Uptake and leakage experiments were performed to study the effects of copper and cadmium on K(+) fluxes in birch (Betula pendula Roth) roots. Labeled rubidium ((86)Rb(+)) was used as a tracer for K(+). Plants were pretreated with Cu or Cd (0, 2, 5 or 25 microM) for 0-300 min and then transferred to radiolabeled nutrient solution (150 microM K(+)) with or without 2,4-dinitrophenol (DNP) to separate the effects of heavy metal on active and passive K(+) influxes. Passive K(+) influx was decreased by pretreatment with Cu but was only slightly affected by pretreatment with Cd. Pretreatment with 2 microM Cu increased active K(+) influx, whereas pretreatment with 25 microM Cu decreased active K(+) influx and intermediate Cu concentrations (5 microM) did not affect active K(+) influx. The pretreatment effects of Cu on active and passive K(+) influxes increased with increasing pretreatment time. During the first hour, pretreatment with Cd decreased active K(+) influx with increasing pretreatment time, whereafter recovery began. To measure K(+) efflux, birch plants were loaded with (86)Rb(+) for 7 days before being exposed to Cu or Cd (0, 1, 3, 5 or 10 microM) in unlabeled nutrient solutions for 24 h. Net efflux of K(+) was measured as (86)Rb(+)-activity in the nutrient solutions 24 h after a heavy metal had been introduced. Efflux of K(+) increased with increasing Cu or Cd concentration in the unlabeled nutrient solution.The data indicate that Cu and Cd affected K(+) influx differently and that recovery mechanism(s), which were induced shortly after heavy metal introduction, counteracted the heavy-metal induced inhibition of active K(+) influx. Efflux of K(+) from plant roots over a 24-h period indicated that Cu and Cd had similar effects on K(+) efflux. There was no evidence of a recovery mechanism counteracting the heavy-metal-induced inhibition of K(+) efflux.  相似文献

The impact on nitrate leaching of agronomic practices designed to immobilize nitrogen in autumn and winter was investigated over 4 years. Experimental treatments (reducing tillage depth, incorporating harvest residues, reducing fertilizer N by growing unfertilized grass or by spring-sown rather than autumn-sown crops) were compared with a control treatment in which autumn crops were sown after burning harvest residues, mouldboard ploughing and seedbed preparation. Winter cover cropping was also compared with winter fallowing. In the first year, incorporation of harvest residues or reducing tillage depth significantly decreased nitrate leaching compared with the control. Unfertilized grass did not affect leaching in the first winter but significantly decreased it in years 2 and 3. When winter cover crops were grown, nitrate leaching was never less than that under an autumn-sown cereal, and in the subsequent year leaching could be significantly greater. Winter fallowing caused the most nitrate leaching over the year. In the winter following a spring-sown crop, leaching under an autumn-sown crop greatly increased. Summed over 4 years, most leaching occurred with the winter fallow—spring cropping treatment; it was 18% more than where a winter cover crop preceded the spring crop. Reducing tillage depth or incorporating harvest residues did not significantly decrease leaching. Unfertilized grass ley followed by an autumn-sown cereal in the fourth year was the only treatment that significantly decrease leaching. Unfertilized grass ley followed by an autumn-sown cereal in the fourth year was the only treatment that significantly reduced leaching loss compared with the control. Incorporating harvest residues resulted in a balance between annual N inputs and outputs. All other treatments required substantial net annual N mineralization to balance annual inputs and outputs.  相似文献

This article suggests a linear functional relationship model for comparing two sets of circular data subject to unobservable errors. Unlike the corresponding and relatively well-studied model for linear data, maximum likelihood estimation for this model is very complicated and no explicit solutions are possible. Using a numerical approximation, we are able to solve the likelihood equations approximately, and to obtain good approximations to the likelihood estimates of the parameters. The quality of our estimates and the feasibility of the estimation method are illustrated via simulation. By establishing a parallel with the model for linear data, we are able to explain the various problems occurring in the process of estimation and to substantiate our numerical results. The interest in the model arose in connection with the study of ocean wave data; an application to such data is also given.  相似文献

