Background: MDM2 (Murine Double Minute2) is an oncoprotein that inhibits the P53 activity. Overexpression of MDM2 gene has been reported in several human tumors. In the present study, we aimed to evaluate the impact of 40-bp insertion/deletion (ins/del) polymorphism on the promoter of MDM2 and susceptibility to breast cancer in a sample of Iranian population. Methods: This case-control study was carried out on 236 patients with breast cancer and 203 healthy individuals. Genomic DNA was extracted from the whole blood by the salting-out method. The 40-bp ins/del polymorphism was determined by using polymerase chain reaction. Results: The findings indicated that MDM2 ins/del variant increased the risk of breast cancer in co-dominant- (odds ratio [OR] = 2.09, 95% CI = 1.14-3.85, P = 0.018, del/del vs. ins/ins), dominant- (OR = 1.49, 95% CI = 1.02-2.18, P = 0.038, ins/del + del/del vs. ins/ins), and recessive- (OR = 1.86, 95% CI = 1.03-3.34, P = 0.038, del/del vs. ins/ins + ins/del) tested inheritance models. The del allele increased the risk of breast cancer (OR = 1.48, 95% CI = 1.11-1.98, P = 0.008) compared with ins allele. Conclusions: Our result revealed that 40-bp ins/del polymorphism in the promoter of MDM2 increased the risk of breast cancer in an Iranian population. Effects of the heavy metal copper(Cu), the metalloid arsenic(As), and the antibiotic oxytetracycline(OTC) on bacterial community structure and diversity during cow and pig manure composting were investigated. Eight treatments were applied, four to each manure type, namely cow manure with:(1) no additives(control),(2) addition of heavy metal and metalloid,(3) addition of OTC and(4) addition of OTC with heavy metal and metalloid; and pig manure with:(5) no additives(control),(6) addition of heavy metal and metalloid,(7) addition of OTC and(8) addition of OTC with heavy metal and metalloid. After 35 days of composting, according to the alpha diversity indices, the combination treatment(OTC with heavy metal and metalloid) in pig manure was less harmful to microbial diversity than the control or heavy metal and metalloid treatments. In cow manure, the treatment with heavy metal and metalloid was the most harmful to the microbial community, followed by the combination and OTC treatments. The OTC and combination treatments had negative effects on the relative abundance of microbes in cow manure composts. The dominant phyla in both manure composts included Actinobacteria, Bacteroidetes, Firmicutes, and Proteobacteria. The microbial diversity relative abundance transformation was dependent on the composting time. Redundancy analysis(RDA) revealed that environmental parameters had the most influence on the bacterial communities. In conclusion, the composting process is the most sustainable technology for reducing heavy metal and metalloid impacts and antibiotic contamination in cow and pig manure. The physicochemical property variations in the manures had a significant effect on the microbial community during the composting process. This study provides an improved understanding of bacterial community composition and its changes during the composting process. 相似文献

Analyses of the sorption of five polysaccharide cleaving soil enzymes by Na-homoionic clay minerals revealed a dependency on the cation exchange capacity of the clays: montmorillonite displayed the highest sorbing capacity, followed by palygorskite and kaolinite. Variation of the experimental conditions using cellulase and montmorillonite showed a maximum sorption at pH 4.8 and temperatures between 25°C and 45°C. Urea had no effect on the sorption, whereas humic acids and acetylation of enzymes lead to a decrease. No sorption was observed in the presence of bivalent cations. A discussion of possible binding mechanisms lead to the conclusion that interactions can be ascribed to van der Waals forces rather than to ionic reactions. It is assumed that the enzymes penetrate into the interlayers of montmorillonite, but not of palygorskite and kaolinite. 相似文献

In January 2011, symptomatic chickpea and faba bean plants were observed in fields located in the Gezira state (Sudan). Faba bean plants showed yellowing and stunting, whereas chickpea plants presented yellowing, reddening and little leaves. The disease etiology was investigated using nested polymerase chain reaction (PCR) with phytoplasma-specific primers which amplify a fragment of the 16S rRNA gene. Sequencing and restriction fragment length polymorphism (RFLP) analyses revealed that the tested phytoplasmas belonged to the group 16SrII. Phylogenetic analyses of the 16S rRNA gene of the obtained sequences indicated that the chickpea and faba bean phytoplasmas from Sudan were more closely related to the phytoplasmas subgroup 16SrII-D. To our knowledge, this is the first report of phytoplasmas from the group 16SrII-D infecting chickpea in Sudan, and faba bean worldwide. 相似文献

Whole genome sequencing of a copper resistant (CuR) black rot strain of Xanthomonas campestris pv. campestris (Xcc) isolated from a broccoli plant in Trinidad revealed a unique operon for copper resistance. The cop genes of strain Xcc-BrA1 were determined to be present on a 160 to 180 kb plasmid shown to be non-conjugative with other xanthomonads. While nucleotide comparison of a putative 8.0 Kbp copLABMGF gene cluster identified in Xcc-BrA1 genome did not reveal any homologous region with other known CuR Xanthomonas strains from diverse origins, the comparison of the translated amino acid sequence indicated similarity with X. citri, X. c. pv. citrumelonis and X. vesicatoria Cop proteins. Cloning of the copLAB gene cluster from Xcc-BrA1 conferred copper resistance to other copper-sensitive xanthomonads. Although Xcc-BrA1 harbors copLAB genes with similar sizes and organization and is able to grow on Cu-amended medium as other CuR xanthomonads, the phylogenetic analysis of nucleotide sequences indicates that the cop cluster in Xcc-BrA1 is unique and distantly related to other copLAB genes from Xanthomonas and Stenotrophomonas. The origin of copper resistance genes in Xcc-BrA1 is likely a result of horizontal gene acquisition from a still unknown phylloplane cohabitant. The findings of this study have implications for the management of crop diseases caused by CuR xanthomonads. Future studies could focus on and determining the distribution, overall importance and appropriate control measures for strains harbouring these unique genes. 相似文献

