At present, Foot-and-Mouth Disease (FMD) has been successfully controlled in most territories of South America, where only Ecuador and Venezuela remain as endemic countries. In this context, the precise characterization of circulating viruses is of utmost importance. This work describes the first molecular epidemiology study performed with the complete VP(1)-coding region of 114 field isolates of FMD virus (FMDV) type O, collected in the Andean countries mainly during 2002-2008. Sequences were aligned and compared to isolates responsible for emergencies in the Southern Cone of the continent between the years 2000 and 2006, and to other representative type O viruses worldwide. The results showed that FMD type O viruses isolated in South America and analyzed up to date are placed in 11 different lineages within the Euro SA topotype. Five of these lineages included viruses circulating in Ecuador and Venezuela during 2002-2008. The last emergencies reported in already-free areas in the Andean region, showed close relationships with viruses circulating in these endemic countries. Andean lineages showed a clear separation from the unique lineage containing viruses responsible for the emergencies in the Southern Cone, reflecting the different livestock circuits and providing evidence that support the ecosystem dynamics in the region. A wide geographical dissemination of the same strain in short time intervals has been observed, pointing to animal movements as the most significant risk parameter. This fact, together with an important generation of viral variants in areas under weak control strategies, reinforce the need of stronger official controls, as well as for establishing multinational cooperative measures in the border areas.  相似文献

Macrolides are important antimicrobials frequently used in human and veterinary medicine in the treatment of pregnant women and pregnant livestock. They may be useful for the control of infectious ovine abortion, which has economic, animal health, and human health impacts. In this study, catheters were surgically placed in the fetal vasculature and amnion of pregnant ewes at 115 (±2) days of gestation. Ewes were given a single dose of 2.5 mg/kg tulathromycin subcutaneously, and drug concentrations were determined in fetal plasma, maternal plasma, and amniotic fluid at 4, 8, 12, 24, 36, 48, 72, 144, and 288 hr after drug administration. Pharmacokinetic parameters in maternal plasma were estimated using noncompartmental analysis and were similar to those previously reported in nonpregnant ewes. Tulathromycin was present in fetal plasma and amniotic fluid, indicating therapeutic potential for use against organisms in these compartments, though concentrations were lower than those in maternal plasma. Time‐course of drug concentrations in the fetus was quite different than that in the ewe, with plasma concentrations reaching a plateau at 4 hr and remaining at this concentration for the remainder of the sampling period (288 hr), raising questions about how tulathromycin may be transported into or metabolized and eliminated by the fetus.  相似文献

Enzymes are protein catalysts of extraordinary efficiency, capable of bringing about rate enhancements of their biochemical reactions that can approach factors of 1020. Theories of enzyme catalysis, which seek to explain the means by which enzymes effect catalytic transformation of the substrate molecules on which they work, have evolved over the past century from the “lock-and-key” model proposed by Emil Fischer in 1894 to models that explicitly rely on transition state theory to the most recent theories that strive to provide accounts that stress the essential role of protein dynamics. In this paper, I attempt to construct a metaphysical framework within which these new models of enzyme catalysis can be developed. This framework is constructed from key doctrines of process thought, which gives ontologic priority to becoming over being, as well as tenets of a process philosophy of chemistry, which stresses environmentally-responsive molecular transformation. Enzyme catalysis can now be seen not as enzyme acting on its substrate, but rather as enzyme and substrate entering into a relation which allows them to traverse the reaction coordinate as an ontologic unity x̄.  相似文献

Fungi inhabiting Japanese pear were isolated from internal tissues of cv. Nijisseiki, and culture filtrates (CFs) of 100 isolates were evaluated for their inhibitory activity against infection by Alternaria alternata Japanese pear pathotype. CFs of 11 isolates inhibited lesion formation on the pear by the pathogen. Among these isolates, CFs of five isolates inhibited spore germination. CFs of the six other isolates inhibited appressorial formation, infection hypha formation, AK-toxin production, or a combination of these actions. Analysis of sequence homology in the rDNA ITS1 regions of these isolates showed that most isolates had high homology with some fungal endophytes.  相似文献