The effects of cadmium (Cd) exposure on sunflower (Helianthus annuus L.) nutrient accumulation remain unclear. However, studies concerning crop improvement for Cd tolerance suggest the use of biotechnology techniques such as tissue culture. It is still unknown whether in vitro cells respond to Cd exposure in a way similar to plants. In this paper, the objectives were (1) to characterize the effects of Cd exposure in macronutrient and micronutrient accumulation in different sunflower organs/tissues and (2) to compare the behavior of two culture systems (plants vs. tissue culture) regarding Cd and nutrient accumulation. To achieve these aims, sunflower plants were grown hydroponically in the presence of Cd (at levels of 0, 5, 50, and 500 μ M). For in vitro cultures, seeds were germinated axenically and leaf explants were then grown on Murashige and Skoog medium (MS). One-month-old calluses were grown on MS medium containing 0, 5, 50, and 500 μ M Cd. After 21 d of exposure to 500 μ M, all plants were dead. The contents of macro- and micronutrients and of Cd were determined by ICPS in 18 d-exposed plants and calluses and in calluses exposed for six months to 50 μ M Cd. At day 18, Cd content increased in leaves, roots, and calluses. Cadmium exposure also decreased the contents of magnesium (Mg), calcium (Ca), iron (Fe), and manganese (Mn) in roots and of Mg, Ca, copper (Cu), Fe, and Mn in shoots. Exposed calluses suffered decreases only in Mg, Ca, and Mn contents. The contents of most of these nutrients in six-month-exposed calluses were similar to those of the control calluses, indicating that these long-term exposed in vitro cells developed mechanisms for regulating the effects of Cd on the accumulation of nutrients.  相似文献

Micrometeorological measurements of nighttime ecosystem respiration can be systematically biased when stable atmospheric conditions lead to drainage flows associated with decoupling of air flow above and within plant canopies. The associated horizontal and vertical advective fluxes cannot be measured using instrumentation on the single towers typically used at micrometeorological sites. A common approach to minimize bias is to use a threshold in friction velocity, u*, to exclude periods when advection is assumed to be important, but this is problematic in situations when in-canopy flows are decoupled from the flow above. Using data from 25 flux stations in a wide variety of forest ecosystems globally, we examine the generality of a novel approach to estimating nocturnal respiration developed by van Gorsel et al. (van Gorsel, E., Leuning, R., Cleugh, H.A., Keith, H., Suni, T., 2007. Nocturnal carbon efflux: reconciliation of eddy covariance and chamber measurements using an alternative to the u*-threshold filtering technique. Tellus 59B, 397–403, Tellus, 59B, 307-403). The approach is based on the assumption that advection is small relative to the vertical turbulent flux (FC) and change in storage (FS) of CO2 in the few hours after sundown. The sum of FC and FS reach a maximum during this period which is used to derive a temperature response function for ecosystem respiration. Measured hourly soil temperatures are then used with this function to estimate respiration RRmax. The new approach yielded excellent agreement with (1) independent measurements using respiration chambers, (2) with estimates using ecosystem light-response curves of Fc + Fs extrapolated to zero light, RLRC, and (3) with a detailed process-based forest ecosystem model, Rcast. At most sites respiration rates estimated using the u*-filter, Rust, were smaller than RRmax and RLRC. Agreement of our approach with independent measurements indicates that RRmax provides an excellent estimate of nighttime ecosystem respiration.  相似文献

