Chronic wasting disease (CWD) is a fatal disease of North American cervids that was first detected in a wild, hunter-shot deer in Saskatchewan along the border with Alberta in Canada in 2000. Spatially explicit models for assessing factors affecting disease detection are needed to guide surveillance and control programs. Spatio-temporal patterns in CWD prevalence can be complicated by variation in individual infection probability and sampling biases. We assessed hunter harvest data of mule deer (Odocoileus hemionus) and white-tailed deer (Odocoileus virginianus) during the early phases of an outbreak in Saskatchewan (i.e., 2002-2007) for targeting the detection of CWD by defining (1) where to look, and (2) how much effort to use. First, we accounted for known demographic heterogeneities in infection to model the probability, P(E), that a harvested deer was infected with CWD given characteristics of the harvest location. Second, in areas where infected deer were harvested we modelled the probability, P(D), of the hunter harvest re-detecting CWD within sample units of varying size (9-54 km(2)) given the demographics of harvested deer and time since first detection in the study area. Heterogeneities in host infection were consistent with those reported elsewhere: mule deer 3.7 times >white-tailed deer, males 1.8 times>females, and quadratically related to age in both sexes. P(E) increased with number of years since the first detection in our study area (2002) and proximity to known disease sources, and also varied with distance to the South Saskatchewan River and small creek drainages, terrain ruggedness, and extent of agriculture lands within a 3 km radius of the harvest. The majority (75%) of new CWD-positive deer from our sample were found within 20 km of infected deer harvested in the previous year, while approximately 10% were greater than 40 km. P(D) modelled at 18 km(2) was best supported, but for all scales, P(D) depended on the number of harvested deer and time since the first infected deer was harvested. Within an 18 km(2) sampling unit, there was an 80% probability of detecting a CWD-positive deer with 16 harvested deer five years after the initial infected harvest. Identifying where and how much to sample to detect CWD can improve targeted surveillance programs early in the outbreak of the disease when based on hunter harvest.  相似文献

Simple wet heat treatments like simple boiling (atmospheric pressure, 100 °C) and pressurized boiling (higher than 100 °C) reduced the polyphenol content of mature dark red seeds of cowpea (Vigna unguiculata (L.) Walp.) cultivar UPL Cp 3 by 61 to 80% and improved in vitro protein digestibility (IVPD) by 6 to 26%. Pressurized steaming (higher than 100 °C) removed 48 to 83% of the polyphenols but increased IVPD by only 1.1 to 4.2%. Dry heat as exemplified by roasting and microwave treatment inactivated 58 to 71% of the tannins but increased IVPD by only 1%. All the heat treatments were effective in removing/inactivating polyphenols though different responses were observed with the resulting in vitro protein digestibilities.  相似文献

An indirect immunofluorescence (IFA) test with a 96-well, flat-bottomed microplate was developed to detect avian pneumovirus (APV) antigen in Vero cell cultures. Samples of nasal turbinates and swabs from infraorbital sinuses and trachea were collected from 4-week-old poults experimentally inoculated with APV. The APV titers by tissue culture IFA staining were compared with that of visual reading of cytopathic effect (CPE). The ability of IFA staining to detect APV antigen correlated well with visualizing CPE. The use of IFA staining of Vero cell cultures allowed detection of APV in substantially less time than the use of visualizing CPE. In addition, the use of IFA allowed specific identification of the virus in cell culture.  相似文献

Somatic hybrids were produced by PEG-induced symmetric and asymmetric protoplast fusions in order to transfer resistance to Alternaria brassicicola, A. brassicae, Phoma lingam, Plasmodiophora brassicae and Turnip mosaic virus (TuMV) into Brassica oleracea var. capitata (cv. ‘Toskama’) and botrytis (cv. ‘Korso’). As resistance donors, ten species belonging to several genera of the family Brassicaceae including wild relatives were used. Of 2,189 plants (somatic hybrids, partially in vitro cloned) tested, 1,616 (73.8%) were resistant against at least one of the pathogens, indicating that, mostly, a successful resistance transfer has taken place. Five hundred and twenty-two hybrids showed multiple resistances to two, three and, in a single case, to four pathogens. Irrespective of the donor parents used in the fusion, a broad variability in symptom manifestation ranging from 0 (without symptoms) to 9 (highly susceptible) could be observed. With regard to the Alternaria pathogens, Sinapis alba, B. nigra and B. juncea were the most effective resistance donors, whereas fusions with Raphanus sativus resulted in the most hybrids with resistance to clubroot and TuMV. As could be shown especially in asymmetric fusions with S. alba, Barbarea vulgaris and Hesperis matronalis, transferred resistance to a pathogen may not correspond with resistance exhibited by the donor parent. Some combinations in which both parents were highly susceptible, e.g. R. sativus (+) B. oleracea var. capitata, yielded hybrids that exhibited strong resistance, e.g. to A. brassicicola, revealing that a new type of resistance might be occurring. With regard to the Alternaria pathogens, resistance expression was very unstable. Many hybrids into which (also variable) resistance of some donors, such as B. vulgaris, S. alba and B. carinata, was transferred became as highly susceptible as those of which the fusion parents did not show any resistance reaction (e.g. R. sativus). For reliable characterization of the resistance response, hybrids should be subjected to several resistance tests during growth period of the host, at least until flowering.  相似文献

