The purpose of this study was conducted to evaluate the use of a system of timed artificial insemination (TAI) and early diagnosis
of pregnancy in Nelore beef cows in order to reduce the breeding season (BS) from 150 to 90 days and analyze the factors affecting
the conception rate of cows selected for TAI. The study included 110 Nelore cows belonging to the Experimental Farm Capim
Branco, Brazil. The body condition scores (BCS) were evaluated, and those above 2.50 were included in the TAI protocol. Five
groups of cows were formed at intervals of 21 days postpartum and then commenced a 9-day progesterone protocol. Progesterone
devices were used three times. Statistical analysis of binary variables was performed by logistic regression in the program
Proc Logistic of Sas. A 5% of significance (P < 0.05) was used to determine the presence of statistical differences and the effect of independent variables on conception
rate. The TAI and early diagnosis of pregnancy reduces the BS from 150 to 90 days without changing the pregnancy rate. The
conception rate was not affected by the presence of the calf (P > 0.05), the corpus luteum (P > 0.05), the BCS (P > 0.05), or the number of reuses of the intravaginal progesterone device (P > 0.05). Therefore, this technique is beneficial for breeding Nelore beef cattle and can be used in anestrous cows with a
BCS above 2.50. Additionally, the progesterone devices can be reused to reduce the cost of the protocol without compromising

Aqueous and ethanolic extracts obtained from nine Chilean marine macro-algae collected at different seasons were examined in vitro and in vivo for properties that reduce the growth of plant pathogens or decrease the injury severity of plant foliar tissues following pathogen infection. Particular crude aqueous or organic extracts showed effects on the growth of pathogenic bacteria whereas others displayed important effects against pathogenic fungi or viruses, either by inhibiting fungal mycelia growth or by reducing the disease symptoms in leaves caused by pathogen challenge. Organic extracts obtained from the brown-alga Lessonia trabeculata inhibited bacterial growth and reduced both the number and size of the necrotic lesion in tomato leaves following infection with Botrytis cinerea. Aqueous and ethanolic extracts from the red-alga Gracillaria chilensis prevent the growth of Phytophthora cinnamomi, showing a response which depends on doses and collecting-time. Similarly, aqueous and ethanolic extracts from the brown-alga Durvillaea antarctica were able to diminish the damage caused by tobacco mosaic virus (TMV) in tobacco leaves, and the aqueous procedure is, in addition, more effective and seasonally independent. These results suggest that macro-algae contain compounds with different chemical properties which could be considered for controlling specific plant pathogens. 相似文献

Equine herpesvirus 1 (EHV-1) is the causative agent of abortion, perinatal foal mortality, neurological and acute respiratory diseases in horses. Conventional laboratory diagnosis involving viral isolation from aborted foetuses is laborious and lengthy and requires processing of samples within 24 h of collection, which is problematic for samples that come from long distances. The aim of this study was to develop a polymerase chain reaction (PCR) assay useful in Argentina to detect DNA sequences of EHV-1 in different tissues from aborted equine foetuses with variable quality of preservation and without the use of conventional DNA fenolic extraction. Several DNA extraction protocols and primers were evaluated. The amplification method was standardized and its specificity was analysed using 38 foetal samples of variable quality of preservation. Of the 38 different foetal tissues, nine livers, six spleens and two lungs in good preservation and eight livers, one spleen and four lungs in a poor state of preservation were positive for PCR. EHV-1 was recovered only from the nine livers, five spleens and two lungs in good preservation. No virus was isolated from the samples that were poorly preserved. Viral isolation was confirmed by cytopathic effect and indirect immunofluorescence. The specificity of the PCR results was confirmed by the restriction endonuclease digestion of PCR products and hybridization. 相似文献

One of the challenges of modern grassland systems is to minimize nitrogen (N) fertilization without negatively affecting the forage yield. Therefore, critical N dilution curves (Nc = ac W?b) have been developed in different species to improve N fertilization management. The aim of this study was to validate a critical N dilution curve for hybrid ryegrasses. Two field experiments were conducted in southern Chile. Treatments were the factorial combination of two hybrid ryegrasses (Shogun and Trojan cultivars) and seven N fertilization rates (0, 50, 100, 200, 350, 525 and 700 kg N/ha). Factors were arranged in a split‐plot design, where forage species were assigned to main plots and N rates to subplots that were randomized into four blocks. A wide range in forage yield and plant N concentration was observed (yield: 0.16 and 3.9 Mg DM/ha and N: 1.6% and 5.1%). The variations in these traits were principally explained by the N levels and harvest times. Relative yield responses of both cultivars were significantly (p < 0.001, R2 = 0.81–0.87) related to the nitrogen nutrition index (NNI) calculated with different critical N dilution curves. However, the NNI calculated with N dilution curves from annual ryegrass best described the relative yield response of hybrid ryegrass. Therefore, this validated critical N dilution curve (%Nc = 4.1W?0.38) will serve as a useful diagnosis tool for improving the N fertilization management of grazing systems for hybrid ryegrasses. 相似文献

