The morphological and physiological development of the intestine, stomach, liver and pancreas of the African catfish, Clarius guriepinus (Burchell 1822), was investigated from hatching until 9 days after the start of exogenous feeding by histological, histochemical and immunohistochemical procedures. During the yolk sac period, lasting 48 hours at 30 C, the digestive system develops from a rather undifferentiated system into a distinct liver and pancreas and a segmented intestine (parts I to III). Insulin and glucagon producing cells were clearly found in the endocrine pancreatic islets from 14 hours after hatching onwards, while at the start of exogenous feeding, the exocrine pancreas showed an intense zymogen colouration. Exogenous feeding started three days after fertilization, i.e., 48 hours after hatching. At that moment, the rearing temperature was lowered to 27.5 C. The first day of feeding was referred to as day zero, changing into day 1 when the first 24 hours of feeding were completed. From the start of exogenous feeding onwards, aminopeptidase, non-specific esterase and ATP-ase were detected in the digestive system. Morphologically, the stomach was completed at day 4 of exogenous feeding. Gastric acid secretion started on day 4 and lead to a pH below 3.3 in the stomach on day 5. During the first four days of exogenous feeding, C-t-gastrin immunopositive cells were found throughout the gut, including the gastric epithelium. After day 4, the C-t-gastrin cells were only detected in the anterior intestine. The completeness of the stomach development was also marked by the appearance of non-specific esterase activity in the stomach epithelial cells on day 5. All data together clearly indicate that in Clarius gariepinus , the ontogeny of a functional digestive system has been completed on day 5 after the start of exogenous feeding, marking thereby the end of the larval period.  相似文献

Resistance of Brahman and Simmentaler cattle to southern African ticks   总被引:1,自引:0,他引:1  
Field collections of ticks from two breeds of cattle in the North Western Transvaal showed the common tick species to be Rhipicephalus appendiculatus, Amblyomma hebraeum, Rhipicephalus evertsi evertsi and Hyalomma marginatum rufipes. The number of these ticks was higher on Simmentaler than on Brahman cattle. Correlation was found between the number of ticks on the cattle and their serum gamma globulin levels indicating an increase in the production of antibodies. Similar correlation was also found between the number of ticks on the cattle and the number of eosinophils in their blood. Resistance was probably acquired by the hosts and, it appears, to a higher degree in Brahman than in Simmentaler.  相似文献

ABSTRACT A new devastating disease in the United States, commonly known as Sudden Oak Death, is caused by Phytophthora ramorum. This pathogen, which previously was described attacking species of Rhododendron and Viburnum in Germany and the Netherlands, has established itself in forests on the central coast of California and is killing scores of native oak trees (Lithocarpus densiflora, Quercus agrifolia, Q. kelloggii, and Q. parvula var. shrevei). The phytosanitary authorities in the European Union consider non-European isolates of P. ramorum as a threat to forest trees in Europe. To date, almost all European isolates are mating type A1 while those from California and Oregon are type A2. The occurrence of both mating types in the same region could lead to a population capable of sexual recombination, which could generate a new source of diversity. To prevent contact between these two populations, a rapid, reliable, and discriminating diagnostic test was developed to easily distinguish the two populations. Based on a DNA sequence difference in the mitochondrial Cytochrome c oxidase subunit 1 (Cox1) gene, we developed a single-nucleotide polymorphism (SNP) protocol to distinguish between isolates of P. ramorum originating in Europe and those originating in the United States. A total of 83 isolates of P. ramorum from Europe and 51 isolates from the United States were screened and all isolates could be consistently and correctly allocated to either the European or the U.S. populations using the SNP protocol.  相似文献

ABSTRACT Specific resistances to isolates of the ascomycete fungus Mycosphaerella graminicola, which causes Septoria tritici blotch of wheat, have been detected in many cultivars. Cvs. Flame and Hereward, which have specific resistance to the isolate IPO323, were crossed with the susceptible cv. Longbow. The results of tests on F1 and F2 progeny indicated that a single semidominant gene controls resistance to IPO323 in each of the resistant cultivars. This was confirmed in F3 families of Flame x Longbow, which were either homozygous resistant, homozygous susceptible, or segregating in tests with IPO323 but were uniformly susceptible to another isolate, IPO94269. None of 100 F2 progeny of Flame x Hereward were susceptible to IPO323, indicating that the resistance genes in these two cultivars are the same, closely linked, or allelic. The resistance gene in cv. Flame was mapped to the short arm of chromosome 3A using microsatellite markers and was named Stb6. Fifty-nine progeny of a cross between IPO323 and IPO94269 were used in complementary genetic analysis of the pathogen to test a gene-for-gene relationship between Stb6 and the avirulence gene in IPO323. Avirulence to cvs. Flame, Hereward, Shafir, Bezostaya 1, and Vivant and the breeding line NSL92-5719 cosegregated, and the ratio of virulent to avirulent was close to 1:1, suggesting that these wheat lines may all recognize the same avirulence gene and may all have Stb6. Together, these data provide the first demonstration that isolate-specific resistance of wheat to Septoria tritici blotch follows a gene-for-gene relationship.  相似文献