Recurrent selection is a method for population improvement which has been used in soybean [Glycine max (L.) Merrill] to modify traits such as grain yield, seed-protein content, seed-oil content, tolerance to iron-deficiency chlorosis, and seed size. Nuclear male-sterility with insect-mediated cross-pollination has been successfully used in recurrent selection schemes in soybean. However, little attention has been given to selection to increase the agronomic performance of male-sterile plants per se. The objective of this study was to evaluate the response of male-sterile lines segregating for male-sterile alleles ms2, ms3, ms6, ms8, and ms9 to phenotypic recurrent selection for increased seed-set after 3 cycles, using a selected group of male parents. Bees halictidae, anthophoridae, andrenidae, and megachilidae were utilized as the pollinator vector. The results indicated that recurrent selection in a favorable environment was successful to increase the number of seeds per male-sterile plant. Although a differential response was observed among populations, the seed-set observed would justify the use of some specific male-sterile selections as female parents in a hybrid soybean seed production system.  相似文献

Breeders are encouraged to develop breeding approaches that strive to integrate food production into the healthy functioning of agro-ecosystems. In the case of legumes, this approach should preserve bee fauna by providing suitable floral resources within the crops themselves. In parallel, legume breeding for sustainable agriculture is linked to the development of environmental services. Foraging places and nesting sites for solitary and social bees are some of the ecological services provided for legumes. Crops with floral attractiveness and rewards for insects can be used to enhance pollinator conservation as well as crop yield and yield stability. We analyze how understanding crop-pollinator relationships (CPR) can contribute to the production of high-yielding and pollinator-friendly varieties by examining: (1) The status of knowledge on mating systems and floral traits; (2) The contribution of CPR understanding to plant breeding for both hybrid-seed production and open-pollinated population improvement.  相似文献

Horses were tested for recognition of objects under various rotations to better understand how they perceive stimuli. Four horses learned to discriminate positive and negative stimuli for three sets of “real-life” three-dimensional objects always presented in one particular orientation (front to left, top up). When the horses reliably performed at an above chance criterion of 80% correct responses for two consecutive runs of 20 trials (P = .0046), the objects were rotated in depth and/or turned upside down, for example, front to right, top to horse; back to horse, bottom up. Overall performance on rotated trials, as well as Trial 1 scores on novel presentations, indicated that the horses were capable of recognizing objects under all rotational conditions. However, there was a degree of individual variation, with some horses performing better than others on certain rotations. Moreover, objects rotated in certain positions were more easily recognized than others. The most significant effect was seen when rotations with the top of the objects visible were compared with those with the bottom visible—the former being more recognizable than the latter. This suggests that the horses were using certain features of the objects on which to base their decisions. These findings provide new information on object perception in horses and address one common belief about why horses startle at objects that should be familiar to them.  相似文献

Coral transplantation was used to evaluate the response of corals to trampling by determining growth and mortality at sites that ranged along a gradient of human use. Human use was measured with observational sampling. A clear progression of coral survivorship along the gradient was evident. Survivorship dropped from 70% at the low impact site to 55% at the medium impact site. Total loss (0% survivorship) was reported from the high impact site after only 8 months, equivalent to less than 200,000 total visitors or 63 people in the water per hour. Where transplanted corals survived, there was no difference in growth, presumably due to the control of activities of people in the water at those sites.  相似文献

Metamorphosis in the flounder has often been compared with the transition of tadpoles into frogs. The dorsal fin rays of the Japanese flounder (Paralichthys olivaceus) elongate during prometamorphosis when thyroid hormone levels are low, and are resorbed during metamorphic climax when thyroid hormone levels are high. Using an in vitro system for the culture of the flounder fin rays, we have examined how various hormones affect the resorption process. Both thyroxine (T4) and triiodothyronine (T3) directly stimulated fin ray shortening, T3 being more potent than T4. Other hormones, such as prolactin, cortisol and sex steroids, did not directly affect the resorption process but modified the tissue's response to thyroid hormones. Similar observations were obtained from in vivo studies. We also monitored the changes in the whole body concentrations of various hormones during early development and metamorphosis, and related these with the thyroid hormone profiles in order to get a better picture of their interactions. The gaps in the present status of research on the role of thyroid hormones during metamorphosis in the Japanese flounder are also discussed.  相似文献
