A gold-immunochromatographic test-strip kit is used for the detection of IgG antibodies against the nucleocapsid protein of Avian Influenza Virus (AIV). Compared with the "gold standard", i.e. hemagglutination inhibition (HI) and agar gel immunodiffusion (AGID) assays, the gold-immunochromatographic test strip has many advantages, such as high specificity, high sensitivity, convenience, is rapid and has low cost. The gold-immunochromatographic test strip provides a unique tool for the on-site surveillance and diagnosis of Avian Influenza.  相似文献

AIM: To investigate whether gold nanoparticles (GNPs) reverses adriamycin (ADM), resistance of human hepatocellular carcinoma drug-resistant cell line HepG2/ADM and to explore the potential mechanism. METHODS: The sensitivities of HepG2 cells and HepG2/ADM cells to ADM were tested by MTT assay before and after GNPs pretreatment. The apoptotic rate was examined by flow cytometry. The concentration of ADM in HepG2/ADM or HepG2 cells was determined by ultraviolet-visible spectrophotometer. The content of glutathione (GSH) in HepG2/ADM or HepG2 cells by DTNB method. RESULTS: The half maximal inhibitory concentrations (IC50) of ADM for HepG2/ADM cells were(29.46±1.73) mg/L and (15.18±0.85) mg/L before and after GNPs pretreatment,respectively. The IC50 of ADM for HepG2 cells was (9.16±2.03) mg/L before pretreatment. The apoptotic rate in GNPs+ADM group was higher than that in ADM group (P<0.05). The concentration of ADM in HepG2/ADM group was lower than that in HepG2 group (P<0.01). After GNPs pretreatment, the concentration of ADM in HepG2/ADM cells was higher than that before pretreatment. The content of GSH in HepG2/ADM group was higher than that in HepG2 group (P<0.01). After GNPs pretreatment, the content of GSH in the HepG2/ADM cells was lower than that before pretreatment. CONCLUSION: Gold nanoparticles can reverse ADM resistance of human hepatocellular carcinoma drug-resistant cell line HepG2/ADM, reduce the content of GSH and increase the concentration of ADM in HepG2/ADM cells.  相似文献

AIM: To investigate the effect of rhynchophylline (Rhy) on blood pressure, cardiac hypertrophy and myocardial fibrosis in spontaneously hypertensive rats (SHR). METHODS: Spontaneously hypertensive rats were randomly divided into model group, high dose (10 mg·kg-1·d-1) and low dose (2.5 mg·kg-1·d-1) group of rhynchophylline, captopril group (17.5 mg·kg-1·d-1). Wistar-Kyoto rats were used as normal control. Respectively, systolic blood pressure was measured by tail cuff every 2 weeks. After 10 weeks, heart weight index and left ventricular weight index were calculated. The myocardial hydroxyproline and plasma angiotensin Ⅱ were detected. Moreover, basic myocardial histopathological changes and myocardial collagen fibres were observed by HE staining and Masson staining, respectively. The protein expression of TGF-β1 and Smad3 in the myocardium was measured by the methods of immunohistochemistry and Western blot. RESULTS: Compared with SHR model group, Rhy significantly reduced blood pressure (P<0.05), the levels of HYP in the myocardium (P<0.05) and the levels of AngⅡ in the plasma (P<0.01). The pathological damages of the myocardial tissues and collagen deposition were attenuated. The protein expression of TGF-β1 and Smad3 was significantly reduced by the treatment with Rhy (P<0.01). CONCLUSION: Rhynchophylline reduces blood pressure and adjusts to improve ventricular remodeling of SHR. The mechanism may be involved in the TGF-β1/Smad pathway and reducing AngⅡ content.  相似文献

AIM: To explore the effects of eplerenone on the expression and activity of aortic endothelial nitric oxide synthase(eNOS) in high salt-induced hypertensive rats.METHODS: Male Wistar rats(4 week old, weighting 50~60 g) were randomly divided into control group, high-salt diet group and eplerenone group. The rats in control group were fed with ordinary rodent animal diet, the rats in high-salt group and eplerenone group were exposed to 5% salt diet for 16 weeks and administrated with the same dosage of saline or eplerenone(40 mg·kg-1·d-1) by gavage for 4 weeks, respectively. Systolic blood pressure(SBP) was measured by tail-cuff every 2 weeks. The rats were sacrificed after 16 weeks and the thoracic aorta was collected. The aldosterone content in the aorta was measured by ELISA. The protein levels of mineralocorticoid receptor(MR) and eNOS were determined by Western blot. The activitie of constitutive NOS(cNOS) was measured by chemocolorimetry. The protein localization of eNOS, neuronal nitric oxide synthase(nNOS) and MR was observed by immunohistochemistry.RESULTS: A process of 8-week high-salt diet increased SBP gradually. SBP in the rats exposure to high salt for 16 weeks was significantly higher than that in control group(P<0.05). After 4 weeks of eplerenone treatment, SBP in the rats was significantly lower than that before treatment(P<0.05). Compared with control group, the aldosterone content in the aorta were significantly increased in high-salt diet group and eplerenone group(P<0.05), the expression level of MR also increased significantly(P<0.05). Compared with control group, both eNOS protein expression(P<0.05) and cNOS activity in high-salt diet group were significantly decreased(P<0.05). The protein expression of eNOS as well as cNOS activity in aorta increased significantly in eplerenone group compared with high-salt diet group(P<0.05).CONCLUSION: Aldosterone content in aorta of high-salt-induced hypertensive rats increases significantly. Aldosterone attenuates the protein expression of eNOS and reduces the enzyme activity through the activation of mineralocorticoid receptor. The selective mineralocorticoid receptor antagonist eplerenone enhances the protein expression of eNOS and its activity, thereby improves eNOS function.  相似文献

