This study reports a functional characterization of a limited segment (QTL) of sheep chromosome 12 associated with resistance to the abomasal nematode Haemonchus contortus. The first objective was to validate the identified QTL through the comparison of genetically susceptible (N) and resistant (R) sheep produced from Martinik × Romane back-cross sheep. The R and N genotype groups were then experimentally infected with 10 000 H. contortus larvae and measured for FEC (every three days from 18 to 30 days post-challenge), haematocrit, worm burden and fertility. Significant differences in FEC and haematocrit drop were found between R and N sheep. In addition, the female worms recovered from R sheep were less fecund. The second step of the characterization was to investigate functional mechanisms associated with the QTL, thanks to a gene expression analysis performed on the abomasal mucosa and the abomasal lymph node. The gene expression level of a candidate gene lying within the QTL region (PAPP-A2) was measured. In addition, putative interactions between the chromosome segment under study and the top ten differentially expressed genes between resistant MBB and susceptible RMN sheep highlighted in a previous microarray experiment were investigated. We found an induction of Th-2 related cytokine genes expression in the abomasal mucosa of R sheep. Down-regulation of the PAPP-A2 gene expression was observed between naïve and challenged sheep although no differential expression was recorded between challenged R and N sheep. The genotyping of this limited region should contribute to the ability to predict the intrinsic resistance level of sheep.  相似文献

Purpose

Humic substances, which are integral components of total organic carbon (TOC), influence soil quality. The study aimed to investigate whether humic and non-humic fractions exhibit early, consistent, and measurable changes and affect TOC sensitivity and storage in a tropical sandy loam soils amended with corn cob biochar.

Materials and methods

There were four treatments with four replicates established in a randomized complete block design. Composite soil samples were taken from plots without biochar (CT), from plots incorporated with 15 t biochar ha?1 (BC-15), and 30 t biochar ha?1 without or with phosphate fertilizer (BC-30 and BC-30+P). The TOC, and humin, humic acid (HA), and fulvic acid (HA) fractions of soil organic carbon were determined for each treatment. The optical densities (400–700 nm) were measured on the soil-free extracts by spectrophotometry; the densities measured at 465 and 665 nm were used to calculate the E465/E665 ratios.

Results and discussion

The BC-30 and BC-30+P plots recorded the highest TOC, humin, humic acid (HA), and fulvic acid (FA) contents with respect to the lowest in the CT. The total exchangeable carbon stratification was significantly higher in all the biochar-treated plots relative to the CT. Spectral analysis showed higher values of E465/E665 (5.02 and 5.15) in the CT and BC-15-treated soils, respectively, compared with the BC-30 and BC-30+P-amended soils with E465/E665 ratios of 2.76 and 2.98, respectively.

Conclusions

Corn cob biochar applied to a tropical sandy loam:

? increased the concentrations of HA and FA and led to increased stratification of TOC, with a stronger effect on HA compared with FA;

? significantly lowered E465/E665 at the high biochar application rate of 30 t ha?1, implying the dominance of high molecular weight humic acid-like substances, and increased degree of aromaticity of the TOC.

Sandy soils, with low productivity, could be improved by compost application to sustain crop production. This study aimed to examine the effect of three compost types (vegetable, fruit and yard waste compost, garden waste compost, and spent mushroom compost) on basic properties of a loamy sand and greenhouse tomato productivity. Disturbed and intact soil samples were taken from a decade-long compost field experiment on loamy sand with three compost types at application rate of 30 m3 ha?1 yr?1 (7.5 ton ha?1 yr?1). The soils were characterized for chemical and physical properties. Tomato was planted in a greenhouse using soil samples from the field and vegetative and yield parameters (plant height, stem diameter, leaf number, and fruit yield), water productivity, and harvest index were evaluated. All compost types significantly increased soil total carbon, total nitrogen, pH, electrical conductivity and significantly decreased bulk density, with no effect on plant available water compared to the control. Fresh and dry fruit weights were significantly increased after compost addition. Plant height, leaf number, stem diameter, and total biomass did not significantly improve after compost addition. Spent mushroom compost had greater effect in improving tomato productivity. A decade-long application of composts on loamy sand improved basic chemical and physical properties which were reflected in increased fruit yield in tomato. Since no negative effect of compost was observed, we suggest that sandy soils may serve as a safe end use option for these composts and potentially support crop growth.  相似文献

