Water soluble plasma proteins were fractionated from hen's egg yolk, and the molecular weight and pI of the most abundant protein species were characterized with gel electrophoresis. The proteins were identified by mass spectrometry. The protein fraction was used to produce oil-in-water emulsions, both at various protein concentrations and at various pH values, and the surface load was determined through serum depletion. The competitive adsorption was studied through the determination of nonadsorbing species with gel electrophoresis. The results show that it was possible to form an oil-in-water emulsion for which droplet size and maximum surface load depended on the protein concentration and pH. Serum albumin and YGP40 adsorbed selectively at the oil/water interface throughout the pH range investigated, and for albumin the selectivity increased close to its pI. It is suggested that this selective adsorption is due to long hydrophobic stretches in the polypeptide chain, which are present in the selectively adsorbing species but absent in less adsorbing species. 相似文献

NG-391 (1) and NG-393 (2), previously reported from undescribed Fusarium species as nerve-cell growth stimulants, were identified from fermentation extracts of the entomopathogenic fungus Metarhizium anisopliae. These compounds are 7-desmethyl analogues of fusarin C and (8Z)-fusarin C, mutagenic toxins from Fusarium species that contaminate corn. A mutant strain of M. anisopliae (KOB1-3) overproduces 1 and 2 by ca. 10-fold relative to the wild-type strain, ARSEF 2575, from which it was derived. Overproduction of these compounds in KOB1-3 imparts a yellow pigmentation to the culture medium of the fungus. These compounds were inactive at 100 mug/disk in antimicrobial disk diffusion assays. Compound 1 was inactive at 100 ppm in a mosquitocidal assay. However, like their fusarin analogues, 1 and 2 exhibited potent S9-dependent mutagenic activity in the Salmonella mutagenicity test. Discovery of these highly mutagenic mycotoxins in M. anisopliae suggests that screening for production of NG-391 and NG-393 in strains that are used as biocontrol agents would be a prudent course of action. The impact of these findings on the use of M. anisopliae as a biocontrol agent is currently unknown and requires further investigation. 相似文献

The impact of 1-methylcyclopropene (1-MCP) on the synthesis and retention of flavonoid compounds during storage and ripening of red Delicious (Malus x domestica Borkh.) apples was investigated. Numerous anthocyanins, flavonols, flavan-3-ols, and a hydroxycinnamic acid from three different fruit harvest maturities were monitored after a 120 day storage and 1 week shelf life period using high-performance liquid chromatography/diode array detector analysis. The total flavonoid concentration was 5% greater in fruit treated with 1-MCP, whereas chlorogenic acid levels were 24% lower. All compounds analyzed increased in concentration during fruit harvest; however, the anthocyanins generally declined after storage, while chlorogenic acid levels increased. 1-MCP treatment resulted in the retention of anthocyanins in the latter stages of storage but did not affect the flavonols and flavan-3-ols. Chlorogenic acid biosynthesis from early and optimal fruit harvest maturities was greatly inhibited by 1-MCP during storage and the 1 week shelf life period. However, 1-MCP did not affect chlorogenic acid concentrations in late-harvested fruit. Results suggest that 1-MCP may inhibit the activity of phenylalanine ammonia-lyase and subsequent biosynthesis of flavonoid compounds. However, because very little postharvest biosynthesis of flavonoids occurs in apples, 1-MCP treatment may be useful for maintaining some of the intrinsic flavonoid levels of red Delicious apples, if applied at the proper harvest maturity. 相似文献

The metabolism of organic pollutants by plants normally requires contaminant direct uptake by cells. Factors affecting this uptake and the later distribution of chemicals within the plant include the physicochemical properties of the compounds (concentration, structure, solubility, log k(ow), diffusion rate) and the biochemical characteristics of the plant. This paper reports the tolerance, uptake, and effects of the pesticides carbaryl, linuron, and permethrin on Lupinus angustifolius germination and growth as well as contaminant intraplant distribution and possible degradation. Lupine plants were grown in hydroponic culture containing either 1 or 5 mg of the individual pesticides, or combinations of these (1, 5, or 10 mg of each), in 100 mL nutrient and water solutions. Analysis of the remaining solutions 8 days post-germination showed the water solutions to have higher remaining pesticide concentrations than nutrient solutions. Furthermore, in the presence of pesticides, germination was more frequent in the water solutions. After 16 days of growth, the plants were harvested, and their tissues were microwaved digested and analyzed by reversed-phase liquid chromatography. Although only minor quantities of each pesticide were detected in plant tissues, their amount in the roots was higher than in the stems. No accumulation was noted in the cotyledons, and only 2% of linuron was detected in the leaves. Mass recovery at the end of the experiment showed that 57, 53, and 55% of carbaryl, linuron, and permethrin, respectively, were degraded and/or bound in an irreversible manner to plant material.
