1. The effect of dialysis on the proacrosin/acrosin system and motility of turkey spermatozoa were examined after 24 and 48 h of liquid storage at 4°C.

2. Fifteen pools of semen diluted in extender were dialysed against Clemson Turkey Semen Diluent (dialysed semen) or stored in aerobic conditions (undialysed semen). Semen quality was assessed by measuring spermatozoa motility, amidase activity of spermatozoa suspension, spermatozoa extract and seminal plasma and anti-trypsin activity of seminal plasma.

3. Extracted amidase activity of dialysed semen was lower than undialysed by 28%. Higher values for speed parameters of spermatozoa were found in dialysed semen in comparison to undialysed, for example, 81.6 µm/s versus 75.0 µm/s for straight-line velocity (VSL), 114.7 µm/s versus 110.3 µm/s for curvilinear velocity (VCL) and 86.6 µm/s versus 79.8 µm/s for average path velocity (VAP).

4. It was concluded that dialysis caused lower amidase activity of spermatozoa and increased speed parameters of progressively motile turkey spermatozoa during storage. Lower extracted amidase activity of dialysed semen reflected better membrane integrity of dialysed semen and suggests that the proacrosin/acrosin system of dialysed spermatozoa is less susceptible to activation compared to undialysed semen.  相似文献

To evaluate protein and lipid requirement of South American catfish surubim (Pseudoplatystoma sp.) juveniles, nine semi‐purified diets containing three levels of protein (40%, 45% and 50%) and three levels of lipid (12%, 16% and 20%) were tested. After 8‐week feeding trial, body weight increase averaged 2124.3 ± 295.7%. Growth performance was significantly affected by dietary level of protein (P < 0.05). At the 40% protein level, increasing level of dietary lipid had a positive effect on final individual mean weight (protein sparing effect). Whole body protein and moisture contents were affected by the dietary level of lipid (P < 0.05). Whole body lipid content positively correlated with the level of dietary lipid (P < 0.05). Cannibalism related mortality was observed despite rearing fish in 24 h dark. Fatty acid composition of fish was affected by the dietary lipid level (P < 0.05). Polyunsaturated fatty acids increased with the increasing level of dietary lipid while saturated fatty acids and monounsaturated fatty acids decreased. Trypsin activity in the digestive tract of surubim was influenced by dietary levels of protein and lipid (P < 0.05). Our preliminary results suggest that the optimum protein/lipid ratio might be close to 45/16% for surubim juveniles.  相似文献

Background

Annotation of proteins with gene ontology (GO) terms is ongoing work and a complex task. Manual GO annotation is precise and precious, but it is time-consuming. Therefore, instead of curated annotations most of the proteins come with uncurated annotations, which have been generated automatically. Text-mining systems that use literature for automatic annotation have been proposed but they do not satisfy the high quality expectations of curators.

Results

In this paper we describe an approach that links uncurated annotations to text extracted from literature. The selection of the text is based on the similarity of the text to the term from the uncurated annotation. Besides substantiating the uncurated annotations, the extracted texts also lead to novel annotations. In addition, the approach uses the GO hierarchy to achieve high precision. Our approach is integrated into GOAnnotator, a tool that assists the curation process for GO annotation of UniProt proteins.

Conclusion

The GO curators assessed GOAnnotator with a set of 66 distinct UniProt/SwissProt proteins with uncurated annotations. GOAnnotator provided correct evidence text at 93% precision. This high precision results from using the GO hierarchy to only select GO terms similar to GO terms from uncurated annotations in GOA. Our approach is the first one to achieve high precision, which is crucial for the efficient support of GO curators. GOAnnotator was implemented as a web tool that is freely available at http://xldb.di.fc.ul.pt/rebil/tools/goa/.