为从众多核桃品质的评价指标中找到主导的影响因子,达到用最少的指标获取最多核桃品质信息的目的,试验以赫章8个单株核桃为试材,通过对表征核桃物理化学特性的12项指标进行测定,利用因子分析,对众多指标进行降维处理,找出少数几个影响核桃品质的主导因子。结果表明:12项原始指标归属为3个相互独立的公因子,累计方差贡献率达到85.960%,其中,出仁率和壳厚度是重要的核桃品质的物理指标,蛋白质含量是重要的化学品质评价指标。核桃的理化品质与亲缘关系之间有一定的联系,利用因子分析结果进行聚类分析发现,1、5、6号核桃理化品质相似,可能具有较近的亲缘关系。4、7、8号单株单独成为3类,说明可能与其它单株的亲缘关系较远。研究结果可在一定程度为核桃资源的品质鉴定、优株鉴选、分类及亲缘关系的确定等提供理论依据。  相似文献

稗草(Echinochloa crusgalli)是稻田中的C4光合型杂草,为了探索稗草ppc基因(Eppc)对水稻遗传转化的可行性及其对光合速率的调节效应,首次将含有稗草根型磷酸烯醇式丙酮酸羧化酶(phosphoenolpyruvate carboxylase,PEPC)基因ppc c DNA的2个植物表达载体p Ubi-Eppc、p Rbc S-Eppc通过农杆菌介导法对水稻进行了遗传转化。对分化植株进行的PCR、RT-PCR、克隆测序和Western杂交等结果均表明稗草ppc基因已经整合到了水稻基因组中,并且在转录和翻译水平都得到了表达。转基因水稻PEPC活性和气体交换参数测定结果表明T0代多数植株的PEPC活性高于对照,最高达到了对照的5.85倍;T0代大多数转基因植株叶片的净光合速率(Pn)比对照提高了20.00%,最大地提高了47.16%,同时叶片水分利用效率(WUE)也得到增强;T6代大部分转化植株的PEPC活性及Pn仍保持高于对照,本研究表明C3根型ppc基因过量表达也可以提高水稻的Pn,且证明稗草PEPC对光合作用具有较强的调节作用。  相似文献

Owing to unique structure and characteristics of carbon nanotubes, much more are attracted attentions since they were discovered. In recent years, carbon nanotubes have been successfully synthesized by various methods. Especially, highly aligned carbon nanotubes were grown by chemical vapor deposition (CVD) and the oriented growth of carbon nanotubes became realized, resulting in many applications of carbon nanotubes and researches on them. In the work, the methods and mechanisms of orientation growth of carbon nanotubes were reviewed, and it was analyzed and discussed that different methods influenced on the process of growing carbon nanotubes. Simultaneously, it was emphatically analyzed that catalyst particles affected on orientation growth of carbon nanotubes during their growth.  相似文献

发芽纸是种子发芽试验必需的材料。国内首次研制成的专用种子发芽纸具有吸水快、持水性好、韧度大、厚度适中等特点。该纸无毒性、幼根不易穿透,且成本低。它既可衬垫于发芽容器中,更宜作发芽纸卷。试用结果表明,它能广泛适用于农作物、蔬菜、牧草、瓜、豆类等各种种子的发芽试验。在植物种子科学研究和生产实际中,经常需要进行种子发芽试验,一般的种子发芽试验常采用纸张作为发芽床。为使种子正常发芽和便于检查发芽结果,要求发芽纸有良好的理化特性。对此,国际种子检验协会对种子发芽纸的质地、灰分、韧度和吸水性能等有明确的规定指标,如要求纸张有足够强度,在发芽试验操作中不致撕破;质地要通透多孔,但幼根不能穿入纸中。国外已有不少国家都生产有专用的种子发芽纸,而国内至今还没有一种专门供种子发芽用的纸张,长期以来使用的是普通滤纸、卫生纸、乃至黄草纸等代用品。这些纸张在协调种子发芽所需的水气条件方面存有不少缺陷,其质地、强度或吸水性等都达不到要求。为了填补国内这一空白,浙江农业大学种子科学中心和浙江民丰造纸厂造纸研究所自1987年开始进行种子发芽纸的研制。经过多次反复研制和测试,已研制成功一种种子发芽纸。它厚度适中,既可平垫于玻皿之中,又可用作发芽纸卷,且成本低于目前代用的普通滤纸。经数家种子部门试用,反应良好。此将发芽纸研制过程中生物学适用性测试的部分内容报告如下。  相似文献

为探讨施肥对小麦田间CO2释放量的影响,设计以四个不同施肥水平为主处理,以三个不同年代的主栽品种碧玛1号、烟农15号、山农11号为研究对象,采用封闭式静态暗箱技术,应用LI-6400型便携式光合测定仪(美国)在作物生长期测定土壤CO2释放量和光合速率。同时测定了作物的生理生化指标(株高、叶绿素含量、旗叶叶面积)和生物量以及环境影响因素土壤温度和水分等。研究结果表明:(1)施肥有利于增加土壤CO2释放量,且中氮处理水平释放量最多。(2)土壤温度和水分对土壤CO2释放量有一定的影响,土温越高,水分越大,CO2释放越多。  相似文献
