The shoot/plantlet regenerationin vitro of seven potato (Solanum tuberosum L.) cultivars from petioles with intact leaflets was assessed using six treatment combinations-a basal medium with or without silver thiosul-phate (STS) or thidiazuron (TDZ) at two concentrations (2 or 0.5 mg/l) of the indoleacetic acid (IAA). The basal medium consisted of Murashige and Skoog (MS) salts and vitamins supplemented with 3 mg/1 6-benzylaminopurine, and 1 mg/1 gibberellic acid, 30 g/l sucrose, and 7.0 g/l PHYTOAGAR. Two full sets repeats and one partial set repeat of independent experiments were conducted and all produced similar results. Silver thiosulphate decreased the regeneration frequency and number of shoots per callus. No significant changes were observed with thidiazuron. Regeneration rates of (100% ) with up to 20 shoots/plantlets per callus were achieved at 2 mg/1 IAA with cultivars Désirée, Kennebec, Niska, and Lenape. These cultivars still showed high regeneration rates (87%–98% ) on media with 0.5 mg/1 IAA, and good regeneration rates were also achieved by the other three cultivars (48%, 94%, and 50% for Chieftain, Russet Burbank, and Shepody, respectively). Even with the single medium protocol (0.5% IAA without thiosulphate or thidiazuron), Désirée, Lenape, and Niska exhibited a regeneration rate of 98%. The use of petiole-with-leaflet explants could be ideal for the regeneration step of potato genetic transformation protocol because of their high regeneration efficiency and their small cut surface area forAgrobacterium elimination after co-incubation.  相似文献

Beef cows in the subtropical USA must be adapted to the stressors of the environment, typically supplied by using Brahman (Br) breeding. Calves produced in the region, however, are usually grown and finished in more temperate regions, and have a perceived reputation for poor ADG and feed efficiency during finishing. Compromised fertility and carcass quality often associated with the Br have increased interest in tropically adapted Bos taurus breed types. The objective of this study was to evaluate 3 breeds [An = Angus (Bos taurus, temperate); Br (B. indicus, tropical); and Ro = Romosinuano (B. taurus, tropical)] and all possible crosses during various segments of post-weaning growth, and for feed efficiency during the finishing phase. Steer calves (n = 473) born over 3 yr were weaned in late September, backgrounded for at least 21 d (BKG), shipped 2,025 km to El Reno, OK, in October, fed a preconditioning diet for 28 d (RCV), grazed wheat (Triticum aestivum L.) pasture from November to May (WHT), finished on a conventional feedlot diet (FIN), and serially harvested after approximately 95, 125, and 150 d on feed. Body weight and ADG during each segment were tested using a mixed model that included calf age at weaning, year (Y), breed of sire (SB), breed of dam (DB), and interactions. In addition, winter treatment (continuous wheat or reduced grazing of wheat with supplement) was included for the wheat and feedlot phases. Sire within SB × SB [and pen (barn × year) for feedlot phase] were considered random. The SB × DB interaction was significant for all traits (P < 0.01) except exit velocity taken at weaning and ADG during FIN, but both traits were affected by 3-way interactions with Y or harvest group. Tropically-adapted purebred steers had greater (P < 0.01) ADG than AnAn through weaning and BKG in FL but the reverse was true during the RCV and WHT segments. Similar, but less pronounced results were noted for F(1) steers with 100% tropical influence compared with those with only 50%. Heterosis was numerically greater for most traits for An × Br (11 to 64%) compared with An × Ro and Br × Ro (3 to 42%), which were similar. In a subset of the steers (n = 261), G:F was not influenced by level of tropical breeding, but tropically adapted steers were more efficient (P < 0.05) by residual feed intake. No heterosis was evident. These data show that in temperate zones, winter is the period when productivity of tropically adapted cattle is compromised.  相似文献

Summary Twenty potato cultivars were tested in replicated trials in three seasons. The weather gradually changed from relatively high humidity and cold air temperatures in February to dry and hot in July. Fresh and dry weights, sucrose, reducing sugar and starch contents were recorded on tubers sequentially harvested over four stages during the growing period of each of the three trials. The data were analyzed for genotype-environment interactions by a path regression model which is composed of a main effect and four multiplicative effects. The model fitted well to the observed data for all traits but reducing sugar. Results of dry tuber weight were used to calculate a heat susceptibility index for each of the cultivars. Four components of the index were identified based on the path model. Comparison of sizes of the components provided information on the degree of response to the environmental stress of each of the cultivars during successive stages of growth. High yielding and heat tolerant cultivars were identified based on the experimental results.  相似文献

