growing importance of this invasive carnivore for the epidemiology of transmissible diseases in Germany. Especially with regard to zoonotic disease outbreaks, the raccoon dog should therefore be paid more attention in disease prevention and eradication strategies.  相似文献

Artemisia vulgaris (mugwort) is a tall (1.0?C2.0 m) high biomass perennial herb which accumulates considerable amounts of metals on contaminated sites. An outdoor pot experiment was conducted on a sandy, slightly alkaline soil of moderate fertility to study the uptake of cadmium and the distribution of Cd in plant tissues of A. vulgaris. Cadmium was applied as CdCl2 (a total of 1 l solution of 0, 10, 50 and 100 mg Cd l?1) to 12-l pots with a height of 25 cm. HNO3- and water-extractable concentrations of Cd were correlated with the applied Cd at 2-cm soil depth, but were not correlated at 20-cm soil depth, suggesting that Cd was either not mobile in the soil or completely taken up by mugwort roots. The Cd concentrations in different organs of A. vulgaris and litter increased with increasing soil contamination. Leaf/soil concentration ratios (BCFs) up to 65.93?±?32.26 were observed. Translocation of Cd to the aboveground organs was very high. The leaf/root Cd concentration ratio (translocation factor) ranged from 2.07?±?0.56 to 2.37 ± 1.31; however, there was no correlation of translocation factors to Cd enrichment, indicating similar translocation upon different soil contamination levels. In summary, A. vulgaris is tolerant to the metal concentrations accumulated, has a high metal accumulating biomass and accumulates Cd up to about 70% in the aboveground parts. Both a high phytoextraction potential and a high value for phytostabilisation would recommend mugwort for phytoremediation.  相似文献

Due to long-lasting historic fly-ash deposition from lignite-fired power plants, soils of forest ecosystems in Central Germany have been affected by a variety of contaminants. In addition to heavy metals and oxides, another fly-ash compound is Black carbon (BC) which supposedly has been accumulated as well. BC as a generic term describing different forms of pyrogenetic carbon is considered as a very stable fraction of the C cycle. In this study, BC content of organic layers of 38 sites from two adjacent forested areas was determined using a simplified benzenepolycarboxylic acid (BPCA) method. Considerable amounts of BC were detected in both areas. It accounted for up to 11.2 and 15.6% of total organic carbon (TOC) in the Oe and Oa horizons, respectively. On average, slightly higher BC contents were found in the Oe (23.0 g BC kg?1), whereas BC/TOC ratios were higher in the Oa. The BC/TOC ratio of the Oe horizon tended to decrease with increasing distance from the former main emitters. In contrast to expectations, BC contents of the Oa horizon tended to increase with increasing distance. The deposition gradient, in previous research found for other fly-ash compounds, could not be observed for BC. Higher values in more remote forests might be taken as an indicator for the influence of additional emission sources. Further research is needed to confirm the results and to elucidate the potential impact of (soot-) BC on humus dynamics in the study area.  相似文献

Radiotherapy with or without surgery is a common choice for brain tumors in dogs. Although numerous studies have evaluated use of three‐dimensional conformal radiotherapy, reports of definitive‐intent, IMRT for canine intracranial tumors are lacking. Intensity‐modulated radiation therapy has the benefit of decreasing dose to nearby organs at risk and may aid in reducing toxicity. However, increasing dose conformity with IMRT calls for accurate target delineation and daily patient positioning, in order to decrease the risk of a geographic miss. To determine survival outcome and toxicity, we performed a multi‐institutional retrospective observational study evaluating dogs with brain tumors treated with IMRT. Fifty‐two dogs treated with fractionated, definitive‐intent IMRT at four academic radiotherapy facilities were included. All dogs presented with neurologic signs and were diagnosed via MRI. Presumed radiological diagnoses included 37 meningiomas, 12 gliomas, and one peripheral nerve sheath tumor. One dog had two presumed meningiomas and one dog had either a glioma or meningioma. All dogs were treated in the macroscopic disease setting and were prescribed a total dose of 45‐50 Gy (2.25‐2.5 Gy per fraction in 18‐20 daily fractions). Median survival time for all patients, including seven cases treated with a second course of therapy was 18.1 months (95% confidence of interval 12.3‐26.6 months). As previously described for brain tumors, increasing severity of neurologic signs at diagnosis was associated with a worse outcome. Intensity‐modulated radiation therapy was well tolerated with few reported acute, acute delayed, or late side effects.  相似文献
