在单因素试验基础上,采用正交试验优化红叶石楠绿叶中类胡萝卜素的提取工艺条件,并用柱层析分离纯化粗提液中的类胡萝卜素。结果表明,在40℃下以丙酮∶乙醇=1∶1为提取剂,料液比1∶10,浸提时间1.5 h,类胡萝卜素含量可达51.26μg/g。该类胡萝卜素粗提液经过柱层析法初步分离纯化,纯化倍数可达1.41。 相似文献

北川白山羊是在特定生态条件下未经系统繁育而形成的优良地方山羊品种,在生长发育、繁殖和抗逆等性状方面具有突出优势。通过对122只北川白山羊个体基因组的扩增,结果从80条RAPD引物中选出12条多态引物,北川白山羊群体平均遗传相似率和遗传多样性指数分别为0.859 8±0.077 6和0.919 8,表明北川白山羊品种具有一定的遗传分化和较丰富的遗传多样性。在12条多态性引物中,SBS06同时对体质量(P=0.028)、体高(P=0.017)和体长(P=0.037)具有显著影响,SBS02对体质量(P=0.033)和体高(P=0.034)具有显著效应,所以推断影响体质量和体高性状的QTL基因座可能与RAPD标记SBS06和SBS02相连锁,可以应用于北川白山羊体质量和体高性状的标记辅助选择以及相应主效基因的进一步研究。 相似文献

AIM: To observe the treatment effect and its immune regulation of human amnion epithelial cells (hAECs) on Alzheimer's disease (AD)-like pathology rat model. METHODS: The hAECs were isolated from amnion with trypsin digestion, and the phenotype of hAECs was analyzed by flow cytometry. SD rats (n=48) were randomly divided into sham control group, model group, medium group and hAECs group. AD-like pathology rat model was induced by bilateral intraventricular injection of lipopolysaccharide (LPS). hAECs (5×105) were injected into the hippocampus of the AD-like pathology rats. At 2 weeks after transplantation, the animals were tested by Morris water maze to observe the function of learning and memory. The pathological change of the brain was observed by HE staining. The expression of amyloid β-protein 42(Aβ42) and Tau protein and the level of acetylcholine (ACh) in the injury brain were determined by immunohistochemistry. The survival and differentiation of hAECs in the hippocampus were measured by immunofluorescent technique. The percentages of lymphocyte subsets in the peripheral blood mononuclear cells were analyzed by flow cytometry. The contents of serum cytokines were detected by cytometric bead array. RESULTS: Compared with model group and medium group, hAECs group showed shortened escape latency (P<0.01), increased frequency of going through the platform (P<0.05), reduced loss of hippocampal neurons, decreased expression of Tau protein and Aβ42 in the hippocampus (P<0.05), increased ACh level in the hippocampus (P<0.05), decreased percentages of Th1 and Th17 subsets, increased percentages of Th2 and Treg cells (P<0.05), decreased concentrations of IFN-γ and IL-2 in the serum, and increased concentration of IL-4(P<0.05). CONCLUSION: hAECs improve the cognitive learning and memory function and alleviate pathologic damage of hippocampus through immune regulation in AD-like pathology rats. 相似文献

The aim of this work was to assess the effect of applying three different doses of fluxapyroxad on microbial activity, community structure and functional diversity as measured by respiration, microbial biomass C, phospholipid fatty acid(PLFA) and community-level physiological profiles(CLPPs). Our results demonstrated that substrate-induced respiration(on day 15) and microbial biomass C(on days 7 and 15) were inhibited by fluxapyroxad, but stimulation was observed thereafter. In contrast, fluxapyroxad addition increased the basal respiration and metabolic quotients(q CO2) and respiratory quotients(QR). Analysis of the PLFA profiles revealed that the total and bacterial biomass(both Gram-positive bacteria(GP) and Gram-negative bacteria(GN)) were decreased within the initial 15 days, whereas those as well as the GN/GP ratio were increased at days 30 and 60. F luxapyroxad input decreased the fungi biomass but increased the bacteria/fungi ratio at all incubation time. Moreover, high fluxapyroxad input(75 mg fluxapyroxad kg–1 soil dry weight) increased the microbial stress level. A principal component analysis(PCA) of the PLFAs revealed that fluxapyroxad treatment s ignificantly shifted the microbial community structure, but all of the observed effects were transient. Biolog results showed that average well color development(AWCD) and functional diversity index(H′) were increased only on day 60. I n addition, the dissipation of fluxapyroxad was slow in soil, and the degradation half-lives varied from 158 to 385 days depending on the concentration tested. 相似文献

本文以白羊草(Bothriochloa ischaemum)为材料,利用其目前已有的转录组序列,通过搜索、比对、功能域多态性分析、进化树构建和基因表达分析等手段,对白羊草R2R3-MYB型转录因子进行挖掘和干旱胁迫下的表达模式分析。结果从白羊草挖掘出43个R2R3-MYB转录因子,它们具有高度保守的[W]-X(19)-[W]-X(19)-[W]-X(n)-[W]-X(18)-[W]结构,主要参与生长发育、次生代谢和逆境响应等生物过程。通过进一步的分析,本研究筛选出8个可能参与干旱胁迫响应的R2R3-MYB转录因子。这为白羊草R2R3-MYB基因的功能研究和开发利用奠定了基础。 相似文献