Increased transport and trade as well as climate shifts play an important role in the introduction, establishment and spread of new pathogens. Arguably, the introduction of bluetongue virus (BTV) serotype 8 in Benelux, Germany and France in 2006 is such an example. After its establishment in receptive local vector and host populations the continued spread of such a disease in a suitable environment will mainly depend on movement of infected vectors and animals. In this paper we explore how wind models can contribute to explain the spread of BTV in a temperate eco-climatic setting. Based on previous work in Greece and Bulgaria filtered wind density maps were computed using data from the European Centre for Medium-Range Weather Forecasts (ECMWF). Six hourly forward wind trajectories were computed at pressure levels of 850hPa for each infected farm as from the recorded onset of symptoms. The trajectories were filtered to remove wind events that do not contribute to possible spread of the vector. The suitable wind events were rastered and aggregated on a weekly basis to obtain weekly wind density maps. Next to this, cumulated wind density maps were also calculated to assess the overall impact of wind dispersal of vectors. A strong positive correlation was established between wind density data and the horizontal asymmetrical spread pattern of the 2006 BTV8 epidemic. It was shown that short (<5km), medium (5-31km) and long (>31km) distance spread had a different impact on disease spread. Computed wind densities were linked to the medium/long-distance spread whilst short range spread was mainly driven by active Culicoides flight. Whilst previous work in the Mediterranean basin showed that wind driven spread of Culicoides over sea occurred over distances of up to 700km, this phenomenon was not observed over land. Long-distance spread over land followed a hopping pattern, i.e. with intermediary stops and establishment of local virus circulation clusters at distances of 35-85km. Despite suitable wind densities, no long range spread was recorded over distances of 300-400km. Factors preventing spread Eastwards to the UK and Northwards to Denmark during the 2006 epidemic are discussed. Towards the east both elevation and terrain roughness, causing air turbulences and drop down of Culicoides, were major factors restricting spread. It is concluded that the proposed approach opens new avenues for understanding the spread of vector-borne viruses in Europe. Future developments should take into consideration both physical and biological factors affecting spread.  相似文献

This study compares the susceptibility to Haemonchus contortus infection in two breeds of sheep endemic to the Canary Islands, the Canaria Hair Breed sheep and the Canaria sheep. Sheep were experimentally infected with 20,000 larvae of H. contortus and animals killed on days 7 and 28 post-infection. No difference between sheep breeds were detected in immature worm counts at days 7 or 28 post-infection. However, in comparison to the Canaria sheep breed, the Canaria Hair Breed sheep showed lower mean faecal egg counts, lower adult worm counts, lower number of eggs in utero and female worm stunting. Overall, these data suggest that the Canaria Hair Breed sheep has a greater resistance to H. contortus infection than Canaria sheep, and that this resistance may act at the level of the adult parasite.  相似文献

Parasite control in wild ruminants is based mainly on anthelmintics. The objective of the present study was to evaluate three parasite control programs in seven herds of captive wild ruminants in 3 consecutive years. In the first year, a biannual spring-summer treatment regime with fenbendazole at 7.5 mg/kg body weight p.o. for 3 days was applied. The next year, an early-season treatment program with three administrations of fenbendazole at the same dosage at 3-wk intervals was used. In the third year, an early-season treatment program with ivermectin (0.2 mg/kg p.o. for 3 days), applied three times at 5-wk intervals, was evaluated. Effectiveness of these control programs was assessed by fecal egg counts and by scores of body condition and fecal consistency at weekly intervals. Involved animal species were Arabian oryx (Oryx leucoryx), scimitar-horned oryx (Oryx dammah), slender-horned gazelle (Gazella leptoceros), Soay sheep (Ovis aries soay), ibex (Capra ibex), red deer (Cervus elaphus hippelaphus), and Nelson's elk (Cervus elaphus nelsoni). With the spring-summer regime, fecal egg counts remained low during the first 5 mo, but from September onward, they slowly increased to significant levels in all seven herds. The early-season program with fenbendazole resulted in fecal egg counts near zero during the entire year in four herds but a significant egg shedding from August onward was noted in Arabian oryx, scimitar-horned oryx, and Soay sheep. The early-season program with ivermectin resulted in very low to zero egg shedding in gazelle, adult Soay sheep, ibex, red deer, and Nelson's elk during the entire grazing season, but failed to prevent high shedding in October in Arabian oryx and scimitar-horned oryx. High contamination of grassy pastures with infective larvae/eggs in the previous year or before the first treatment may explain failure of the treatment regimes in some herds.  相似文献

Morphological abnormalities in wild animals can be indicators of the underlying health of a population and may be determined through routine photographic surveys. Here, we assess unusual rostrum conditions in Indian Ocean humpback dolphins (Sousa plumbea) inhabiting South African coastal waters to understand the rate of prevalence of abnormal rostrums and formulate hypotheses on potential causes. Photographic data were collated from systematic boat surveys and opportunistic sightings, obtained between April 1998 and March 2021 in various regions along the South African coast. Overall, 31 unique individuals were found with abnormal rostrum conditions, varying from slight misalignments to severe wounds and/or aberrant morphologies. In most cases, injuries were likely caused by natural events during the animal's life history such as interactions with sharks and/or reef-associated hunting strategies. Mark–recapture data indicated that individuals had survived with these injuries for up to 10 years. This study reports the highest incidence of rostrum abnormalities in the species. As numbers reflect only those that have survived their injuries, they are considered a minimum estimate. A better understanding of the cause(s) of these injuries is important given the endangered status of this species.  相似文献