The prevalence of mycoplasmas in the respiratory tracts of 148 pneumonic calves originating from 25 herds in the Netherlands is reported. Four types of culture media were used to isolate mycoplasmas: solid modified Edward medium, 2 types of Friis media, and A7B differential agar medium. Mycoplasmas were isolated both from nasal swab specimens and lung lavage fluids collected from live calves and from nasal mucosa and lung tissue specimens collected post mortem. All of the mycoplasma strains isolated could be identified as either Ureaplasma diversum (isolated from 80% of 25 herds), Mycoplasma dispar (92%), M. bovirhinis (88%), M. bovis (20%), M. bovigenitalium (4%), M. arginini (16%), or M. canis (12%). Isolation rates of M. dispar and U. diversum were considerably higher from lung lavage fluids than from nasal swab specimens. M. bovis was detected only in fattening herds and not in dairy herds. The respiratory tracts of 75% of the calves examined contained at least 2 mycoplasma species. In total, 25 different combinations of mycoplasma species were detected in specimens collected from noses and lungs. The pathogenic species U. diversum and M. dispar had not been isolated before in the Netherlands.  相似文献

OBJECTIVE: To determine the prevalence and clinical implications of anthelmintic resistance in cyathostomes of horses. DESIGN: Prospective study. ANIMALS: 80 horses on 10 farms in a 5-county region of northeast Georgia. PROCEDURE: On each farm, horses were stratified in descending order according to pretreatment fecal egg count (FEC), blocked into groups of 4, and then randomly assigned to 1 of 4 treatment groups: no treatment (controls), and treatment with pyrantel pamoate, fenbendazole, or ivermectin. Fecal samples were collected 24 hours prior to treatment and 2, 4, and 6 weeks after treatment for determination of FEC. Mean percentage of reduction in FEC was then calculated for each treatment group. For horses from each farm, the efficacy of each anthelmintic was categorized on the basis of mean percentage of reduction in FEC at 2 weeks after treatment (< 80% reduction = ineffective; 80 to 90% reduction = equivocal; and > 90% reduction = effective). RESULTS: Pyrantel pamoate was effective at reducing FEC in horses from 7 farms, ineffective in horses from 2 farms, and equivocal in horses from 1 farm. Fenbendazole was ineffective at reducing FEC in horses from 9 farms and equivocal in horses from 1 farm. Ivermectin was effective at reducing FEC in horses from all 10 farms. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that cyathostome resistance to fenbendazole is highly prevalent, and resistance to pyrantel pamoate is high enough to warrant concern. Resistance to ivermectin was not detected. On the basis of these data, it appears that ivermectin continues to be fully effective in horses. However, too few farms were used in this study to determine the prevalence of cyathostome resistance to ivermectin. Therefore, the efficacy of ivermectin should continue to be monitored closely.  相似文献

Coastal lands around Bay of Bengal in Central Godavari Delta are mainly agriculture fields and two times annually paddy crops putting in the study area. Canals of Godavari River are the main source of water for irrigation. Geophysical and geochemical investigations were carried out in the study area to decipher subsurface geologic formation and assessing seawater intrusion. Electrical resistivity tomographic surveys carried out in the watershed-indicated low resistivity formation in the upstream area due to the presence of thick marine clays up to thickness of 20-25?m from the surface. Secondly, the lowering of resistivity may be due to the encroachment of seawater in to freshwater zones and infiltration during tidal fluctuation through mainly the Pikaleru drain, and to some extent rarely through Kannvaram and Vasalatippa drains in the downstream area. Groundwater quality analyses were made for major ions revealed brackish nature of groundwater water at shallow depth. The in situ salinity of groundwater is around 5,000?mg/l and there is no groundwater withdrawal for irrigation or drinking purpose in this area except Cairn energy pumping wells which is using for inject brackish water into the oil wells for easy exploration of oil. Chemical analyses of groundwater samples have indicated the range of salt concentrations and correlation of geophysical and borehole litholog data in the study area predicting seawater-contaminated zones and influence of in situ salinity in the upstream of study area. The article suggested further studies and research work that can lead to sustainable exploitation/use and management of groundwater resources in coastal areas.  相似文献
