Comparative stability studies of poly(2-methyl-2-oxazoline) and poly(ethylene glycol) brush coatings

Non-fouling surfaces that resist non-specific adsorption of proteins, bacteria, and higher organisms are of particular interest in diverse applications ranging from marine coatings to diagnostic devices and biomedical implants. Poly(ethylene glycol) (PEG) is the most frequently used polymer to impart surfaces with such non-fouling properties. Nevertheless, limitations in PEG stability have stimulated research on alternative polymers that are potentially more stable than PEG. Among them, we previously investigated poly(2-methyl-2-oxazoline) (PMOXA), a peptidomimetic polymer, and found that PMOXA shows excellent anti-fouling properties. Here, we compare the stability of films self-assembled from graft copolymers exposing a dense brush layer of PEG and PMOXA side chains, respectively, in physiological and oxidative media. Before media exposure both film types prevented the adsorption of full serum proteins to below the detection limit of optical waveguide in situ measurements. Before and after media exposure for up to 2 weeks, the total film thickness, chemical composition, and total adsorbed mass of the films were quantified using variable angle spectroscopic ellipsometry (VASE), X-ray photoelectron spectroscopy (XPS), and optical waveguide lightmode spectroscopy (OWLS), respectively. We found (i) that PMOXA graft copolymer films were significantly more stable than PEG graft copolymer films and kept their protein-repellent properties under all investigated conditions and (ii) that film degradation was due to side chain degradation rather than due to copolymer desorption. 相似文献

Seven new deoxyisoaustamide derivatives (1–7) together with known compounds (8–10) were isolated from the coral-derived fungus Penicillium dimorphosporum KMM 4689. Their structures were established using spectroscopic methods, X-ray diffraction analysis and by comparison with related known compounds. The absolute configurations of some alkaloids were determined based on CD and NOESY data as well as biogenetic considerations. The cytotoxic and neuroprotective activities of some of the isolated compounds were examined and structure-activity relationships were pointed out. New deoxyisoaustamides 4–6 at concentration of 1 µM revealed a statistical increase of PQ(paraquat)-treated Neuro-2a cell viability by 30–39%. 相似文献

The wood bulk density, bark mass and decomposition rate constants of cut stumps of the main European boreal tree species were assessed along a 40-year chronosequence of clear-felled sites with and without prescribed burning. Using the single exponential model, the annual decomposition rate constants k of above-ground stumps were calculated as 0.048, 0.052 and 0.068 year−1 for Scots pine (Pinus sylvestris), Norway spruce (Picea abies) and birch (Betula sp.), respectively. Bark decomposed faster than wood and bark fragmentation increased the rate of decomposition. There was a significant negative effect of burning on decomposition rate for pine wood, and for pine and spruce bark but not for spruce and birch wood or for birch bark. The decomposition of bark of all species was slower with larger diameter stumps but only slightly slower in the case of birch wood. Our results suggest (i) using different decomposition rate constants for wood and bark, (ii) taking into account fragmentation as it greatly increases the volume loss, and (iii) adjusting of k in carbon dynamics studies on burned sites. Such refinements to estimates of coarse woody debris decomposition constants could aid in identification of ecosystems and management scenarios necessary to maximize carbon storage and conserve biodiversity. Prescribed burning for restoration purposes decreases decomposition rates and consequently ensures longer persistence of stumps for maintaining biodiversity in intensively managed forests. 相似文献

Twelve Galago senegalensis from the Moscow Zoo were presented with papular to nodular (2-11 mm) lesions on the pinnae, containing a white, waxy material. Microscopic examination revealed large numbers of mites consistent with the morphology of Demodex spp. mites. Nine animals were treated with ivermectin, 600 μg/kg/day topically, orally or subcutaneously for 3-10 months, while one remained untreated. All the treated animals achieved clinical remission. The control animal was still affected and died 11 months later due to pneumonia and possible eosinophilic leukaemia. No adverse effects were noted in any animals during the treatment. No animal relapsed in the 13-19 months follow-up period. To the authors' knowledge, this is the first report of demodicosis in G. senegalensis. The use of ivermectin in G. senegalensis was safe, although its effectiveness in the treatment of demodicosis needs further investigation. 相似文献