Oak mortality is often associated with a complex of decline factors. We describe the morphological and physiological responses of coast live oak, Quercus agrifolia Née, in California to an invasive insect, the goldspotted oak borer (GSOB), Agrilus auroguttatus Schaeffer (Coleoptera: Buprestidae), and evaluate drought as a potential inciting factor. Morphological traits of 356 trees were assessed and physiological traits of 70 of these were monitored intensively over one growing season. Morphological characteristics of tree health included crown thinning and dieback; bole staining resulting from larval feeding; density of GSOB adult exit holes; and holes caused by woodpecker feeding. These characteristics were used to rank GSOB infestation/injury into four classes, and taken together, they explained 87% of the variation in a principal component analysis. Drought stress on various size/age and infestation classes of Q. agrifolia was measured by assessing branchlet pre-dawn and solar noon xylem water potential, leaf cell turgor potential, and water use efficiency over one growing season. Both morphological and physiological traits were highly variable in mature and old growth trees. Early summer plant water status (branchlet xylem water potential and water use efficiency) was similar between uninfested and newly colonized trees, suggesting that GSOB are not pre-selecting drought-stressed Q. agrifolia for oviposition. By late summer, leaf water and cell turgor potentials were lower in infested than in uninfested mature trees, suggesting that GSOB infestation causes drought stress in these trees. Among the tree size/age classes, infested old growth trees exhibited the greatest change in water use efficiency over the growing season, and showed greater morphological injury symptoms of decline than infested mature trees. Morphological attributes of decline in Q. agrifolia associated with GSOB were correlated weakly with increasing physiological drought stress among infestation classes of trees. We propose that the collection of morphological responses of Q. agrifolia to GSOB described here can be used to monitor the future expansion of the GSOB distribution as well as the GSOB-induced decline of Q. agrifolia in California.  相似文献

Organic matter and abiotic conditions seem to influence distribution patterns of Enchytraeidae. In this study effects of changes in management practices on population dynamics of enchytraeids were determined. At two sites (in Athens and Griffin, GA) parts of a fescue (FE) field were converted into conventional tillage (CT) and no-tillage (NT) plots and changes in densities and depth distribution of enchytraeids were followed for 3 years. A site at Watkinsville, containing various soil textures and characterised by very low organic carbon content, which was converted into no-tillage 4 years earlier, was also sampled. Significant reductions in enchytraeid densities, after conversion of fescue into CT, were only found at Griffin. The management practices affected the vertical distribution of enchytraeids. In fescue and NT more enchytraeids were found in the 0–5cm than in the 5–15cm layer. In conventional tillage fields enchytraeids were more evenly distributed over the profile or more abundant in the 5–15cm layer. Management also affected the timing of population dynamics in the different plots. At two sample dates high abundances were found in CT plots only, not in any of the other plots. Enchytraeids were larger at Athens than at Griffin and Watkinsville and contained more soil particles in their gut. At Athens enchytraeids will presumably contribute more to the development of soil structure than at the other sites. We evaluated the commercial and recreational fishery landings over the past 22 years, first at the national level, then for populations of concern (those that are overfished or experiencing overfishing), and finally by region. Recreational landings in 2002 account for 4% of total marine fish landed in the United States. With large industrial fisheries excluded (e.g., menhaden and pollock), the recreational component rises to 10%. Among populations of concern, recreational landings in 2002 account for 23% of the total nationwide, rising to 38% in the South Atlantic and 64% in the Gulf of Mexico. Moreover, it affects many of the most-valued overfished species-including red drum, bocaccio, and red snapper-all of which are taken primarily in the recreational fishery.  相似文献

Application of a comprehensive wave climate program to seven major deltas indicates that deltaic configurations and coastal landform combinations depend to a considerable degree on the wave power adjacent to the shore and on the river discharge relative to wave forces. Nearshore wave power is not correlative with deepwater wave power but, owing to frictional attenuation, is a function of the subaqueous slope. The subaqueous slope, in turn, depends partially on the slope and width of the continental shelf but primarily on the rate at which the river can supply sediments to the nearshore zone. River-dominated configurations result only when the river is able to build flat offshore profiles, which reduce nearshore wave power; where the subaqueous slope is steep, waves reach the shore comparatively undiminished and wave-built forms prevail.  相似文献
