This study evaluated the Mycobacterium tuberculosis protein antigen MPT-51, the trimeric antigen 85 (Ag85) complex, and Bacillus Calmette-Guérin (BCG) in an indirect ELISA to diagnose bovine tuberculosis (TB) from serum samples. Serum was collected from 208 intra-dermal tuberculin test (ITT)-positive and 54 ITT-negative animals from a region where bovine TB is endemic. Using the Ag85 and BCG antigens, the indirect ELISA was able to discriminate ITT-positive from ITT-negative animals. This level of discrimination was not achieved when using the MPT-51 antigen. The highest sensitivity (Se) and specificity (Sp) of the test was found when BCG was used (Se, 82%; Sp, 91%). Further work in different epidemiological settings and with larger numbers of animals will be required to validate these findings.  相似文献

This trial was conducted to evaluate if the effect of condensed tannin (CT) is associated with a true protein source on intake, ruminal and total digestibility, ruminal digestion rate, protein efficiency, and microbial efficiency in beef steers fed high concentrate diet (87% of DM). Four Bos indicus steers (407 ± 12 kg of BW) fitted with rumen cannula were assigned to a 4 × 4 Latin square design, arranged in a 2 × 2 factorial arrangement. Treatments consisted of either inclusion (0.4% of DM) or exclusion of condensed tannin (CT) from quebracho extract (76% of CT) with or without the use of soybean meal (SBM) as source of true protein. The level of inclusion was calculated to provide a daily intake of 1 g/10 kg of BW of CT. Intake of DM and nutrients was not affected (P > 0.10) by CT inclusion. However, there was an effect (P < 0.10) of CT inclusion on ether extract digestibility. An interaction (P < 0.10) was observed between CT and SBM on ruminal digestibility and digestion rate of crude protein (CP): when mixed with soybean meal, CT decreased the ruminal digestibility and, consequently, reduced the digestion rate of CP. Intake of CP increased (P < 0.10) with the inclusion of SBM. No differences in DM passage rate were observed (P > 0.10) among treatments. Effects of the interaction (P < 0.10) between CT and SBM were observed on flux of rumen undegradable protein (RUP), metabolizable protein (MP), and on the ratio MP:CP. In the presence of soybean meal, the addition of CT increased (P < 0.10) the flux of RUP and MP, and improved the ratio MP:CP. The yield of microbial protein on the abomasum and the microbial efficiency did not differ among treatments (P > 0.10). There was no difference (P > 0.10) on the pH, VFA, and ruminal ammonia (N-NH3) with the addition of condensed tannin. The N-NH3 increased and the ruminal pH decreased with the inclusion of soybean meal (P < 0.10). The utilization of condensed tannin as an additive in beef cattle diets with high level of concentrate and soybean meal as a source of true protein implies positive effects on crude protein utilization, decreasing digestion rate and ruminal digestibility of crude protein and consequently increasing the levels of metabolizable protein, with no changes in the ruminal fermentation parameters.  相似文献

Beer contains a very complex mixture of nutrients, which in this work are identified to some extent by high-field high-resolution nuclear magnetic resonance (NMR) one- and two-dimensional methods. The (1)H NMR spectrum of beer shows a predominance of strongly overlapped peaks arising from several carbohydrates. Minor components are clearly observed both in the aliphatic and in the aromatic regions of the spectrum. With the aid of two-dimensional methods, spectral assignment was carried out, enabling the identification of approximately 30 compounds and identifying about the same number of spin systems for further assignment. The variability of the spectral profile of beers differing in type and label was studied by principal component analysis (PCA), and it was found that, although some distinction is achieved on the basis of the aliphatic and sugar compositions, clearer separation between ales and lagers is obtained by PCA of the aromatic profiles alone. The potential of this technique as a rapid and informative quality control tool is discussed.  相似文献

The use of high-resolution NMR and high-resolution diffusion-ordered spectroscopy (DOSY) for the characterization of selected Port wine samples of different ages with the aim of identifying changes in composition is described. Conventional 1D and 2D NMR methods enabled the identification of about 35 compounds, including minor components such as some medium-chain alcohols, amino acids, and organic acids. High-resolution (HR) DOSY extended sample characterization, increasing the number of compounds identified and NMR assignments made, by providing information on the relative molecular sizes of the metabolites present. Port wines of different ages were found to differ mainly in their content of (a) organic acids and some amino acids, (b) an unidentified possible disaccharide, and (c) large aromatic species. The relative amount of these last high Mw aromatics is seen to decrease significantly in the oldest wine, as expected from the known formation and precipitation of anthocyanin-based polymers during red wine aging.  相似文献

Millet is a grass that responds to potassium fertilization and Alfa 01 and Alfa 02 are potential sources of this nutrient. Thus, the aim of this study was to evaluate the potential of as potassium sources by measuring accumulation of this nutrient in millet. The crop was grown in Ustoxic Quartzipsamment (RQo) soil. The experiment was set up in randomized blocks with three repetitions. The treatments consisting of three sources (KCl, Alfa 02, Alfa 01), two rates (200 and 400 kg ha?1 K2O) and with no K fertilization (control). Potassium chloride was used as the standard to which the other sources of potassium were compared. Two consecutive crops of millet were grown in the same pots. The first crop received potassium source application but not the second. At the end of experiment, shoot dry matter production, shoot potassium concentration, shoot accumulated potassium, soil potassium and relative agronomic efficiency were determined. The relative agronomic efficiency, calculated from crops, was 100% for KCl, was 84% for Alfa 02 and was 11% for Alfa 01. In the shoot dry matter production and accumulated potassium only Alfa 01 was not statistically different of control. Millet absorbed potassium from the Alfa 02 source in both the first and second crops.  相似文献

