This review addresses the quantification of anthropogenic pollutants in lacustrine sediments by multidisciplinary analyses including: chronostratigraphy using radioisotopes (137Cs) and radiocarbon dates (14C), trace metal analysis, faecal indicator analysis, as well as antibiotic-resistant genes by molecular analysis. Sediment cores from lakes Lucerne and Geneva that are located at a distance of 150?km from each other reveal a synchronous increase in anthropogenic trace metals (Pb, Cu, Zn, and Mn) following the industrial revolution in Europe about 1850. In both lakes, the peak of water pollution by toxic metals due to discharge of industrial wastewaters was reached in the middle of the twentieth century. During the second part of the twentieth century, both sites show a decrease in metal pollution following the implementation of wastewater treatment plants. On the contrary, the Vidy Bay of Lake Geneva where the treated wastewaters from the city of Lausanne are released since 1964 points out a dramatic increase in trace metal deposition. Later, a high increase in organic matter deposition, in bacteria (Escherichia coli and Enterococcus faecalis) activity as well as antibiotic-resistant genes and bacteria occurred into the bay, simultaneously with the eutrophication of the large and deep perialpine lakes in the 1970s due to excessive external nutrient loading.  相似文献

Aged plants are more difficult to infect than young plantlets. This modification of susceptibility is described as mature plant resistance (MPR). For potato virus Y (PVY), MPR is known to lead to low infection rates of plants inoculated at the postflowering stage and a decrease in the number of infected daughter tubers. However, the impact of inoculation date on the capacity of PVY to accumulate in daughter tubers has not been studied so far. Field and greenhouse experiments were carried out to better understand PVY epidemiology and to help potato growers to evaluate consequences of early/late infections on the quality of their crops. In field trials, potato plants (cv. Bintje) were covered by insectproof nets from planting to harvest except for a 14-day period to expose plants to natural PVY infections. Under controlled conditions, potato plants were mechanically inoculated with PVY at different dates from preflowering stages (early inoculations) to postflowering stage (late inoculations). At harvest, daughter tubers were individually collected and analysed to define proportions and viral load of infected tubers according to the time between virus inoculation and harvest. Our results showed that although the age of plants at the time of inoculation can modify their susceptibility to PVY infection, in return, early and late PVY inoculations lead to similar rates of infected tubers at the plant scale and equivalent viral accumulation in infected tubers. All together, these data revealed that both early/late infections are high risks for the sanitary quality of potato tubers.  相似文献

The present study was conducted to determine the spatial heterogeneity of bulk density, soil moisture, inorganic N, microbial biomass C, and microbial biomass N in the ridge tillage system of Turiel compared to conventional mouldboard ploughing on three sampling dates in May, July, and August. The soil sampling was carried out under vegetation representing the ridge in a high spatial resolution down the soil profile. Bulk density increased with depth and ranged from 1.3 g cm−3 at 10 cm depth to 1.6 g cm−3 at 35 cm in ploughed plots and from 1.0 g m−3 at 5 cm to 1.4 g m−3 at 35 cm in the ridges. In the ploughed plots, the contents of microbial biomass C and microbial biomass N remained roughly constant at 215 and 33 μg g−1 soil, respectively, throughout the experimental period. The microbial biomass C/N ratio varied in a small range around 6.4. In the ridged plots, the contents of microbial biomass C and microbial biomass N were 5% and 6% higher compared to the ploughed plots. Highest microbial biomass C contents of roughly 300 μg g−1 soil were always measured in the crowns in July. The lowest contents of microbial biomass C of 85–137 μg g−1 soil were measured in the furrows. The ridges showed strong spatial heterogeneity in bulk density, soil water content, inorganic nitrogen and microbial biomass.  相似文献

The polypyrimidine tract binding protein (PTB) is a 58-kilodalton RNA binding protein involved in multiple aspects of messenger RNA metabolism, including the repression of alternative exons. We have determined the solution structures of the four RNA binding domains (RBDs) of PTB, each bound to a CUCUCU oligonucleotide. Each RBD binds RNA with a different binding specificity. RBD3 and RBD4 interact, resulting in an antiparallel orientation of their bound RNAs. Thus, PTB will induce RNA looping when bound to two separated pyrimidine tracts within the same RNA. This leads to structural models for how PTB functions as an alternative-splicing repressor.  相似文献

The long-term (9 years) effect of pig slurry applications vs mineral fertilization on denitrifying activity, N2O production and soil organic carbon (C) (extractable C, microbial biomass C and total organic C) was compared at three soil depths of adjacent plots. The denitrifying activities were measured on undisturbed soil cores and on sieved soil samples with acetylene method to estimate denitrification rates under field or potential conditions. Pig slurry applications had a moderate impact on the C pools. Total organic C was increased by +6.5% and microbial biomass C by ≥25%. The potential denitrifying activity on soil suspension was stimulated (×1.8, P<0.05) 12 days after the last slurry application. This stimulation was still apparent, but not significant, 10 months later and, according to both methods of denitrifying activity measurement (r 2=0.916, P<0.01 on sieved soil; r 2=0.845, P<0.001 on soil cores), was associated with an increase in microbial biomass C above a threshold of about 105 mg kg−1. The effect of pig slurry on denitrification and N2O reduction rates was detected on the surface layer (0–20 cm) only. However, no pig slurry effect could be detected on soil cores at field conditions or after NO3 enrichments at 20°C. Although the potential denitrifying activity in sieved soil samples was stimulated, the N2O production was lower (P<0.03) in the plot fertilized with pig slurry, indicating a lower N2O/(N2O + N2) ratio of the released gases. The pig-slurry-fertilized plot also showed a higher N2O reduction activity, which is coherent with the lower N2O production in anaerobiosis.  相似文献

The mycotoxin T-2 toxin, produced by various Fusarium species, is a widespread contaminant of grain and grain products. Knowledge about its toxicity and metabolism in the human body is crucial for any risk assessment as T-2 toxin can be detected in processed and unprocessed food samples. Cell culture studies using cells of human origin represent a potent model system to study the metabolic fate of T-2 toxin as well as the cytotoxicity in vitro. In this study the metabolism of T-2 toxin was analyzed in a cell line derived from human colon carcinoma cells (HT-29) and primary human renal proximal tubule epithelial cells (RPTEC) using high-performance liquid chromatography coupled with Fourier transformation mass spectrometry (HPLC-FTMS). Both cell types metabolized T-2 toxin to a variety of compounds. Furthermore, cell cycle analysis in RPTEC proved the apoptotic effect of T-2 toxin and its metabolites HT-2 toxin and neosolaniol in micromolar concentrations.  相似文献

In recent years, arsenic (As) has received increased attention as humans may be exposed to it through occupational and environmental exposure. Tobacco (Nicotiana tabacum L.) like other crops can uptake this element from the soil, which may lead to human exposure. Here, we report on a survey on arsenic in cured or processed tobacco leaves obtained from Africa, Asia, Europe, South and North America. A total of 1,431 leaf samples of flue-cured, burley, and Oriental tobaccos were obtained from various sampling locations during 2002 to 2004. Arsenic concentration in the samples averaged 0.4?±?0.6 μg g?1 as determined by inductively coupled plasma-mass spectrometry. Recorded values from most samples showed that concentrations of arsenic were usually found at the lower end of the distribution. Significant differences were found among tobacco types, sampling locations, and crop years. Arsenic concentrations were rather low in the majority of regions investigated, which is compatible with data from the literature. However, sample size was small and sampling geographically restricted. Our results would need to be validated with a larger dataset.  相似文献