Data on the biomass and productivity of southeast Asian tropical forests are rare, making it difficult to evaluate the role of these forest ecosystems in the global carbon cycle and the effects of increasing deforestation rates in this region. In particular, more precise information on size and dynamics of the root system is needed. In six natural forest stands at pre-montane elevation (c. 1000 m a.s.l.) on Sulawesi (Indonesia), we determined above-ground biomass and the distribution of fine (d < 2 mm) and coarse roots (d > 2 mm), estimated above- and below-ground net production, and compared the results to literature data from other pre-montane paleo- and neotropical forests. The mean total biomass of the stands was 303 Mg ha−1 (or 128 Mg C ha−1), with the largest biomass fraction being recorded for the above-ground components (286 Mg ha−1) and 11.2 and 5.6 Mg ha−1 of coarse and fine root biomass (down to 300 cm in the soil profile), resulting in a remarkably high shoot:root ratio of c. 17. Fine root density in the soil profile showed an exponential decrease with soil depth that was closely related to the concentrations of base cations, soil pH and in particular of total P and N. The above-ground biomass of these stands was found to be much higher than that of pre-montane forests in the Neotropics, on average, but lower compared to other pre-montane forests in the Paleotropics, in particular when compared with dipterocarp forests in Malesia. The total above- and below-ground net primary production was estimated at 15.2 Mg ha−1 yr−1 (or 6.7 Mg C ha−1 yr−1) with 14% of this stand total being invested below-ground and 86% representing above-ground net primary production. Leaf production was found to exceed net primary production of stem wood. The estimated above-ground production was high in relation to the mean calculated for pre-montane forests on a global scale, but it was markedly lower compared to data on dipterocarp forests in South-east Asia. We conclude that the studied forest plots on Sulawesi follow the general trend of higher biomasses and productivity found for paleotropical pre-montane forest compared to neotropical ones. However, biomass stocks and productivity appear to be lower in these Fagaceae-rich forests on Sulawesi than in dipterocarp forests of Malesia.  相似文献

Norway spruce (Picea abies (L.) Karst.) is an economically important, but relatively drought-sensitive tree species that might suffer from increasing drought intensities and frequencies, which are predicted to occur in parts of central Europe under future climatic change. In a throughfall exclusion experiment using sub-canopy roofs, we tested the hypothesis that enhanced drought leads to an increase in fine root mortality, and also to a higher, subsequent fine root growth. Fine root production and mortality were assessed using two independent approaches, sequential soil coring (organic layer) and direct observations in minirhizotrons (organic layer plus upper mineral soil). Six weeks of throughfall exclusion resulted in mild drought stress, i.e. a reduction in average soil moisture from 20 to 12 vol.% during the treatment. Based on the sequential coring data, experimental drought did not result in significant changes in fine root biomass during the 6-week treatment period, but caused an increase in fine root mortality by 61% in the 6 weeks following the drought treatment. Remarkably, fine root production showed a synchronous increase in this period, which more than compensated for the loss due to increased mortality. The minirhizotron data confirmed that the drought treatment increased fine root loss in the organic layer. Based on this method, however, root loss occurred during the drought period and was not compensated by increased root production. The mild drought stress was mainly restricted to the organic layer and did not significantly influence fine root dynamics in the mineral soil. We calculated that the drought event resulted in an extra input of about 28 g C m−2 and 1.1 g N m−2 to the soil due to increased fine root mortality. We conclude that even periods of mild drought significantly increase fine root mortality and the associated input of root-derived C to the soil organic matter pool in temperate Norway spruce forests.  相似文献

Endocrine-disrupting chemicals (EDCs) are widespread used and can interfere on hormone regulation with adverse consequences for both biota and human. Vitellogenin (vtg) is a yolk precursor protein synthesized by the liver in response to estrogen. In order to characterize the vtg of tropical fish Rhamdia quelen and establish a molecular biomarker, adult male individuals were exposed to 17-β-estradiol (E2) for vtg induction and anti-R. quelen vtg polyclonal antibodies production. Vitellogenic female fish were used as positive control group. E2-induced vtg was characterized as a glycolipophosphoprotein of high molecular mass with peptide mass fingerprint very similar in E2-exposed male and vitellogenic female fish. A polyclonal serum containing anti-R. quelen vtg antibodies was produced and showed high specificity and sensibility to detect the vtg of three fish species: R. quelen, Piaractus mesopotamicus and Prochilodus lineatus. Wildlife and laboratory studies reported that EDCs released into the environment may alter the levels of plasma vtg in male fish, making this protein a valuable biomarker of xenoestrogens exposure. Then, we propose the use of anti-R. quelen vtg as a tool for biomonitoring studies and water quality assessment in Brazil and South American countries where the three fish species occur.  相似文献