During the last decade, the production of off‐season maize has increased in several regions of Brazil. Growing maize during this season, with sowing from January through April, imposes several climatic risks that can impact crop yield. This is mainly caused by the high variability of precipitation and the probability of frost during the reproduction phases. High production risks are also partially due to the use of cultivars that are not adapted to the local environmental conditions. The goal of this study was to evaluate crop growth and development and associated yield, yield components and water use efficiency (WUE) for maize hybrids with different maturity ratings grown off‐season in a subtropical environment under both rainfed and irrigated conditions. Three experiments were conducted in 2001 and 2002 in Piracicaba, state of São Paulo, Brazil with four hybrids of different maturity duration, AG9010 (very short season), DAS CO32 and Exceler (short season) and DKB 333B (normal season). Leaf area index (LAI), plant height and dry matter were measured approximately every 18 days. Under rainfed conditions, the soil water content in the deeper layers was reduced, suggesting that the extension of the roots into these layers was a response to soil water limitations. On average, WUE varied from 1.45 kg m−3 under rainfed conditions to 1.69 kg m−3 under irrigated conditions during 2001. The average yield varied from 4209 kg ha−1 for the hybrids grown under rainfed conditions to 5594 kg ha−1 under irrigated conditions during 2001. Yield reductions under rainfed conditions were affected by the genotype. For the hybrid DKB 333B with a normal maturity, yield was reduced by 25.6 % while the short maturity hybrid Exceler was the least impacted by soil water limitations with a yield reduction of only 8.4 %. To decrease the risk of yield loss, the application of supplemental irrigation should be considered by local farmers, provided that this practice is not restricted by either economic considerations or the availability of sufficient water resources.  相似文献

Infestations by rhabditiform nematodes and acarids of the genus Raillietia are considered the primary causes of external otitis in cattle in tropical regions. Recently, yeasts of the genus Malassezia have been associated with a relatively high percentage of otitis cases, but the occurrence of other yeasts and mycelial fungi has not yet been reported in the literature. This work studied the presence of fungi in the ear canal of 45 cattle with external parasitic otitis. The results were positive for yeasts of the genus Malassezia in 31 (68.9 %) of the 45 cultures in Mycosel medium supplemented with olive oil. The 45 cultures in Sabouraud dextrose medium revealed the growth of seven (15.5 %) yeasts of the genus Candida, five (11.1 %) Rhodotorulamucilaginosa, two (4.4 %) fungi of the genus Aspergillus and eight ‘Micelia sterilia’. Future studies may confirm and elucidate the importance of these agents in the aetiology of bovine otitis.  相似文献

The role of metallothionein (MT) and zinc (Zn) in hepatic copper (Cu) accumulation in calves from a region in North-West Spain has been investigated. In this region there is intensive pig farming, and animals with liver Cu concentrations above normal are not uncommon. Concentrations of hepatic MT were not related to Cu accumulation but were strongly dependent on the Zn status of the animal. When analysing the metal content bound to MT it was observed that Cu-MT values, in the same way as Zn-MT, were directly correlated with MT concentrations in the liver, indicating that although Cu is a poor inducer of MT synthesis, it can compete with Zn for MT binding sites. The ability of Cu to displace Zn from MT is highly dependent on the Cu:Zn ratio in the cell, Cu being the main metal in MT at the higher levels of Cu exposure in Galician cattle. In spite of this, the percentage of the total hepatic Cu bound to MT is very low in these animals, indicating that cattle have a very limited capacity to accumulate Cu-MT in the liver, and may therefore have a greater susceptibility to Cu toxicosis.  相似文献

Summary In this study we investigated the effect of the acetyl-L-carnitine (ALC) supplementation on the myenteric neurons of the jejunum of rats made diabetic at the age of 105 days by streptozotocin (35 mg/kg body weight). Four groups were used: non-diabetic (C), non-diabetic supplemented with ALC (CC), diabetic (D), diabetic supplemented with ALC (DC). After 15 weeks of diabetes induction the blood was collected by cardiac puncture to evaluate glycaemia and glycated haemoglobin. Next the animals were killed and the jejunum was collected and subjected to whole-mount preparation to evidence the myenteric neurons through the histochemical technique of the NADH-diaphorase. The neuronal counts were made in 80 microscopic fields, in tissue samples of five animals of each group. The profiles of the cell bodies of 1000 neurons per group were analysed. Diabetes induced a significant increase in the area of the cell body and decrease in the number of NADH-diaphorase positive myoenteric neurons. ALC suplementation to the diabetic group promoted smaller hypertrophic effects and less neuronal loss than in the myoenteric neurons of the diabetic rats, and in addition diminished the body weight decrease and reduced the fasting glycaemia.  相似文献