Parentage analysis in aquaculture determines genealogical relationships between broodstock and progeny when the parents are unknown. Thus, parentage analysis is a useful tool to establish pedigree reports in molecular‐assisted selection programs. Here, we evaluated 10 heterologous microsatellite markers for parentage assignment in abalone hybrids produced from 43 abalone broodstocks of red abalone (Haliotis rufescens) and Japanese abalone (H. discus hannai). The allele frequencies, exclusion probabilities and broodstock contributions were calculated using CERVUS, PAPA and GERUD software. The polymorphic information content (PIC) values showed that most of the microsatellite loci were highly informative (>0.7) and more than 90% of parentage assignment was possible with a minimum of 5–6 microsatellite markers. Parentage assignment for hybrid and pure‐red progeny showed a better performance than pure‐Japanese progeny. This result could be due to the high level of allele loss in the parental genotypes. In addition, results indicated that only two sires contributed over 80% and 90% of red and hybrid progenies, respectively. This study gives a new molecular tool to support marker‐assisted selection in abalone hybrids produced in Chile. 相似文献

Two introduced abalone species are currently produced in Chile, red abalone Haliotis rufescens and Japanese abalone Haliotis discus hannai. However, red abalone accounts for 99% of total production, while the Japanese abalone has not adapted well to Chilean coastal waters. This study reports the hatching, growth and thermal tolerance performance in interspecific hybrids produced between red (R) and Japanese (J) abalone. Our results show that egg age and sperm concentration were critical factors to produce hybrids. The cross R♀ × J♂ showed a fertilization rate of 55.3 ± 3.5% using 20‐min‐old eggs and sperm concentrations of 14 × 106 cells mL?1, while the reciprocal cross (J♀ × R♂) was not successful. Further, larval development stages were similar in RR, JJ and RJ hybrid abalones. Among the experimental trials, settlement rate varied from 12.3% to 18.6% and final survival from 20.1% to 31.7%, being the RJ hybrid rates intermediate between parental species. The final shell lengths were similar between RR and RJ hybrids, but significantly higher in JJ abalones. In addition, thermal tolerance was ascertained due its pivotal role for the abalone physiology. Thus, RJ hybrids showed the highest HSP70 gene expression and offers new possibilities to expand Chilean abalone production in warm waters zones. 相似文献

The aim of this study was to evaluate a range of statistical and geostatistical methods for their usefulness in providing insights into how highly pathogenic avian influenza (HPAI) subtype H5N1 might spread through a national population of village poultry. The insights gained allow the generation of disease dispersion hypotheses. The case study data set consisted of 161 outbreaks of HPAI subtype H5N1 in village poultry reported in Romania between October 2005 and June 2006. Reports of village outbreaks (%) occurred in three waves: October-December (14%), February-March (16%), and May-June (68%). Risk mapping - based on variography and kriging - was used to visualize the evolution of the epidemic. Outbreaks first appeared in eastern and southern Romania, particularly within an area that forms part of the Danube River Delta. The largest phase of the epidemic affected villages in all parts of central, southern, and eastern Romania, but outbreaks were clustered in central Romania. Outbreaks spread in an east to west direction. By using geostatistical visualisation and spatial statistics, the evolution of the epidemic could be characterised into two parts: disease introduction, local spread, and sporadic outbreaks, and long-distance disease spread with rapid epidemic propagation. This is consistent with the hypothesis that the environment and landscape (specifically the Danube River Delta) played a critical role in the introduction and initial spread of HPAI subtype H5N1 during the autumn and winter of 2005, and that the movement of poultry might have introduced the infection into central Romania during the spring and summer of 2006. Further research focusing on the spatio-temporal interface between the two parts of the epidemic might reveal how and why it progressed from a confined, local epidemic to a large, national epidemic.