The effects of supplemental dietary phosphatidylcholine (PtCho) on the growth performance, survival and digestive enzyme activity of Caspian brown trout (Salmo trutta caspius Kessler 1877) alevins were investigated in this study. Alevin (initial weight 0.8 ± 0.12 g) was fed for 5 weeks with an isoproteic‐ and isolipidic‐formulated diet with increased levels of PtCho from 0 to 60 g kg?1 dry matter and decreasing levels of soybean oil. The increase in dietary PtCho up to 4% led to an increase in alevin final weight, suggesting that moderate PtCho levels are needed during this stage of Caspian brown trout. Survival was not affected by the dietary PtCho level. Phosphatidylcholine incorporation into the diet caused higher phospholipase A2‐specific activity. Phosphatidylcholine did not show a beneficial effect on the specific activity of amylase and protease. Lipase‐specific activity was significantly higher in the PtCho groups compared with the control group. The hepatosomatic index (HSI) was significantly influenced by the dietary PtCho level. The results of the present study indicated that the dietary supplementation of PtCho in the diet of Caspian brown trout alevin improved growth and lipolytic enzyme activity. 相似文献

Background
Recently, botulinum neurotoxin (BoNT)-derived recombinant proteins have been suggested as potential botulism vaccines. Here, with concentrating on BoNT type E (BoNT/E), we studied two of these binding domain-based recombinant proteins: a multivalent chimer protein, which is composed of BoNT serotypes A, B and E binding subdomains, and a monovalent recombinant protein, which contains 93 amino acid residues from recombinant C-terminal heavy chain of BoNT/E (rBoNT/E-HCC). Both proteins have an identical region (48 aa) that contains one of the most important BoNT/E epitopes (YLTHMRD sequence).Methods
The recombinant protein efficiency in antibody production, their structural differences, and their BoNT/E-epitope location were compared by using ELISA, circular dichroism, computational modeling, and hydrophobicity predictions.Results
Immunological studies indicated that the antibody yield against rBoNT/E-HCC was higher than chimer protein. Cross ELISA confirmed that the antibodies against the chimer protein recognized rBoNT/E-HCC more efficiently. However, both antibody groups (anti-chimer and anti-rBoNT/E-HCC antibodies) were able to recognize other proteins. Structural studies with circular dichroism showed that chimer proteins have slightly more secondary structures than rBoNT/E-HCC.Conclusion
The immunological results suggested that the above-mentioned identical region in rBoNT/E-HCC is more exposed. Circular dichroism, computational protein modeling and hydrophobicity predictions indicated a more exposed location for the identical region in rBoNT/E-HCC than the chimer protein, which is strongly in agreement with immunological results. We investigated the effects of soybean phosphatidylcholine (PtCho) on the growth, fatty acid (FA) profile, and body composition
of juvenile Caspian brown trout. Juvenile trout, initial average weight 0.8 ± 0.12 g, were fed semi-purified formulated diets
supplemented with 2, 4, and 6% PtCho for 5 weeks. Results showed that fish with diets containing 4 or 6% PtCho had significantly
greater growth. The FA profiles of both the polar lipids (PL) and neutral lipids (NL) in juvenile tissues were influenced
by dietary FA, although the effects of PtCho supplementation were more marked for PL than for NL. Fish fed PtCho in the diet
also had higher levels of 18:2n-6, 20:4n-6 and n-6, and significantly lower levels of docosahexaenoic acid and eicosapentaenoic
acid in both PL and NL. The lipid contents of whole-body tissues and liver were positively affected by PtCho percentage in
the diet. Specific growth rate, hepatosomatic index, visceralsomatic index, and protein efficiency ratio were also significantly
influenced by dietary PtCho. The results of our study indicate that dietary PtCho has a growth-promoting effect and that juvenile

The distribution variability of soil electrical conductivity (EC), pH, clay, sand, CaCO3, organic carbon (OC) and available potassium (K) in the Naqade region was investigated using a geostatistical method and Geographical Information System (GIS) technique. Two hundred and eighty-two topsoil (0–30 cm) samples were randomly collected and analyzed. pH and clay followed a normal distribution, whereas sand EC, CaCO3, OC and K were log-transformed. The highest variation was observed for soil EC, and the lowest for soil pH. In the variography analysis, spherical, exponential and gaussian models were best fit on experimental semivariograms. The minimum effective spatial autocorrelation was 1500 m for OC and the maximum effective spatial autocorrelation was 4000 m for sand and K. Strong spatial correlations were noted with sand and CaCO3 (<25%), whereas values were moderate for clay, EC, OC and K (25–75%). Ordinary kriging was utilized for the interpolation of estimated variable determinations in unsampled sites. It was found that soil properties in this study area were strongly influenced by both environmental and natural factors. The results can be used as a source of information for the development and implementation of any further land management and soil and water conservation plans. 相似文献