Plant response to increasing atmospheric CO2 partial pressure (pCO2) depends on several factors, one of which is mineral nitrogen availability facilitated by the mineralisation of organic N. Gross rates of N mineralisation were examined in grassland soils exposed to ambient (36 Pa) and elevated (60 Pa) atmospheric pCO2 for 7 years in the Swiss Free Air Carbon dioxide Enrichment experiment. It was hypothesized that increased below-ground translocation of photoassimilates at elevated pCO2 would lead to an increase in immobilisation of N due to an excess supply of energy to the roots and rhizosphere. Intact soil cores were sampled from Lolium perenne and Trifolium repens swards in May and September, 2000. The rates of gross N mineralisation (m) and NH4+ consumption (c) were determined using 15N isotopic dilution during a 51-h period of incubation. The rates of N immobilisation were estimated either as the difference between m and the net N mineralisation rate or as the amount of 15N released from the microbial biomass after chloroform fumigation. Soil samples from both swards showed that the rates of gross N mineralisation and NH4+ consumption did not change significantly under elevated pCO2. The lack of a significant effect of elevated pCO2 on organic N turnover was consistent with the similar size of the microbial biomass and similar immobilisation of applied 15N in the microbial N pool under ambient and elevated pCO2. Rates of m and c, and microbial 15N did not differ significantly between the two sward types although a weak (p<0.1) pCO2 by sward interaction occurred. A significantly larger amount of NO3 was recovered at the end of the incubation in soil taken from T. repens swards compared to that from L. perenne swards. Eleven percent of the added 15N were recovered in the roots in the cores sampled under L. perenne, while only 5% were recovered in roots of T. repens. These results demonstrate that roots remained a considerable sink despite the shoots being cut at ground level prior to incubation and suggest that the calculation of N immobilisation from gross and net rates of mineralisation in soils with a high root biomass does not reflect the actual immobilisation of N in the microbial biomass. The results of this study did not support the initial hypothesis and indicate that below-ground turnover of N, as well as N availability, measured in short-term experiments are not strongly affected by long-term exposure to elevated pCO2. It is suggested that differences in plant N demand, rather than major changes in soil N mineralisation/immobilisation, are the long-term driving factors for N dynamics in these grassland systems.  相似文献

Inverting radiative transfer (R-T) models against remote sensing observations to retrieve key biogeophysical parameters such as leaf area index (LAI) is a common approach. Even if new inversion techniques allow the use of three-dimensional (3D) models for that purpose, one-dimensional (1D) models are still widely used because of their ease of implementation and computational efficiency. Nevertheless, they assume a random distribution of foliage elements whereas most canopies show a clumped organization. Due to that crude simplification in the representation of the canopy structure, sizeable discrepancies can occur between 1D simulations and real canopy reflectance, which may further lead to false LAI values. The present investigation aims to appraise to which extent the incorporation of a clumping index (noted λ) into 1D R-T model could improve the simulations of Bidirectional Reflectance Distribution Function (BRDF). Canopy BRDF is simulated here for three growth stages of a maize crop with the Discrete Anisotropic Radiative Transfer (DART) model in the visible and near infrared spectral bands, for two contrasted soil types (dark and bright) and different levels of heterogeneity to represent the canopy structure. 3D numerical scenes are based on in-situ structural measurements and associated BRDF simulations are thus considered as references. 1D scenarios assume either that leaves are randomly distributed (λ = 1) or clumped (λ < 1). If BRDF simulations seem globally reliable under the assumption of a random distribution in near infrared, it can also lead to relative errors on the total BRDF up to 30% in the red spectral band. It comes out that the use of a clumping index in a 1D reflectance model generally improves BRDF simulations in the red considering a bright soil, which seems relatively independent of LAI. In the near infrared, best results are usually obtained with homogeneous canopies, except with the dark soil. Clearly, influent factors are mainly the LAI and the spectral contrast between soil and leaves.  相似文献