Plant growth responses to biochar addition: an Australian soils perspective   总被引:1,自引:0,他引:1  
The use of biochar as an agricultural amendment has attracted much attention owing to its potential to improve soil condition and plant growth; however, production outcomes are often uncertain. Although soil type is a major driver of plant productivity, there are relatively few biochar studies that directly compare plant growth responses across a range of soil types. We tested the wheat growth response to biochar derived from poultry litter and from wheat straw applied at 1, 5 and 10 t ha?1 (approximately 0.13, 0.67 and 1.33 % w/w) in four soils representing major agricultural regions in Australia: an acidic arenosol (Western Australian cereal belt), an acidic rhodic ferralsol (Northern New South Wales), a neutral vertisol (Queensland cropping) and an alkaline haplic calcisol (Eyre Peninsula in South Australia). In the neutral vertisol, where plant growth was vigorous in the control treatments, biochar had little impact, whereas in the alkaline calcisol, there was a small significant increase in shoot biomass at high (10 t ha?1) application rates. Plant growth responses in the acidic soils were most evident but demonstrated a strong contrast to one another. In the acidic arenosol, negative growth impact correlated with increasing electrical conductivity, while in the acidic ferralsol a small rate-dependent increase in pH correlated with relatively large gains in biomass, possibly due to improved phosphorus nutrition and alleviated Al toxicity. Moving towards effective integration of biochar as a management tool will not only require stratification based on soil types, but wider consideration of the main plant production constraints, such as pH, pertinent to a particular system.  相似文献

Nematodes have difficult control and complex handling, but considering the physiological and biochemical changes that micronutrients promoting in plants, there is possibility that the supply with these chemicals increases the resistance of plants against nematodes. Thus, the present study aimed to assess the effect of the application of boron and zinc on the reproduction of Meloidogyne incognita, embryonic development and juvenile nematode hatching. Nematode reproduction was evaluated in tomato plant inoculated with 2000 eggs and treated in the aerial part with boron or zinc at the following doses: 0, 1/2, 1×, 2× and 4× the manufacturer's recommendation (100 and 30?g/L, respectively), with the plants assessed 60 days after inoculation. For assessment of embryonic development and juvenile hatching, 1?mL nematode suspension was placed in Petri dishes containing 9?mL of the same doses of boron and zinc, and assessment occurred four and eight days after incubation. Results obtained showed that boron controlled nematode population at the dose of 400?g/L and promoted juvenile hatching when used at maximum dosage on the eighth day. Zinc reduced the number of galls and the number of eggs at the dose of 60?g/L, but did not exhibit direct effect on nematode.  相似文献

Animals may respond to spatial and temporal heterogeneity by altering their movement patterns. The time an animal spends in an area of a given size is termed ȁ8first-passage timeȁ9 and can be used to identify the scales at which different movement processes occur. Using first-passage time and 2-h observations, we identified nested spatial scales representing three movement behaviours for elk (Cervus elaphus) – inactive/resting (moves < 50 m), active/foraging (x̄ = 276.7 m, SD = 56.6), and active/relocating (x̄ = 1628.3 m, SD = 436.6). Our ability to identify inactive behaviour was limited by GPS accuracy. The scale separating relocating and foraging behaviour ranged 550–1650 m across individuals and varied quadratically with the mean patch size of cutover forest in an animal’s home range. We classified path segments into the 3 movement behaviours and related behaviours to local environmental conditions. Elk were likely to be inactive in areas having a low predicted use by wolves (Canis lupus), farther than 50 m from anthropogenic linear clearings, and where microclimatic conditions were cool (high shrub cover and north to east-facing slopes). In contrast, elk were most likely to forage in areas having intermediate levels of herbaceous biomass and low movement costs. Elk were most likely to be relocating when in areas of high wolf use, when close to linear clearings, and in energetically costly situations such as moving upslope. We discuss how elk use of potential foraging habitats may be restricted in this landscape by risks imposed by predators, humans, or both.  相似文献