Efficacy and mechanism of action of marine alkaloid 3,10-dibromofascaplysin (DBF) were investigated in human prostate cancer (PCa) cells harboring different levels of drug resistance. Anticancer activity was observed across all cell lines examined without signs of cross-resistance to androgen receptor targeting agents (ARTA) or taxane based chemotherapy. Kinome analysis followed by functional investigation identified JNK1/2 to be one of the molecular targets of DBF in 22Rv1 cells. In contrast, no activation of p38 and ERK1/2 MAPKs was observed. Inhibition of the drug-induced JNK1/2 activation or of the basal p38 activity resulted in increased cytotoxicity of DBF, whereas an active ERK1/2 was identified to be important for anticancer activity of the alkaloid. Synergistic effects of DBF were observed in combination with PARP-inhibitor olaparib most likely due to the induction of ROS production by the marine alkaloid. In addition, DBF intensified effects of platinum-based drugs cisplatin and carboplatin, and taxane derivatives docetaxel and cabazitaxel. Finally, DBF inhibited AR-signaling and resensitized AR-V7-positive 22Rv1 prostate cancer cells to enzalutamide, presumably due to AR-V7 down-regulation. These findings propose DBF to be a promising novel drug candidate for the treatment of human PCa regardless of resistance to standard therapy. 相似文献

The selective inhibition technique by specific antibiotics (streptomycin, cycloheximide) applied to substrate-induced respiration (SIR) measurement was used to test the relative contribution of fungi to bacteria (F/B ratio) to the overall microflora-induced activity in soils of European Russia. Investigated soils covered a wide climatic transect and different ecosystem types including managed vs. natural ecosystems. Before direct comparison among sites, the antibiotic inhibition technique was optimized for soil characteristics. Once the optimal concentration was set, the combined effect of the two antibiotics resulted in average 60% inhibition of SIR. The analyzed sites (in total 47) including various biomes (tundra, middle taiga, southern taiga, subtaiga, dark coniferous forests outside the boreal region, steppe, mountain forests and arable sites), were characterized by a wide range of soil pHw (3.95–7.95), soil organic carbon (0.69–24.08%), soil microbial biomass carbon (149–5028 µg C g?1 soil) and soil basal respiration (0.24–8.28 µg CO2-C g?1 soil h?1). In all the analyzed sites, a predominance of fungal over bacteria activity was observed with F/B ratios always higher than one (4.9 on average). The origin of cultivated potatoes has remaining questions. In this study, 237 accessions of all cultivated species and 155 accessions of wild species closely related to cultivated potatoes, including their putative ancestors, were analyzed using 15 plastid microsatellites (SSRs) to investigate genetic diversity and their relationships with the wild species. We here used polymorphic plastid SSRs we developed from potato plastid genome sequences as well as already known plastid SSR markers. All 15 loci were polymorphic and identified a total of 127 haplotypes. Dramatic decreases in levels of genetic diversity were revealed in landraces in comparison with wild ancestor species. The plastid SSR results showed a decrease in haplotype number and diversity from Peru to both north and south. Phylogenetic analysis revealed two distinct groups. One of them, group A, contained the majority of accessions of cultivated species of the Solanum tuberosum Andigenum group including all accessions of cultivated diploid and triploid cytotypes of this group (S. chaucha, S. phureja, and S. stenotomum by a former taxonomic system) and most of tetraploid accessions of the S. tuberosum Andigenum group (S. tuberosum subsp. andigenum), and the majority of accessions of wild ancestors from the northern members of the S. brevicaule complex. Another group B comprised most of the wild species accessions and almost exclusively hybrid cultivated species which have introgressed plastid genomes from the other wild gene pools. Lack of clustering of traditional cultivated species (as used above) support a revised group classification of S. tuberosum. 相似文献
