Xanthohumol (XN) is the major prenylated chalcone of hops and hence an ingredient of beer. Despite many advances in understanding of the pharmacology of XN, one largely unresolved issue is its low bioavailability in the human organism. Also, not much is known about its actual concentrations and pharmacokinetics in liver and intestinal cells. Therefore, the uptake, intracellular distribution, and kinetics of XN were studied in various cell types, namely, hepatocellular carcinoma cells (HuH-7), hepatic stellate cells (HSC), primary cultured hepatocytes, and colorectal adenocarcinoma cells (Caco-2). Fluorescent microscopy allowed for the first time visualization and tracing of the uptake and intracellular distribution of XN. A rapid accumulation of XN concentrations that were up to >60-fold higher than the concentration present in the ambient culture medium was observed. Fluorescence recovery after photobleaching experiments revealed that most XN molecules are bound to cellular proteins, which may alter properties of cellular factors.  相似文献

European domestic pigs are derived from the European wild pig by genetic selection and differ in their muscular-growth ability. Thus, it was the aim of this study to investigate the consequences of genetic selection on muscle composition and fibre size in the gracilis muscle. Fibre typing based on the ATPase reaction revealed that this muscle in wild pigs is composed mainly of type-I and -IIa fibres, whereas, in domestic pigs, type-IIb fibres were predominant. For all fibre types, domestic pigs had about threefold larger fibres than wild pigs. Type-I fibres tended to be the smallest fibres in domestic pigs, but the largest fibres in wild pigs, which may be due to long-term training effects in the free-ranging animals. Giant fibres as an indicator of degeneration were obvious only in the domestic-pig samples. Their occurrence, as well as the larger fibre size and the predominance of type-IIb fibres in domestic pigs, may be ascribed to high concentrations of growth hormone. It is concluded that selection for muscular growth may favour metabolic dysbalances within the muscle and, therefore, degenerative processes.  相似文献

The effects of total gastrectomy in six young swine were followed and described during an experimental period up to 18 months. Two of the gastrectomized pigs (Nos. 1 and 2) were medicated with cycobemin every second week. Two other gastrectomized pigs (Nos. 11 and 14) were medicated with iron-dextran twice a week. Three non-gastrectomized swine receiving the same feed as the gastrectomized animals were observed as controls for up to eight months.One of the gastrectomized non-medicated pigs (No. 4) did not gain weight, while the other gastrectomized animals had a normal appearance and gained weight but less than the controls. The non-medicated as well as the cycobemin-medicated gastrectomized animals developed a microcytic hypochrome anemia corresponding to the anemia in iron deficiency. Histochemically loss of iron in the depots (liver, spleen, bone marrow and intestine) was demonstrated in the gastrectomized animals except those treated with iron-dextran. Histological and histochemical examinations of the nervous system did not show any abnormalities.  相似文献

Norway Spruce is the economically most important tree species in Europe and has been cultivated in plantations on a large-scale at low elevations, far outside its natural range. In the Bohemian Forest, it naturally occurs in pure stands above 1150 m a.s.l. and as a mixed tree species from 650 to 1150 m a.s.l. An understanding of natural distributions and the diversity along temperature gradients at various elevations is important for conservation, pest management, and predictions of future species assemblages by global warming. Here we investigated the species richness of canopy arthropods in spruce trees along a gradient from 300 to 1300 m a.s.l. using flight-interception traps. We analyzed species richness by combining diversity partitioning with a moving window approach after standardizing sample size per plot. Total richness decreased linearly as the elevation increased, which reflected declining temperatures and a declining regional species pool. Phytophages (herbivores excluding xylophages) were the most influenced. Richness did not peak at the transition zones of the three ecological elevation zones, neither for all species, nor for any of the separate functional groups. However, the proportion of both beetle and true bug spruce specialists significantly increased with elevation and actually doubled in richness above 1000 m a.s.l., where spruce is naturally dominating. Our results indicate that even planted spruce trees at lower elevations maintain high levels of species richness. Further climate warming will promote overall species richness, especially of phytophages, at all elevations. However, spruce specialists may be seriously threatened by global warming.  相似文献

As shown earlier, pig rations with high starch and purine content initiate mucosal hypertrophy by stimulating mitotic activity and DNA formation in the small intestine, whereas in the colon butyrate inhibits apoptosis and thus increases crypt depth. It was the aim of this study to combine these effects by targeting fat-coated butyrate into the small intestine where it usually does not occur, and to investigate effects on mucosal development and function. Three groups of five pigs were fed 3.6 kg/day of either a low-energy ration [deficit group, 6.6 MJ metabolizable energy (ME)/kg] or a high-energy ration (13.7 MJ ME/kg) that was supplemented with brewing yeast as a source of purines. The third ration was of high energy and contained purines and was additionally supplemented with coated butyrate (13.5 MJ ME/kg; 29 g calcium butyrate/kg). Rations were fed for 5 days. After killing, tissue samples were obtained from the proximal, medial and distal parts of jejunum for histology. Chyme samples were obtained from the ileum of all animals and used for sucrase determination. Villus size was not changed by feeding, but butyrate had an effect on plica height and area mainly in the medial jejunum. Odor emission from swine facilities is determined by microbial breakdown of amino acids or carbohydrates in the pig colon. It was the aim to influence apoptosis and thus amino acid availability for odor formation by feeding resistant starch (300 g kg(-1) feed) over the whole fattening period to 40 pigs. Concentrations of 12 key components (indoles, volatile fatty acids, methanethiol) were measured in feces and headspace over the slurry duct and compared to 40 normally fed controls in a separate compartment. Concentrations of substances resulting from amino acids were reduced in feces by 70% (indoles) and 8% (branched chain fatty acids) and in the headspace by 72% and 20%. Resistant starch only led to minor increases of straight chain fatty acid concentration. Maximal reduction occurred for 3-methyl-1H-indole (skatole) which is the main determinant of malodor so that the results point to promising strategies for reducing pig odor emission.  相似文献

Background

Only few pharmacologic compounds have been validated for treatment of atrial fibrillation (AF) in horses. Studies investigating the utility and safety of flecainide to treat AF in horses have produced conflicting results, and the antiarrhythmic mechanisms of flecainide are not fully understood.

Objectives

To study the potential of flecainide to terminate acutely induced AF of short duration (≥15 minutes), to examine flecainide‐induced changes in AF duration and AF vulnerability, and to investigate the in vivo effects of flecainide on right atrial effective refractory period, AF cycle length, and ventricular depolarization and repolarization.

Animals

Nine Standardbred horses. Eight received flecainide, 3 were used as time‐matched controls, 2 of which also received flecainide.

Methods

Prospective study. The antiarrhythmic and electrophysiologic effects of flecainide were based on 5 parameters: ability to terminate acute pacing‐induced AF (≥15 minutes), and drug‐induced changes in atrial effective refractory period, AF duration, AF vulnerability, and ventricular depolarization and repolarization times. Parameters were assessed at baseline and after flecainide by programmed electrical stimulation methods.

Results

Flecainide terminated all acutely induced AF episodes (n = 7); (AF duration, 21 ± 5 minutes) and significantly decreased the AF duration, but neither altered atrial effective refractory period nor AF vulnerability significantly. Ventricular repolarization time was prolonged between 8 and 20 minutes after initiation of flecainide infusion, but no ventricular arrhythmias were detected.

Conclusions and Clinical Importance

Flecainide had clear antiarrhythmic properties in terminating acute pacing‐induced AF, but showed no protective properties against immediate reinduction of AF. Flecainide caused temporary prolongation in the ventricular repolarization, which may be a proarrhythmic effect.  相似文献
